AIM： To asses the expression of myeloid dendritic cells （CD11c＋） subset during acute HCV hepatitis and its possible involvement in natural history of the infection.METHODS： We enrolled 11 patients with acute hepatitis C （AHC）（Group A）, 10 patients with acute hepatitis A （AHA）（as infective control-Group B） and 10 healthy donors （group C） in this study. All patients underwent selective flow cytometry gating strategies to assess the peripheral number of the myeloid dendritic cells （mDCs） to understand the possible role and differences during acute hepatitis.RESULTS： Eight of 11 patients with acute HCV hepatitis did not show any increase of mDCs compared to healthy individuals, while a significant decrease of mDCs was found in absolute cell count （z=-2.37, P〈0.05） and percentage （z=-2.30; P〈 0.05） as compared with AHA. On the contrary, The remaining three patients of the group A had a higher mDCs number and percentage as occur in group B. Interestingly, after six months, those patients did not show any increase of mDCs subset were chronically infected, while the three subjects with an increase of peripheral mDCs, as in HAV acute infection, resolved the illness.CONCLUSION： The lack of increase of mDCs during acute hepatitis C might be an important factor involved in chronicization of the infection.

Congenital heart disease (CHD) is a major birth defect worldwide. However, the aetiology of CHD remains unclear and the detailed molecular mechanisms underlying CHD pathogenesis have not been fully understood. The human regulator of calcineurin (RCAN1) gene is highly expressed in human hearts and brains (Fuentes et al., 1995) and composed of seven exons that are alternatively spliced and/or transcribed by differential promoters to produce different isoforms (Fig. 1A) (Yang et al., 2000). RCAN1.1 (NM_004414) and RCAN1.4 (NM_203418) are the two major isoforms differentially expressed in many tissues and cells. RCAN1.1 is constitutively expressed while the expression of RCAN1.4 is induced by diverse stimuli (Yang et al., 2000; Lange et al., 2004). RCAN1.4 negatively regulates NFAT-mediated transcription (Lange and Yutzey, 2006; Qin et al., 2006), which is an important regulator for early heart development (de la Pompa et al., 1998). Although SNPs rs149048873, rs193289374 and rs143081213 in the RCAN1.1 promoter were not associated with sporadic CHD (Guo et al., 2015; Li et al., 2015), the function of RCAN1.4 in CHD remains elusive.

[Objectives]The demand for ponies has been increasing in recent years,but the stock of Debao ponies is far from enough to supply market *** this context,this paper aims to achieve the purpose of conservation,purification and rejuvenation.[Methods]Under the conditions of same age,semen collection frequency,season,feeding environment,feed nutrition level,etc.,the semen of Debao pony and American pearl pony were collected,respectively,and the quality of the semen collected was analyzed.[Results]There was no significant difference in semen volume between the two breeds of ponies;the sperm vitality of the firstly-collected semen of ponies of which the semen has not been collected for a long time was *** semen quality of American pearl pony was slightly better than that of Debao pony.[Conclusions]When the semen quality of American pearl pony is stable,it can be used for cross-breeding with the Debao pony mare,highlighting the hybrid vigor of the two breeds,thereby achieving the purpose of reducing the size of ponies.

Jasmonates（JAs） are rapidly induced after wounding and act as key regulators for wound induced signaling pathway. However, what perceives the wound signal and how that triggers JA biosynthesis remains poorly understood. To identify components involved in Arabidopsis wound and JA signaling pathway, we screened for mutants with abnormal expression of a luciferase reporter, which is under the control of a wound-responsive promoter of an ethylene response factor（ERF） transcription factor gene, RAP2.6（Related to APetala 2.6）.The rea1（RAP2.6 expresser in shoot apex） mutant constitutively expressed the RAP2.6-LUC reporter gene in young *** with the typical JA phenotypes including shorter petioles,loss of apical dominance, accumulation of anthocyanin pigments and constitutive expression of JA response gene, rea1 plants also displayed cell death and accumulated high levels of JA in response to wounding. The phenotype of rea1 mutant is caused by a gain-of-function mutation in the C-terminus of a mechanosensitive ion channel MscS-like 10（MSL10）. MSL10 is localized in the plasma membrane and is expressed predominantly in root tip, shoot apex and vascular tissues. These results suggest that MSL10 is involved in the wound-triggered early signal transduction pathway and possibly in regulating the positive feedback synthesis of JA.

Dear Editor,Plant tissue culture involves callus formation and de novo shoot ***,explants from differentiated tissues are used to generate a pluripotent cell mass,called callus,on auxin-rich callus-inducing medium(CIM),followed by shoot regeneration on cytokinin-rich shoot-inducing medium(SiM).Callus results from division of pericycle-like cells(Atta et al.,2009;Sugimoto et al.,2010);its cellular identity resembles that of lateral root primordia(Atta et al.,2009;Sugimoto et al.,2010).Callus acquires cellular pluripotency by forming root stem cell niches on CIM(Sugimoto et al.,2010)。

β-defensin peptides are a large family of antimicrobial peptides. Although they kill microbes in vitroand interact with immune cells, the precise role of these genes in vivo remains uncertain. Despite their inducible presence at mucosal surfaces, their main site of expression is the epididymis. Recent evidence suggests that a major function of these peptides is in sperm maturation. In addition to previous work suggesting this, work at the MRC Human genetics Unit, Edinburgh, has shown that homozygous deletion of a cluster of nine β-defensin genes in the mouse results in profound male sterility. The spermatozoa derived from the mutants had reduced motility and increased fragility. Epididymal spermatozoa isolated from the cauda region of the homozygous mutants demonstrated precocious capacitation and increased spontaneous acrosome reactions compared with those from wild-types. Despite this, these mutant spermatozoa had reduced ability to bind to the zona pellucida of oocytes. Ultrastructural examination revealed a disintegration of the microtubule structure of mutant-derived spermatozoa isolated from the epididymal cauda region, but not from the caput. Consistent with premature acrosome reaction and hyperactivation, spermatozoa from mutant animals had significantly increased intracellular calcium content. This work demonstrates that in vivo β-defensins are essential for successful sperm maturation, and that their disruption alters intracellular calcium levels, which most likely leads to premature activation and spontaneous acrosome reactions that result in hyperactivation and loss of microtubule structure of the axoneme. Determining which of the nine genes are responsible for the phenotype and the relevance to human sperm function is important for future work on male infertility.

Evaluation of SRAP markers efficiency in genetic diversity of Aspergillus flavus from peanut-cropped soils in China

In order to evaluation the efficiency of SRAP markers on genetic diversity of Aspergillus flavus,we screened out 17sets of primer pairs which could produce clear and reproducible SRAP bands from 150 SRAP primer *** size of SRAP fragments ranged from 120 to 2100 *** pair Me10/Em9 produced the maximum number of polymorphic bands(12 bands),while Me8/Em13 produced the fewest number of polymorphic bands(only 1).Through analysis genetic diversity ability of different sets of primer pairs,the set of 12 primer pairs was selected for SRAP genetic marker of *** analysis was performed based on the genetic similarity coefficients,which ranged from 0.53 to 0.89.A dendrogram assembled using the unweighted pair-group method with arithmetic averages grouped *** samples into 5 main *** results suggested that SRAP marker is a useful molecular technology for the diversity of *** from peanut soils in China.
