Multiple pollutants from crop and livestock production in the Yangtze River: status and challenges

The rapid increase in the proportion of cash crops and livestock production in the Yangtze River Basin has led to commensurate increases in fertilizer and pesticide inputs. Excessive application of chemical fertilizer, organophosphorus pesticides and inappropriate disposal of agricultural waste induced water pollution and potentially threaten Agriculture Green Development(AGD). To ensure food security and the food supply capacity of the Yangtze River Basin, it is important to balance green and development, while ensuring the quality of water bodies. Multiple pollutants affect the transfer, adsorption, photolysis and degradation of each other throughout the soil-plant-water system. This paper considers the impact of multi-pollutants on the nitrogen and phosphorus cycles especially for crops, which are related to achieving food security and AGD. It presents prospective on theory, modeling and multi-pollutant control in the Yangtze River Basin for AGD that are of potential value for other developing regions.

To isolate and verify novel genes from qinghao (Artemisia annua) based on the development-specific and environment-induced transcriptomics, leaves have been harvested from the flowering A. annua plants and exposed to low temperature for isolation of total RNAs and cloning of full-length cDNAs and cDNA fragments, or expressed sequence tags (ESTs). After being sequenced and browsed for homol- ogy, these sequences have been submitted to GenBank. Among the accessed 75 sequences, 4 full-length cDNAs are highly homologous to the known A. annua genes, but 71 ESTs are absent in the sequence records of A. annua genes, in which 34 sequences are homologous to other plant genes, including 24 identified protein-coding sequences and 10 unidentified protein-coding sequences, while other 37 sequences are not present in the sequence records of any plant genes, representing the first cloned plant genes. In order to investigate the responsive patterns of A. annua genes to extreme envi- ronmental stresses, especially low temperature, the expression levels of 3 critical qinhaosu (artemisi- nin) biosynthetic genes, ADS, CYP71AV1 and CPR, have been measured in pre- and post-chilling A. annua seedlings cultured in vitro by semi-quantitative PCR (SQ-PCR). Consequently, ADS and CYP71AV1 genes are strongly induced by chilling, but CPR gene is not significantly affected by such treatment. Furthermore, induction of these genes by chilling can be potently suppressed by Ca2+ channel inhibitor LaCl3 or Ca2+ chelator EGTA, suggesting a putative involvement of Ca2+-CaM signal transduction pathway in chilling-induced overexpression of ADS and CYP71AV1 genes. The real-time fluorescent quantitative PCR (RFQ-PCR) assay of A. annua seedlings exposed to chilling has shown that the expression level of CaM gene is up-regulated for more than 2.5 folds, thereby confirming our above inference on the relevance of Ca2+-CaM-mediated signal transduction to chilling-induced gene overexpression. Inhibitory effect of gold nanoparticles conjugated with interferon gamma and methionine on breast cancer cell line

Objective: To develop a gold nanoparticles complex conjugated with interferon-gamma(IFN-g) and methionine along with application of hyperthermia using near-infrared laser beams for the treatment of cancer ***: Gold nanorods(10 nm) were conjugated with IFN-g and methionine using carbodiimide family and characterized after purification by dialysis bags. Breast cancer cells were cultured and incubated with gold nanorods at different concentrations followed by irradiation with near-infrared laser beam. Samples were then evaluated for their viability in order to determine the effect of treatment and variables by MTT ***: Zetasizer results confirmed the conjugation of gold nanorods with methionine and IFN-g. The median percentage of cell viability in 0.30 mg/m L concentration of gold nanorods was 82%. The cell viability reached to 85% at the same concentration of gold nanorods, which existed in the assayed complex. The results of MTT assay showed that the 0.60 mg/m L concentration of gold nanoparticles complex was toxic on tumor cells(P < 0.05). After exposure to hyperthermia, the viability of cells at 6 min decreased to77% in 0.30 mg/m L concentration of gold nanorods ***: The size and concentration of gold nanorods was not cytotoxic. However,their presence during irradiation near-infrared laser increased the number of dead cells during the treatment of cells.

Chemical profiling of Curcuma aeruginosa Roxb. rhizome using different techniques of solvent extraction

Objective:To investigate the possible phytochemical constituents of Curcuma aeruginosa Roxb.(C. aeruginosa) rhizome using two different techniques of direct solvent extraction. Methods: Two different techniques of direct solvent extractions, i.e. methyl tert-butyl ether(MTBE) extraction and two-phase methanol/chloroform(M/C) system, were used in this study. The analysis of the phytochemical constituents in MTBE and M/C extracts was performed using gas chromatography-mass spectrometry/mass spectrometry. The mass spectra of the compounds was matched with the NIST 08 mass spectral library. Results: The present study revealed that the extraction using two-phase M/C have resulted in higher metabolite coverage compared to the extraction with MTBE. Direct solvent extraction using MTBE revealed the presence of 27 compounds; whereas, M/C allowed the extraction of 18 and 36 compounds in polar(methanol) and nonpolar(chloroform) fractions respectively. The major compounds detected in the MTBE extract that based on the peak area percentage were methenolone(16.64%), cycloisolongifolene, 8,9-dehydro-9-formyl-(15.93%), labd-13-en-15-oic acid,8,12-epoxy-12-hydroxy-γ-lactone(10.77%), propiolic acid, 3-(1-hydroxy)-2 isopropyl-1,5-methylcyclohexyl)(7.84%), 4-oxo-β-isodamascol(5.17%), velleral(3.11%) and Z-α-farnesene(2.00%). The most prevailing major compounds identified in the polar fraction of the M/C extraction were α-D glucopyranoside, 1,3,4,6 tetrakis-O-(TMS)(trimethylsilyl)-β-D-fructofuranosyl 2,3,4,6-tetrakis-O-(TMS)-(38.08%), d-glucose, 2,3,4,5,6-pentakis-O-(TMS)-, O-methyloxime(14.61%), D-fructose, 1,3,4,5,6-pentakis-O-(TMS)-, O-methyloxime(5.28%), isocitric acid(TMS)(3.06%), oxalic acid, bis(TMS) ester(2.96%), hexadecanoic acid, TMS ester(2.16%), citric acid, ethyl ester, tri-TMS(1.91%) and butanedioic acid, [(TMS) oxy]-, bis(TMS) ester(1.14%); whereas in the nonpolar extract, among the major compounds detected were cycloisolongifolene, 8, 9-dehydro-9-formyl(15.70%), propioli

Plants produce a myriad of structurally and functionally diverse metabolites that play many different roles in plant growth and development and in plant response to continually changing environmental conditions as well as abiotic and biotic stresses. This metabolic diversity is, to a large extent, due to chemical modification of the basic skeletons of metabolites. Here, we review the major known plant metabolite modifications and summarize the progress that has been achieved and the challenges we are facing in the field. We focus on discussing both technical and functional aspects in studying the influences that various modifications have on biosynthesis, degradation, transport, and storage of metabolites, as well as their bioactivity and toxicity. Finally, we discuss some emerging insights into the evolution of metabolic pathways and metabolite functionality.

Effect and Analysis of Mechanized Production on Ratooning Ability of Guitang 47

[Objectives] This study was conducted to explore the effect of machine harvesting on ratooning ability of Guitang 47, and to provide basis for mechanized production of Guitang 47. [Methods] On the basis of mechanized planting and management of newly planted sugarcane Guitang 47, a comparative test of machine harvesting and manual harvesting was conducted, during which the plant generation rate, sugarcane yield, sucrose content and related agronomic traits of ratoon cane were continuously investigated in two years after machine harvesting of the newly planted sugarcane. [Results] There were no significant differences in the plant generation rate, plant height, stalk diameter, millable cane, sugarcane yield and sucrose content from manual harvesting of Guitang 47 in the first and second years after machine harvesting. In the second year after machine harvesting, the yields of ratoon cane and the sugar content reached 101.70 and 15.32 t/hm^2, respectively, which were equivalent to those of manual harvesting. [Conclusions] Guitang 47 has strong ratooning ability and lodging resistance and strong resistance to rolling and crushing, and is adaptive to full mechanized production. It should be promoted and accelerated in sugarcane areas lacking labor to reduce costs and increase benefits.

Phylogenetic Analysis of Epibacterial Communities on the Surfaces of Four Red Macroalgae

Macroalgal surfaces are prone to being attached by bacteria. Epibacterial community structures on marine macroalgae are host-specific but temporally and spatially variable. In this study, we investigated the structure of epibacterial communities on the surfaces of four red macroalgae, Gracilaria lemaneiformis, Gloiopeltisfurcata, Mazzaella sp. and Porphyra yezoensis, by analyzing the sequences of 16S rRNA gene libraries. Healthy individuals of all macroalgae species were collected in winter from a farm at Dalian, China. The results showed that the epibacterial communities were mainly dominated by ct-Proteobacteria, 7-Proteobacteria and Bacteroidetes. Deinococcus-Thermus, Spirochaetes and e-Proteobacteria were also found. The majority of cloned sequences shared the greatest similarity to those of culturable organisms. A large portion of sequences from the ct-Proteobacteria homed in Roseobacter clade, i.e., genera Ahrensia, Roseovarius, Litoreibacter, Octadecabacter, Thaiassobacter and Sulfitobacter, while members of Bacteroidetes mainly belonged to family Flavobacteriaceae. The cloned sequences could be separated into 66 OTUs at 0.01 distance value, and rare common OTUs were found among libraries. At genus level, Pseudoa#eromonas dominated Gr. lemaneiformis and GI. furcata libraries, accounting for 72.2% and 47.3%, respectively. Sulfitobacter dominated P. yezoensis library, accounting for 35.4%. A previously undefined cluster within Deinococcus-Thermus dominated Mazzaella sp. library, accounting for 24.6% of the all. These results indicated that a broad range of bacteria inhabited the surfaces of these macroalgae.

Climate change mitigation is a major challenge of human ***,to this end,many countries including China are committed to achieving carbon neutrality within a few *** is a major sugarcane and sugar beet producing country and has one of the largest carbon footprint for sugarcane and sugar beet production globally.A comprehensive study was conducted on sugarcane and sugar beet crops grown in China for greenhouse gas(GHG)emissions mitigation potential,economic crop production from a sustainable sugar production ***-term trend analysis showed that yield and GHG emissions of sugarcane and sugar beet crops increased but the ratio of income to cost *** equation model analysis revealed nitrogen fertilizer and labor as the major drivers of GHG emissions for both sugarcane and sugar *** sugarcane and sugar beet,the path coefficient of N fertilizer were‒0.964 and‒0.835 and that of labor were 0.771 and 0.589,*** transitioning the current cropping system to an improved model with optimized labor,N input and machinery use,the GHG emissions and total annual cost of sugarcane and sugar beet production can be reduced by 32%and 24%,respectively,by 2030,compared to a business-as-usual *** is the first integrated and comparative study of environmental and economic sustainability of sugarcane and sugar beet production in *** findings will enable all stakeholders of Chinese sugarcane and sugar beet industries to transform them into environmentally and economically sustainable sugar production.
