Mechanism and experimental study on the fruit detachment of Chinese wolfberry through reciprocating vibration

In order to realize the efficient and high-quality mechanical picking for Chinese wolfberry,firstly,the forced reciprocating vibration picking principle of the Chinese wolfberry branch was studied,and the mechanical model of vibration picking was established based on the simplified cantilever model,and the response analysis and solution of all positions for the branch were carried *** the same time,the critical mechanical model of fruit detachment under the condition of fruit hanging on branches was established,and the theoretical values of inertia force for each component of the branch were ***,through actual measurement and finite element modeling,the natural frequency and forced vibration response simulation for each component of the branch of Chinese wolfberry terminal branch model were both studied,and the relationship between single-point periodic excitation force and high-quality fruit shedding parameters was ***,according to the conclusion of the picking model,a test bench with many groups of adjustable parameters was ***,the last branch of fruit-hanging Chinese wolf berry for Ningqi No.1 was taken as the experimental object,a four-level orthogonal experiment was designed with three factors:frequency,amplitude and entrance ***,the net picking rate,damage rate and false picking rate were taken as the evaluating indicators,referring to the comprehensive scores of the three *** was concluded that the primary and secondary relations of factors affecting the picking effect are frequency,amplitude and entrance angle,and the best operation parameters are frequency of 20 Hz,amplitude of 15 mm,and entrance angle of 45°,then,a hand-held vibration picker with setting parameters was trial-produced,and the optimal parameter combination was verified in the Chinese wolfberry planting base of the National Chinese wolfberry Engineering and Technology Research *** results showed that the net picking rate of rip

The electrochemical nitrate reduction reaction(NO_(3)RR)to ammonia under ambient conditions is a promising approach for addressing elevated nitrate levels in water bodies,but the progress of this reaction is impeded by the complex series of chemical reactions involving electron and proton transfer and competing hydrogen evolution ***,it becomes imperative to develop an electro-catalyst that exhibits exceptional efficiency and remarkable selectivity for ammonia synthesis while maintaining long-term *** the magnetic biochar(Fe-C)has been synthesized by a two-step mechanochemical route after a pyrolysis treatment(450,700,and 1000℃),which not only significantly decreases the particle size,but also exposes more oxygen-rich functional groups on the surface,promoting the adsorption of nitrate and water and accelerating electron transfer to convert it into *** showed that the catalyst(Fe-C-700)has an impressive NH_(3)production rate of 3.5 mol·h^(−1)·gcat^(−1),high Faradaic efficiency of 88%,and current density of 0.37 A·cm^(−2)at 0.8 V *** hydrogen electrode(RHE).In-situ Fourier transform infrared spectroscopy(FTIR)is used to investigate the reaction intermediate and to monitor the *** oxygen functionalities on the catalyst surface activate nitrate ions to form various intermediates(NO_(2),NO,NH_(2)OH,and NH_(2))and reduce the rate determining step energy barrier(*NO_(3)→*NO_(2)).This study presents a novel approach for the use of magnetic biochar as an electro-catalyst in NO_(3)RR and opens the road for solving environmental and energy challenges.

Adult neurogenesis,the process of creating new neurons,involves the coordinated division,migration,and differentiation of neural stem *** process is restricted to neurogenic niches located in two distinct areas of the brain:the subgranular zone of the dentate gyrus of the hippocampus and the subventricular zone of the lateral ventricle,where new neurons are generated and then migrate to the olfactory *** has been thought to occur only during the embryonic and early postnatal stages and to decline with age due to a continuous depletion of neural stem ***,recent years have seen tremendous progress in our understanding of adult brain neurogenesis,bridging the knowledge gap between embryonic and adult ***,we discuss the current status of adult brain neurogenesis in light of what we know about neural stem *** this notion,we talk about the importance of intra cellular signaling molecules in mobilizing endogenous neural stem cell prolife *** on the current understanding,we can declare that these molecules play a role in targeting neurogenesis in the mature ***,to achieve this goal,we need to avoid the undesired proliferation of neural stem cells by controlling the necessary checkpoints,which can lead to tumorigenesis and prove to be a curse instead of a blessing or hope.

Soil salinization poses a threat to maize production worldwide,but the genetic mechanism of salt tolerance in maize is not well ***,identifying the genetic components underlying salt tolerance in maize is of great *** the current study,a teosinte-maize BC2F7 population was used to investigate the genetic basis of 21 salt tolerance-related *** total,125 QTLs were detected using a high-density genetic bin map,with one to five QTLs explaining 6.05–32.02%of the phenotypic variation for each *** total phenotypic variation explained(PVE)by all detected QTLs ranged from 6.84 to 63.88%for each *** all 125 QTLs,only three were major QTLs distributed in two genomic regions on chromosome 6,which were involved in three salt tolerance-related *** addition,10 pairs of epistatic QTLs with additive effects were detected for eight traits,explaining 0.9 to 4.44%of the phenotypic ***,18 QTL hotspots affecting 3–7 traits were *** one hotspot(L5),a gene cluster consisting of four genes(ZmNSA1,SAG6,ZmCLCg,and ZmHKT1;2)was found,suggesting the involvement of multiple pleiotropic ***,two important candidate genes,Zm00001d002090 and Zm00001d002391,were found to be associated with salt tolerance-related traits by a combination of linkage and marker-trait association ***00001d002090 encodes a calcium-dependent lipid-binding(CaLB domain)family protein,which may function as a Ca^(2+)sensor for transmitting the salt stress signal downstream,while Zm00001d002391 encodes a ubiquitin-specific protease belonging to the C19-related *** findings provide valuable insights into the genetic basis of salt tolerance-related traits in maize and a theoretical foundation for breeders to develop enhanced salt-tolerant maize varieties.

In order to find out the matching principle of excitation force and energy consumption of reciprocating vibrating Chinese wolfberry picking device,the energy consumption mechanism of reciprocating vibrating Chinese wolfberry picking device is *** to the structural characteristics and operating principle of the picking device,the no-load and load movement and force characteristics of the crank bearing and vibrating component are analyzed,and the theoretical model of energy consumption of the reciprocating vibration picking device is jointly constructed,and the simulation analysis is carried *** results show that the vibrating component and load mass have a significant influence on torque,the load air resistance phase has a significant effect on torque,and the load air resistance and friction coefficient have no significant influence on ***,by building an AC servo motor torque detection system and a torque sensing detection system,verification experiments are carried out,the maximum torque of the preset system is 1.3 N∙m,the rated power is 400 W,the motor frequency is 20 Hz,the amplitude is 15 mm,and the total mass of the vibrating component is 0.143 *** results show that,the no-load operation,the change trend of detected torque is consistent with simulation,the torque model is verified to be *** maximum torque of simulation and detection are 0.52 N∙m and 0.57 N∙m respectively,and the error between test and simulation is 9.6%.For load operation,the maximum torque of five groups of branch loads of 20 g,60 g,100 g,140 g,180 g and 220 g are detected to be 0.73 N∙m,0.74 N∙m,0.75 N∙m,0.82 N∙m and 0.83 N∙m,respectively,and the relationship model between load and torque is obtained by *** research results can provide a theoretical basis which can configure a suitable motor in the reciprocating vibration Chinese wolfberry picking device with a certain load limit.

Acaricidal effect of the antimicrobial metabolite xenocoumacin 1 on spider mite control

The two-spotted spider mite,Tetranychus urticae Koch,is one of the most harmful pests in many agroecosystems *** effectively manage this pest,there is an urgent need to develop novel bio-active acaricides that support integrated pest management strategies targeting *** this study,we explored the acaricidal effects of xenocoumacin 1 (Xcn1) on *** and its predator Neoseiulus californicus using the highly puri?ed ***1 was extracted and purified from the cell-free supernatant of the Xenorhabdus nematophila CB6 mutant constructed by the easy promoter activated compound identi?cation (easyPACId) *** the concentration of Xcn1 exceeded 100μg mL~(–1),the survival rate of spider mite adults declined to below 40%and the fecundity was decreased by 80%at six days *** concentrations of 25 and 50μg mL~(–1),Xcn1 signi?cantly impeded spider mite development by inhibiting the ***,neither concentration had any adverse effects on the survival or reproduction of the predatory mite *** results from laboratory and semi-?eld experiments consistently demonstrated the effectiveness of the antimicrobial metabolite Xcn1 in controlling pest mites at both the molecular and physiological *** study offers a promising possibility that combines the compatible biocontrol agents of Xcn1 and predatory mites for integrated pest mite control.

Straw return increases crop production by improving soil organic carbon sequestration and soil aggregation in a long-term wheat-cotton cropping system

Straw return is a promising strategy for managing soil organic carbon(SOC)and improving yield ***,the optimal straw return strategy for sustainable crop production in the wheat(Triticum aestivum L.)-cotton(Gossypium hirsutum L.)cropping system remains *** objective of this study was to quantify the long-term(10 years)impact of carbon(C)input on SOC sequestration,soil aggregation and crop yields in a wheat-cotton cropping system in the Yangtze River Valley,*** treatments were arranged with a single-factor randomized design as follows:no straw return(Control),return of wheat straw only(Wt),return of cotton straw only(Ct),return of 50%wheat and 50%cotton straw(Wh-Ch)and return of 100%wheat and 100%cotton straw(Wt-Ct).In comparison to the Control,the SOC content increased by 8.4 to 20.2%under straw return.A significant linear positive correlation between SOC sequestration and C input(1.42-7.19 Mg ha^(−1)yr^(−1))(P2 and 1-2 mm at the 0-20 cm soil depth were also significantly elevated under straw return,with the greatest increase of the aggregate stability in the Wt-Ct treatment(28.1%).The average wheat yields increased by 12.4-36.0%and cotton yields increased by 29.4-73.7%,and significantly linear positive correlations were also detected between C input and the yields of wheat and *** average sustainable yield index(SYI)reached a maximum value of 0.69 when the C input was 7.08 Mg ha^(−1)yr^(−1),which was close to the maximum value(SYI of 0.69,C input of 7.19 Mg ha^(−1)yr^(-1))in the Wt-Ct ***,the return of both wheat and cotton straw was the best strategy for improving SOC sequestration,soil aggregation,yields and their sustainability in the wheat-cotton rotation system.

Decreased soil pH weakens the positive rhizosphere effect on denitrification capacity

The stimulating effect of rhizosphere on denitrification is considered to be an unavoidable loss of soil nitrogen（N） and detrimental to crop N use efficiency,which is regulated by crop growth and soil properties. Soil acidification, occurring rapidly in many intensive farming lands, affects both crop growth and soil properties, thereby altering rhizosphere effect on denitrification. However, the mechanism by which soil acidification regulates rhizosphere denitrification still remains unclear. Here, we determined the denitrification capacity（DC） and associated community compositions of nirK-and nirS-type denitrifiers in maize rhizosphere and bulk soils at four acidity gradients（pH = 6.8, 6.1, 5.2, and 4.2）. Results showed that the stimulating effect of rhizosphere on DC strongly depended on soil pH. Compared to bulk soil, rhizosphere soil had significantly higher DC at pH 5.2, but not at pH of 4.2. With increasing soil acidity, the stimulation of rhizosphere on DC（calculated as the difference in DC between rhizosphere and bulk soils） decreased from 8.01 to 0.01 mg N ***, soil acidification significantly reduced the differences in dissolved organic carbon（DOC） and abundance of key nirK-type denitrifier taxa between rhizosphere and bulk soils, both of which were positively related to the stimulation of rhizosphere on DC. These findings demonstrated that soil acidification could weaken the positive rhizosphere effect on denitrification via regulated C availability and associated nirK-type denitrifier community, potentially reducing N loss risk in rhizosphere soil. The independent role of soil p H should be fully considered when modelling N behaviour in plant-soil systems.

Transcriptome and metabolome analysis of preharvest internal browning in Nane plum fruit caused by high temperature

The preharvest internal browning of Nane plum fruit,with no visible effects on the appearance of the fruit,has become a serious problem in recent years in its production area in Guangdong Province,*** study investigated the effects of environmental factors,including temperature,on Nane plum internal *** orchards at different elevations with different incidences of internal browning were *** fruits with different internal browning incidence levels,the internal browning mechanism was analyzed with transcriptome and metabolome *** results revealed decreased internal browning at high *** treatment significantly reduced internal browning,whereas bagging and insect-proof net-covering treatments significantly increased internal *** bagging and net coverings increase the local ambient temperature,the findings suggest that high temperature is an important factor influencing the internal browning of Nane *** metabolome experiments showed that with increased internal browning,the levels of phenolic hydroxyls such as catechol increased,with simultaneous increases in hydrogen peroxide content and oxidase *** can be speculated that the oxidation of phenolic hydroxyl substances is the main cause of the preharvest browning of Nane *** analysis revealed the increased expression of calcium signaling-related and downstream effector genes and indicated an important role of calcium in internal browning,possibly due to its increased content in the ***,with increasingly serious internal browning,genes related to photosynthesis were down-regulated,while genes related to senescence were up-regulated,thus suggesting the up-regulation of the process of cell senescence during internal *** conclusion,heat stress should be eliminated to reduce preharvest internal browning in Nane plum.

Whole-genome genotyping(WGG)stands as a pivotal element in genomic-assisted plant ***theless,sequencing-based approaches for WGG continue to be costly,primarily owing to the high expenses associated with library preparation and the laborious *** prior development of foreground and background integrated genotyping by sequencing(FBI-seq),we discovered that any sequence-specific primer(SP)inherently possesses the capability to amplify a massive array of stable and reproducible non-specific PCR products across the ***,we further improved FBI-seq by replacing the adapter ligated by Tn5 transposase with an arbitrary degenerate(AD)*** protocol for the enhanced FBI-seq unexpectedly mirrors a simplified thermal asymmetric interlaced(TAIL)-PCR,a technique that is widely used for isolation of flanking ***,the improved TAIL-PCR maximizes the primer-template mismatched annealing capabilities of both SP and AD *** addition,leveraging of next-generation sequencing enhances the ability of this technique to assay tens of thousands of genome-wide loci for any *** cost-effective,user-friendly,and powerful WGG tool,which we have named TAIL-PCR by sequencing(TAIL-peq),holds great potential for widespread application in breeding programs,thereby facilitating genome-assisted crop improvement.
