Activation of stress-response genes by retrograde signaling-mediated destabilization of nuclear importin IMPα-9 and its interactor TPR2

Stress-induced retrograde signal transmission from the plastids to the nucleus has long puzzled plant *** address this,we performed a suppressor screen of the ceh1 mutant,which contains elevated 2-C-methyl-d-erythritol-2,4-cyclopyrophosphate(MEcPP)levels,and identified the gain-of-function mutant impα-9,which shows reversed dwarfism and suppressed expression of stress-response genes in the ceh1 background despite heightened *** genetic and biochemical analyses established that the accumulation of MEcPP initiates an upsurge in Arabidopsis SKP1-like 1(ASK1)abundance,a pivotal component in the proteasome degradation *** increase in ASK1 prompts the degradation of IMPα-***,we uncovered a protein-protein interaction between IMPα-9 and TPR2,a transcriptional co-suppressor and found that a reduction in IMPα-9 levels coincides with a decrease in TPR2 ***,the interaction between IMPα-9 and TPR2 was disrupted in impα-9 mutants,highlighting the critical role of a single amino acid alteration in maintaining their *** of their interaction results in the reversal of MEcPP-associated *** immunoprecipitation coupled with sequencing analyses revealed that TPR2 binds globally to stress-response genes and suggested that IMPα-9 associates with the *** function together to suppress the expression of stress-response genes under normal conditions,but this suppression is alleviated in response to stress through the degradation of the suppressing *** biological relevance of our discoveries was validated under high light stress,marked by MEcPP accumulation,elevated ASK1 levels,IMPα-9 degredation,reduced TPR2 abundance,and subsequent activation of a network of stress-response *** summary,our study collectively unveils fresh insights into plant adaptive mechanisms,highlighting intricate interactions among retrograde signaling,the proteasome,and nuclear transport machinery.

Intraspecific floral colour variation in three Pedicularis species

Our study aimed to compare the essential oil(EO)concentration and composition of several Apiaceae species growing in the Northern region of *** yields of EOs from materials ranged from 0.03%(root EO of Angelica acutiloba and aerial parts EO of Heracleum bivittatum)-0.27%(leaf EO of Xyloselinum vietnamense).Gas chromatography-mass spectrometry(GC-MS)allowed the identiﬁcation of 74 components in the EOs of six Apiaceae species,making up 94.4%–100.0%of the *** EO from Angelica acutiloba,(Z)-ligustilide accounted for an extremely large proportion(94.9%).EO of Angelica pubescens was dominated by six character-istic components includingα-pinene(21.5%),β-phellandrene(18.1%),p-cymene(12.2%),3-methylnonane(8.7%),o-cymene(8.1%),and D-sylvestrene(6.2%).The EO from Cryptotaenia japonica was characterized by high amounts ofα-selinene(48.7%),β-selinene(23.7%),and trans-β-farnesene(5.4%).The EOs from leaves and stems of Xyloselinum vietnamense were characterized by high concentrations of sabinene(69.8%and 33.8%),4-terpineol(8.7%and 7.4%)andβ-pinene(4.0%and 6.5%)while EOs from aerial parts and root of Xylo-selinum leonidii comprise four characteristic monoterpenes includingα-pinene(28.2%and 52.8%),β-pinene(7.9%and 10.3%),β-phellandrene(7.6%and 15.3%),and sabinene(3.0%and 4.1%).Additionally,cryptone is also one of the major components in the EO of Xyloselinum leonidii(13.2%in the aerial parts oil and 2.8%in the root oil).In the EOs isolated from the aerial parts and root of Heracleum bivittatum,α-pinene(22.5%and 70.2%)andβ-pinene(43.2%and 20.0%)were the predominant *** appeared in the EO from aerial parts of Heracleum bivittatum with a relatively high concentration(13.5%)while bornyl acetate(5.1%)was also one of the main components in the EO from its aerial parts but was not detected in other Apiaceae species in the present *** databases help identify and control the quality of plant material studied from the family Apiaceae growing in Vietnam.

Differential roles of C-3 and C-6 phosphate monoesters in affecting potato starch properties

The effects of starch phosphate monoester content(SPC),namely C-3(C3P)and C-6 phosphate monoesters(C6P),on the starch properties were investigated using four potato starches with varied SPC/C3P/C6P and two nonphosphorylated maize starches with a similar range of amylose content(AC)as *** starch property results showed that a higher SPC is associated with lower turbidity,storage and loss modulus after storage,and water solubility,but higher swelling power(SP)and pasting *** findings suggested that SPC inhibited molecular rearrangement during storage and starch leaching during heating,and enhanced swelling and viscosities due to increased hydration and water uptake caused by the repulsion effect of phosphate groups and a less ordered crystalline *** SPC also resulted in lower resistant starch(RS)content in a native granular state but higher RS after *** correlations further indicated that SPC/C3P/C6P were positively correlated with peak(r^(2)=0.925,0.873 and 0.930,respectively),trough(r^(2)=0.994,0.968 and 0.988,respectively),and final viscosities(r^(2)=0.981,0.968 and 0.971,respectively).Notably,SPC,mainly C3P,exhibited a significantly positive correlation with SP(r^(2)=0.859)and setback viscosity(r^(2)=0.867),whereas SPC,mainly C6P,showed a weak positive correlation with RS after retrogradation(r^(2)=0.746).However,SPC had no significant correlations with water solubility,turbidity and rheology properties,which were more correlated with *** findings are helpful for the food industry to select potato starches with desired properties based on their contents of SPC,C3P,or C6P.

Diabetes and Hypertension Are Associated with Food Insecurity in a Cameroonian Population: A Case-Control Study

Diabetes and hypertension are the most prevalent cardiovascular risk factors. Recent studies showed an increase in the prevalence of food insecurity in our country. The aim of this study was to assess how food insecurity affects the dietary habits, socio-demographic characteristics and metabolic profile of individuals with diabetes or hypertension. This case-control study was conducted among diabetic and hypertensive participants (cases) and diabetic and hypertensive normal (controls) during the screening campaigns for nutrition-related chronic diseases. The sociodemographic, clinical and biochemical parameters of the participants were analyzed. Logistic regression analyses were performed to identify factors associated with diabetes and hypertension in the study population. Bivariate analyses showed that male gender (OR = 1.972;95% CI: 1.250 - 3.089), regular alcohol consumption (OR = 2.012;95% CI: 1.294 - 3.130), low fruit consumption (OR = 1.590;95% CI: 1.016 - 2.488), low dietary diversity (OR = 2.915;95% CI: 1.658 - 5.127) and abdominal obesity (OR = 1.893, CI 95% 1.203 - 2.978) were significantly associated with hypertension. In addition, low fruit consumption (OR = 1.829;95% CI 1.092 - 3.064), low legume consumption (OR = 3.515;95% CI 1.861 - 6.635), and hypertriglyceridaemia (OR = 2.241, 95% CI 1.139 - 4.408) were significantly associated with diabetes. The indirect association observed between food insecurity and diabetes and hypertension suggests the need for nutritional policies aimed at popularizing the production and consumption of fruits and legumes. Similarly, health services need to be aware and informed of the important role that food insecurity can play in the development of diabetes and hypertension.

Decoding the biochemical dialogue: metabolomic insights into soybean defense strategies against diverse pathogens

Soybean, a crucial global leguminous crop, confronts persistent threats from diverse pathogens, exerting a profound impact on global *** genetic dimensions of soybean-pathogen interactions have garnered attention, the intricate biochemical responses remain poorly elucidated. In this study, we applied targeted and untargeted liquid chromatography coupled to mass spectrometry(LC-MS) metabolite profiling to dissect the complex interplay between soybeans and five distinct pathogens. Our analysis uncovered 627 idMS/MS spectra,leading to the identification of four main modules, encompassing flavonoids, isoflavonoids, triterpenoids, and amino acids and peptides,alongside other compounds such as phenolics. Profound shifts were observed in both primary and secondary metabolism in response to pathogenic infections. Particularly notable were the bidirectional changes in total flavonoids across diverse pathogenic inoculations, while triterpenoids exhibited a general declining trend. Noteworthy among the highly inducible total flavonoids were known representative antipathogen compounds(glyceollin I), backbone forms of isoflavonoids(daidzein, genistein, glycitein, formononetin), and newly purified compounds in this study(prunin). Subsequently, we delved into the biological roles of these five compounds, validating their diverse functions against pathogens: prunin significantly inhibited the vegetative growth and virulence of Phytophthora sojae; genistein exhibited a pronounced inhibitory effect on the vegetative growth and virulence of Phomopsis longicolla; daidzein and formononetin displayed significant repressive effects on the virulence of P. longicolla. This study underscores the potent utility of metabolomic tools, providing in-depth insights into plant-pathogen interactions from a biochemical perspective. The findings not only contribute to plant pathology but also offer strategic pathways for bolstering plant resistance against diseases on a broader scale.
