A Novel Poly(3-hexylthiophene) Engineered Interface for Electrochemical Monitoring of Ascorbic Acid During the Occurrence of Glutamate-Induced Brain Cytotoxic Edemas

Although neuroelectrochemical sensing technology offers unique benefits for neuroscience research,its application is limited by substantial interference in complex brain environments while ensuring biosafety requirements.In this study,we introduced poly(3-hexylthiophene)(P3HT)and nitrogen-doped multiwalled carbon nanotubes(N-MWCNTs)to construct a composite membrane-modified carbon fiber microelectrode(CFME/P3HT-N-MWCNTs)for ascorbic acid(AA)detection.The microelectrode presented good linearity,selectivity,stability,antifouling,and biocompatibility and exhibited great performance for application in neuroelectrochemical sensing.Subsequently,we applied CFME/P3HT-N-MWCNTs to monitor AA release from in vitro nerve cells,ex vivo brain slices,and in vivo living rat brains and determined that glutamate can induce cell edema and AA release.We also found that glutamate activated the N-methyl-d-aspartic acid receptor,which enhanced Na^(+) and Cl^(−) inflow to induce osmotic stress,resulting in cytotoxic edema and ultimately AA release.This study is the first to observe the process of glutamate-induced brain cytotoxic edema with AA release and to reveal the mechanism.Our work can benefit the application of P3HT in in vivo implant microelectrode construction to monitor neurochemicals,understand the molecular basis of nervous system diseases,and discover certain biomarkers of brain diseases.

Ribosomopathies encompass a spectrum of disorders arising from impaired ribosome biogenesis and reduced functionality.Mutation or dysexpression of the genes that disturb any finely regulated steps of ribosome biogenesis can result in different types of ribosomopathies in clinic,collectively known as ribosomopathy genes.Emerging data suggest that ribosomopathy patients exhibit a significantly heightened susceptibility to cancer.Abnormal ribosome biogenesis and dysregulation of some ribosomopathy genes have also been found to be intimately associated with cancer development.The correlation between ribosome biogenesis or ribosomopathy and the development of malignancies has been well established.This work aims to review the recent advances in the research of ribosomopathy genes among human cancers and meanwhile,to excavate the potential role of these genes,which have not or rarely been reported in cancer,in the disease development across cancers.We plan to establish a theoretical framework between the ribosomopathy gene and cancer development,to further facilitate the potential of these genes as diagnostic biomarker as well as pharmaceutical targets for cancer treatment.

In light of the evolving nature of various diseases,time becomes a crucial factor in diagnosis and identifying the underlying causes.A point-of-care device provides a rapid diagnosis of a disease without using complex and advanced instruments,which are costly and difficult to transport.A paper-based device is a relatively frugal solution wherein the paper is used as a substratum in which the reactions are carried out.These methods are non-invasive,and the sample collection is relatively easy.Saliva is one such body fluid in which various biomarkers are present for numerous diseases.Bioanalysis of saliva has attracted more attention recently due to its non-invasiveness and robustness.Exploiting the discovery of clinical biomarkers from salivary analysis has the potential to revolutionize the healthcare sector by providing crucial information to monitor the health status of individuals and disease progression which enables personalized treatment.This review provides the limitation of the traditional methods in clinical applications and highlights the significance of saliva as a non-invasive biological fluid that is a source of multiple biomarkers associated with various diseases.It also provides insights into the different paper-based colorimetric microfluidic devices developed against salivary biomarkers in the past decade.The major challenges in the point-of-care application and the future prospects have been discussed as well.Further,we also emphasize the importance of this approach in dental disease diagnosis which is least explored and holds potential applications.

The recent article published in Signal Transduction and Targeted Therapy sheds light on the significance of N6-methyladenosine(m6A)methyltransferase-like protein METTL14 in mitigating the progression of metabolic dysfunction-associated fatty liver disease(MAFLD).1 In this study,Wang et al.elucidated the downregulation of METTL14 in hepatocytes from both MAFLD patients and high-fat diet(HFD)-induced MAFLD mouse models,underscoring its pivotal role in maintaining hepatic lipid and redox homeostasis in normal livers.

Biochemistry and transcriptome analysis reveal condensed tannins alleviate liver injury induced by regulating cholesterol metabolism pathway

Free cholesterol has been considered to be a critical risk factor of nonalcoholic fatty liver disease(NAFLD).It remains unknown whether dietary intake of condensed tannins(CTs)have distinguishable effects to alleviate liver damage caused by a high cholesterol diet.Male C57BL/6 mice were fed a high cholesterol diet for 6 weeks,and given CTs treatment at a dosage of 200 mg/(kg·day)at the same time.The results indicated that compared with mice fed a normal diet,a high cholesterol diet group resulted in significant weight loss,dysregulation of lipid metabolism in blood and liver,and oxidative stress in the liver,but CTs treatment dramatically reversed these negative effects.Hematoxylin and eosin(H&E)staining and frozen section observation manifested that CTs treatment could effectively reduce the deposition of liver cholesterol and tissue necrosis caused by high cholesterol intake.CTs alleviated liver injury mainly by regulating the expression of related genes in cholesterol metabolism pathway and AMPK phosphorylation.Our results confirmed that CTs have remarkable cholesterol lowering and anti-liver injury effects in vivo.

The visualization of drugs in living systems has become key techniques in modern therapeutics.Recent advancements in optical imaging technologies and molecular design strategies have revolutionized drug visualization.At the subcellular level,super-resolution microscopy has allowed exploration of the molecular landscape within individual cells and the cellular response to drugs.Moving beyond subcellular imaging,researchers have integrated multiple modes,like optical near-infrared II imaging,to study the complex spatiotemporal interactions between drugs and their surroundings.By combining these visualization approaches,researchers gain supplementary information on physiological parameters,metabolic activity,and tissue composition,leading to a comprehensive understanding of drug behavior.This review focuses on cutting-edge technologies in drug visualization,particularly fluorescence imaging,and the main types of fluorescent molecules used.Additionally,we discuss current challenges and prospects in targeted drug research,emphasizing the importance of multidisciplinary cooperation in advancing drug visualization.With the integration of advanced imaging technology and molecular design,drug visualization has the potential to redefine our understanding of pharmacology,enabling the analysis of drug micro-dynamics in subcellular environments from new perspectives and deepening pharmacological research to the levels of the cell and organelles.

Inflammatory cytokine storms can trigger disease exacerbation and even death and have reached a consensus in the clinical treatment of acute organ failure.However,the existing strategies remain a great challenge to efficiently suppress inflammatory cytokine storms for promoting organ repair and regeneration.Herein,fully human umbilical cord(UC)-derived adhesive materials(UCAM)that integrate decellularized extracellular matrix(ECM)nanofiber hydrogel and homologous mesenchymal stem cells(MSCs)are demonstrated to greatly suppress inflammatory cytokine storms,demonstrating high efficacy in treating acute liver failure(ALF)in rats with 90%hepatectomy.The UC-derived adhesive materials have the capacity to secrete a significant quantity of cytokines by MSCs to recruit activated immune cells to migrate into their ECM nanofiber networks,segregating them away from the infection area and thereby greatly suppressing the inflammatory cytokine storms.As expected,the UC-derived adhesive materials can significantly promote hepatocyte proliferation to achieve functional recovery and regeneration of the liver,significantly improving the survival rate in rats.Our fully human UC-derived adhesive materials provide a new avenue in suppressing inflammatory cytokine storms for promoting organ regeneration that would be really utility in clinical organ transplantation-related treatment.

Three-dimensional(3D)printing has attracted increasing research interest as an emerging manufacturing technology for devel-oping sophisticated and exquisite architecture through hierarchical printing.It has also been employed in various advanced industrial areas.The development of intelligent biomedical engineering has raised the requirements for 3D printing,such as flexible manufacturing processes and technologies,biocompatible constituents,and alternative bioproducts.However,state-of-the-art 3D printing mainly involves inorganics or polymers and generally focuses on traditional industrial fields,thus severely limiting applications demanding biocompatibility and biodegradability.In this regard,peptide architectonics,which are self-assembled by programmed amino acid sequences that can be flexibly functionalized,have shown promising potential as bioinspired inks for 3D printing.Therefore,the combination of 3D printing and peptide self-assembly poten-tially opens up an alternative avenue of 3D bioprinting for diverse advanced applications.Israel,a small but innovative nation,has significantly contributed to 3D bioprinting in terms of scientific studies,marketization,and peptide architectonics,including modulations and applications,and ranks as a leading area in the 3D bioprinting field.This review summarizes the recent progress in 3D bioprinting in Israel,focusing on scientific studies on printable components,soft devices,and tissue engineering.This paper further delves into the manufacture of industrial products,such as artificial meats and bioinspired supramolecular architectures,and the mechanisms,physicochemical properties,and applications of peptide self-assembly.Undoubtedly,Israel contributes significantly to the field of 3D bioprinting and should thus be appropriately recognized.

Evolution of stellated gold nanoparticles:New conceptual insights into controlling the surface processes

Understanding the surface processes(deposition and surface diffusion)that occur at or close to the surface of growing nanoparticles is important for fabricating reproducibly stellated or branched gold nanoparticles with precise control over arm length and spatial orientation of arms around the core.By employing a simple seed-mediated strategy,we investigate the key synthetic variables for precise tuning of in situ surface processes(competition between the deposition and surface diffusion).These variables include the reduction rate of a reaction,the packing density of molecules/ions on the high surface energy facets,and temperature.As a result,the thermodynamically stabilized nanoparticles(cuboctahedron and truncated cube)and kinetic products(cube,concave cube,octapod,stellated octahedron,and rhombic dodecahedron)in different sizes with high quantitative shape yield(>80%)can be obtained depending on the reduction rate of reaction and the packing density of molecules/ions.With computer simulation,we studied the stability of stellated(branched structure)and non-stellated(non-branched structure)gold nanoparticles at high temperature.We construct a morphology phase diagram by varying different synthetic parameters,illustrating the formation of both stellated and non-stellated gold nanoparticles in a range of reaction conditions.The stellated gold nanoparticles display shape-dependent optical properties and can be self-assembled into highly ordered superstructures to achieve an enhanced plasmonic response.Our strategy can be applied to other metal systems,allowing for the rational design of advanced new stellated metal nanoparticles with fascinating symmetry dependent plasmonic,catalytic,and electronic properties for technological applications.

Silk-based biomaterials have gained significant importance making them a promising choice for the future of med-ical technology due to their versatility and biocompatibility.They can be fabricated and tailored through various processing methods such as electrospinning,freeze-drying,and 3D printing,to achieve specific properties and structures namely sponges,hydrogels,films,and scaffolds that can be utilized for different biomedical applica-tions.Biocompatibility,a unique property of silk-based biomaterials,has been demonstrated through both in vivo and in vitro studies and to date many studies have reported the successful use of these silk-based biomaterials in different fields of medicine.In this review,we have elaborately discussed different types of silk,their structural composition,and biophysical properties.Also,the current review focuses on highlighting various biomedical ap-plications of engineered and fabricated silk-based biomaterials which aid in the treatment of certain infections and diseases related to skin,eyes,teeth,bone,heart,nerves,and liver.Furthermore,we have consolidated the advancements of silk-based biomaterials in the different fields of biotechnology such as sensors,food coating and packaging,textiles,drug delivery,and cosmetics.However,the research in this field continues to expand and more significant observations must be generated with feasible results for their reliable use in different biomedical applications.
