Phytodiversity and medicinal plant distribution in pasturelands of North Western Himalaya in relation to environmental gradients

This study aimed at elucidating phytodiversity of pasturelands of North-western Kashmir Himalayan biotic province in relation to various environmental gradients, with added focus on assessing distribution of important threatened medicinal plant taxa across surveyed pasturelands. A total of 16 sites that spanned across a broad altitudinal gradient(2502-4120 m) were selected. Phytosociological data like density, cover, frequency and abundance of vascular flora were collected and processed using gradient analysis. Soil moisture, nitrogen, organic carbon, acidity and electrical conductivity, altitude, degree of slope and human disturbance were recorded from each site. Analysis of species composition showed a total of 293 species of vascular plants, belonging to 192 genera and 64 families recorded from surveyed sites. Rank abundance exhibited that majority of species are locally rare and patchily distributed, with over 60% of species recording frequencies between 0%-20% and none showing frequencies more than 80%. Species richness showed significant variations and ranged from 20(Parihasmaidan) to 114(Nagbarren) for macro and 3.53±2.11(Thajwas) to 8.76± 1.01(Marsar) for micro-scale of measurement. The ShannonWiener(H') and Simpson's diversity(Ds) indices also varied greatly and were recorded highest(H'= 2.76 & Ds= 0.36) for Marsar and lowest(0.87 & 0.62) for Parihasmaidan. The diversity attributes suggested that pasturelands with lower species richness and diversity on macro-scale were not always species poor and less diverse on micro-measurement scale. The life form groupings highlighted the hemicryptophytic character of vegetation and species with highest importance value index were Sibbaldia cuneata(5.71), Rumex nepalensis(2.89), Juniperus wallichiana(2.88), Cynodon dactylon(2.83), Poa annua(2.49) and Sambucus wightiana(2.41). Expression levels of aminopeptidase-N genes in the lightbrown apple moth, Epiphyas postvittana

Five aminopeptidase-N genes （EpAPN1-5） of the tortricid moth Epiphyas postvittana have been isolated from a midgut expressed sequence tag （EST） library. Relative RNA expression of these genes was measured by quantitative reverse transcription polymerase chain reaction using actin as a reference gene. Measurements were made on various tissues of fifth instar larvae, over all stages of the life cycle and under differing dietary conditions： different protein sources and levels, and in presence of trypsin and metalloprotease inhibitors. Gene expression for all five genes was the greatest in midgut tissue, but was also detected in the hindgut, fat body and Malpighian tubules. EpAPN4 was consistently the highest expressed, with EpAPN3 at about half that level; EpAPN5 was the least expressed. During larval stages expression was high, generally increasing over the instars, after an early peak as neonates or first instars. Expression in other life stages was much lower. Males and females showed differing expression in the pupal and adult stages： female expression was higher in the pupa, this reversed in the adult. Gene expression levels and ratios both changed with diet. A natural apple leaf diet depressed levels. Diets with the most impaired amino acid balance induced the greatest change; generally EpAPN1 increased by the greatest proportion. The addition of proteinase inhibitor also increased gene expression, and it was noteworthy that the trypsin inhibitor addition, which has been shown to double aminopeptidase activity, also doubled levels of gene expression.

Physiological mechanisms of resistance to cold stress associated with 10 elite apple rootstocks

A study was conducted in attempting to identify the cold-resistant apple rootstocks and to establish a comprehensive evaluation system. In this study, 10 elite apple dwarfing rootstocks(GM256, JM7, M26, M7, SC1, SH1, SH38, SH6, M9, and T337) were employed for the experiment and the following parameters were investigated under different low temperature stress conditions(0, –15, –20, –25, –30, and –35°C): the changes of the relative electrical conductivity(REC), anthocyanin content, protein content, soluble sugar content, soluble starch content, proline content, malondialdehyde(MDA) content, superoxide dismutase(SOD) activity, and peroxidase(POD) activity of the dormant branches. The inflection temperature that could represent the plant tissue semi-lethal temperature(LT) was obtained by the measurements of REC. The LTwas used to evaluate eight other indices. The results showed that there was no significant correlation between LTand POD activity as well as between the soluble sugar, protein and proline contents at 0 and –15°C. Soluble starch content at 0 and –15°C and anthocyanin content at –15–(–30)°C were significantly but negatively correlated to the LT50 and the MDA content at 0–(–20)°C was significantly positively correlated to the LT. Statistical analysis based on principal component analysis and LT50 showed that cold resistant apple rootstocks in the decreasing order from high to low as GM256, SH6, SH38, SH1, SC1, M26, M7, JM7, T337, and M9.

Characterization of two novel heat shock protein 70s and their transcriptional expression patterns in response to thermal stress in adult of Frankliniella occidentalis (Thysanoptera:Thripidae)

Heat shock protein 70（HSP70） is one of the most important members in the heat shock protein family, and plays important roles in the thermotolerance of insect. To explore the molecular mechanism of thermotolerance of Frankliniella occidentalis adults, the difference in the expression of HSP70s in F. occidentalis male or female adults under the thermal stress was studied under the laboratory conditions. Two full length c DNAs of HSP70s gene（Fohsc704 and Fohsc705） were cloned from F. occidentalis by using RT-PCR and RACE. The genomic sequence was demonstrated by genomic validation, and the position and size of the intron were analyzed by sequence analysis of c DNA. Real-time PCR was used to analyze the HSP70 expression patterns. The c DNA of Fohsc704 and Fohsc705 possessed 2 073 and 1 476 bp which encoded 690 and 491 amino acids（aa） with a calculated molecular weight of 75 and 54 k Da, respectively. Four introns in Fohsc704 and six introns in Fohsc705 protein were found. However, the HSP70 protein sequences in our study were ended with EKKN and GIFL, which were different from the reported Fo HSP70s. Various expression patterns of Fohsc704 and Fohsc705 were found in both genders of F. occidentalis under thermal stress. The expression of Fohsc704 and Fohsc705 reached to the highest level at –12 and –8°C in male adults, respectively, and Fohsc705 expressed the highest level at 33°C in female adults. In conclusion, HSP70s of F. occidentalis in our study are novel heat shock proteins. There were difference in expression patterns of the two hsc70s in genders of F. occidentalis, and the two HSP70s play important roles in the thermotolerance of F. occidentalis.

Chemical pre-treatments improve seed germination and seedling growth in Semecarpus kurzii:an ethnomedicinally important plant

Semecarpus kurzii *** an important ethnomedicinal plant used for curing tumours,inflammation,fever and pain,and is also a source of iso ricinoleic *** the present investigation,effect of chemical pretreatments on germination and seedling growth revealed significant *** pre-treatments could improve germination and treatment with KNO_3 （9.9 mM）was significantly superior（94%） to the untreated control（76%）.Mean daily germination varied among the treatments（0.90–4.14 seeds per day）,when compared with 2.27 seeds per day in *** germination time was significantly reduced from 22.14 days（control） to 16.40 days（19.8 mM KNO_3） and 17.16 days（1.45 mM GA_3）.Treatment with KNO_3（19.8 mM） and GA_3（1.45 and 2.90 mM）significantly reduced the time for 50% *** with higher concentrations of GA_3 and KNO_3 exhibited significantly faster germination initiation（11.0 and 12.0 days,respectively） over the control（17.0 days）.Seedling vigour index was not improved by *** treatments did not improve shoot growth,collar thickness,or number of seedling leaves; however,root growth was significantly improved over *** with GA_3（2.90 mM） promoted leaf elongation but reduced leaf width *** with KNO_3 and GA_3 could be recommended to obtain healthy seedlings for establishing new plantations of this important medicinal plant species.

Physiological changes and expression characteristics of ZIP family genes under zinc def iciency in navel orange(Citrus sinensis)

Zinc （Zn） deficiency is widespread among citrus plants, but information about the mechanisms for Zn deficiency response in these plants is scarce. In the present study, different navel orange （Citrus sinensis （L.） Osbeck） leaves with various yellowing levels were sampled in our experimental orchard, and upon estimation of nutrient contents, Zn deficiencies were diagnosed as mild, moderate, and severe. Further analysis of chlorophyll content, photosynthetic characteristics, antioxidant enzyme activities, and expression levels of Zn/Iron-regulated transporter-like protein （ZIP） family genes were conducted in the sampled Zn-deficient leaves. The results showed that chlorophyll contents and net photosynthetic rate （Pn） seemed to decrease with reduced Zn contents. In addition, comparison of severe Zn-deficient and normal leaves revealed that activities of peroxidase （POD） and catalase （CAT） increased significantly, whereas that of Zn-containing enzymes such as Cu/Zn superoxide dismutase （Cu/Zn-SOD） significantly reduced with decreasing Zn contents. As expected, expression of the ZIP family genes, ZIP1, ZIP3, and ZIP4, was induced by Zn deficiencies. These results deepen our understanding of Zn deficiency in citrus plants as well as provide useful preliminary information for further research.

Proteomic Analysis of Fruit Bending in Cucumber (Cucumis sativus L.)

In cucumber, fruit shape is an important quality criterion, and fruit bending is known to limit growth, yield, and taste. To investigate the post-transcriptional changes that regulate fruit bending and to better understand the underlying molecular mechanisms, we generated a proteomic profile of the abdomen and back of cucumber bending fruit. Two-dimensional gel electrophoresis （2-DE） allowed the detection of approximately 900 distinct protein spots in each gel, 32 of which were differentially expressed in the abdomen and back of bending cucumber fruit. Ten of the differentially expressed proteins were analyzed using matrix-assisted laser ionization time of flight mass spectrometry （MALDI-TOF/MS）. A search of primary databases showed that the identified proteins are involved in various metabolic processes and cellular responses, including photosynthesis metabolism, energy metabolism, defense and stress response, and regulation. The identified proteins included large subunits of ribulose-1,5-bisphosphate carboxylase/oxygenase, which are involved in photosynthesis and photorespiratory metabolism, and isocitrate dehydrogenase, which is involved in the tricarboxylic acid cycle. It is possible that imbalances in catabolic and anabolic processes directly affect the bending of cucumber fruit. The predicted function of the cobalamin- independent methionine synthase isozyme is closely related to ethylene biosynthesis; fruit bending may be regulated by ethylene, or by ethylene signaling crosstalk during fruit development. The 14-3-3 protein is usually considered to be a regulation-related protein, which plays a role in regulating cell hyperplasia, cell differentiation during growth, and apoptosis during senescence. Involvement of guanosine triphosphate （GTP）-binding proteins in signal transmission is known to regulate the development of cells in cucumber fruits and to play a role in fruit shape variation. Haploid Induction via In vitro Gynogenesis in Tomato(Solanum lycopersicum L.)

In order to determine the potential for haploid induction via in vitro gynogenesis in tomato, the ovules and protoplasts of embryo sacs from the hybrids Zhongza 101 and Zhongza 105 were cultured. An efficient method of ovule isolation was established in this study. Using this method, 100-150 ovules could be isolated from one ovary. Isolated ovules were cultured on three induction media to induce gynogenesis in vitro. During culture, ovules were enlarged markedly, with opaque white color. When observed microscopically, there were cell divisions and cell clumps in embryo sacs. Subsequently, the cell clumps in embryo sacs ceased growth, likely because the integument grew faster than embryo sacs did and hindered the fiarther development of embryo sacs. Therefore, subsequent callus morphogenesis might be originated from the integument. Thousands ofcalli from the two tomato varieties were obtained. Five diploid plants were regenerated after 15 months of subculturing. To eliminate the hindering effect of integument on embryo sac cells, the protoplasts of embryo sacs were prepared and cultured. After 48 hours of culture, the protoplasts of embryo sacs doubled in size and gradually formed clusters of cells. These results suggested that gynogenesis might be a potential way for haploid induction in tomato.

Effects of Rest Grazing on Organic Carbon Storage in Stipa grandis Steppe in Inner Mongolia, China

This study was aimed to evaluate the potential effects of rest grazing on organic carbon storage in Stipa grandis steppe of Inner Mongolia, China. Using potassium dichromate heating method, we analyzed the organic carbon storage of plant and soil in Stipa grand＆ steppe after rest grazing for 3, 6, and 9 yr. The results indicated that as the rest grazing ages prolonged, the biomass of aboveground parts, litter and belowground plant parts （roots） of the plant communities all increased, meanwhile the C content of the biomass increased with the rest grazing ages prolonging. For RG0, RG3a, RG6a, and RG9a, C storage in aboveground vegetation were 60.7, 76.9, 82.8 and 122.2 g C m2, respectively; C storage of litter were 5.1, 5.8, 20.4 and 25.5 g C m^-2, respectively; C storage of belowground roots （0-100 cm） were 475.2, 663.0, 1 115.0 and 1 867.3 g C m^-2, respectively; C storage in 0-100 cm soil were 13.97, 15.76, 18.60 and 32.41 kg C m^-2, respectively. As the rest grazing ages prolonged, the organic C storage in plant communities and soil increased. The C storage ofbelowground roots and soil organic C was mainly concentrated in 0-40 cm soil body. The increased soil organic C for RG3a accounted for 89.8% of the increased carbon in vegetation-soil system, 87.2% for RG6a, and 92.6% for RG9a. From the perspective of C sequestration cost, total cost for RG3a, RG6,, and RG9a were 2 903.4, 5 806.8 and 8 710.2 CNY haq, respectively. The cost reduced with the extension of rest grazing ages, 0.15 CNY kg^-1 C for RG3a, 0.11 CNY kg-~ C for RG6a and 0.04 CNY kg℃ for RG9a. From the growth characteristics of grassland plants, the spring was one of the two avoided grazing periods, timely rest grazing could effectively restore and update grassland vegetation, and was beneficial to the sustainable use of grassland.
