Adenine nucleotide translocase 2(Ant2)is required for individualization of spermatogenesis of Drosophila melanogaster

Successful completion of spermatogenesis is crucial for the perpetuation of the *** Drosophila,spermatid individualization,a process involving changes in mitochondrial structure and function is critical to produce functional mature ***2,encoding a mitochondrial adenine nucleotide translocase,is highly expressed in male testes and plays a role in energy metabolism in the ***,its molecular function remains ***,we identified an important role of Ant2 in spermatid *** Ant2 knockdown testes,spermatid individualization complexes composed of F-actin cones exhibited a diffuse distribution,and mature sperms were absent in the seminal vesicle,thus leading to male *** most striking effects in Ant2-knockdown spermatids were decrease in tubulin polyglycylation and disruption of proper mitochondria derivatives *** apoptotic cells were also observed in Ant2-knockdown *** further investigate the phenotype of Ant2 knockdown in testes at the molecular level,complementary transcriptome and proteome analyses were *** the mRNA level,868 differentially expressed genes were identified,of which 229 genes were upregulated and 639 were downregulated induced via Ant2 ***-labeling proteome analysis revealed 350 differentially expressed proteins,of which 117 proteins were upregulated and 233 were *** expression of glutathione transferase(GstD5,GstE5,GstE8,and GstD3),proteins involved in reproduction were significantly regulated at both the mRNA and protein *** results indicate that Ant2 is crucial for spermatid maturation by affecting mitochondrial morphogenesis.

Population dynamics of marine ciliate Euplotes vannus (Protozoa, Ciliophora) in different artificial seawaters

To study population dynamics of marine ciliates in different artificial seawaters （ASW）, the population growth dynamics of a common marine ciliate Euplotes vannus were investigated using beef extract media and rice media for five types of ASW and natural seawater （NSW）. The results show that：（1） the population growth rate was in the order of NSW〉Flack ASW〉Nakamula ASW〉Schmadz ASW〉Oshima ASW〉Subow ASW and was considerably higher in rice media than in beef extract media （apart from Subow ASW）; （2） the maximum density of E. vannus in stationary phase in each treatment was ranked as Hack ASW〉Nakamula ASW〉Schmadz ASW〉NSW〉Oshima ASW〉Subow ASW, and was again higher in rice media than in beef extract media （except for Subow ASW）; （3） the exponential and stationary phases were longer in rice media than in beef extract media; （4） strains of E. vannus that had been domesticated for 〉1 year in ASW grew significantly slower, with lower maximum density and longer stationary phase than those isolated and maintained in NSW. It was demonstrated that：（1） E. vannus may grow well in Flack, Nakamula and Schmads ASW compared with NSW （mainly in terms of growth rate）; and （2） Oshima ASW is the preferred choice for stock cultures of E. vannus, but the ASWs Flack, Nakamula and Schmadz are preferred for mass culture. These findings suggest that these three ASWs are effective for the cultivation of marine protozoa for experimental studies on ecology, toxicology and molecular biology.

Molecular phylogeny and ultrastructure of a new population of Urostyla grandis(Ciliophora, Hypotrichida) from Harbin, China

A new population of Urostyla grandis, collected from a freshwater pond of northeastern China was investigated by living observation, silver impregnation, and molecular phylogeny based on small subunit ribosomal rRNA(SSU rRNA). Besides, its ultrastructural characteristics were investigated carefully by scanning electron microscopy(SEM) and transmission electron microscopy(TEM) *** descriptions of some inner ultrastructures were provided and some new organelles were also found. Especially, the oral inner microtubule system was illustrated by SEM and TEM for the ?rst time and a new type extrusome was reported. These results may supply complement taxonomic data and ingestion mechanism on ciliates in genus Urostyla.

Ciliates of the heterotrich family Folliculinidae are widespread in various habitats and are distinguished by their transparent loricae of various shapes,conspicuous peristomial lobes,and dimorphic life *** usually attach firmly to the surface of substrates,feed on bacteria and microalgae,and play a significant role in energy flow and material cycling in the microbial food ***,little is known regarding their biodiversity and *** this work,we establish the terminology of the family Folliculinidae and select six crucial features for genus *** on previous studies,we revise the classification of Folliculinidae,supply improved diagnoses for each of the 33 folliculinid genera,and provide a key to their ***,phylogenetic analyses based on small subunit ribosomal DNA(SSU rDNA)sequences revealed that the family is monophyletic and comprises two subclades(subclades I II)which can be identified by the flexibility of their peristomial lobes and the sculpturing of their ***,we investigate the evolutionary relationships of folliculinids using the six chosen generic features.

Insights into the phylogeny of sporadotrichid ciliates (Protozoa, Ciliophora: Hypotricha) based on genealogical analyses of multiple molecular markers

The sporadotrichid ciliates are an especially diverse group. A number of investigators have studied the morphological, morphogenetic, and molecular relationships among members of this group. Despite this, a consistent classification is still lacking and several important questions about the phylogenetic relationships within this group remain unsolved. To improve our understanding of these relationships, we constructed phylogenetic trees using the nucleotide sequences of the small-subunit rRNA (SSrRNA) gene and amino acid sequences of actin I and α-tubulin. Analyses of SSrRNA gene sequences indicated that: 1) the Sporadotrichida sensu Lynn (2008) and the Oxytrichidae are polyphyletic; 2) the Uroleptus species, which are classified to urostylids, formed a sister group with the oxytrichids; 3) Halteria grandinella, which is grouped morphologically with oligotrich species, clustered within the oxytrichids. These results are congruent with previous studies based on SSrRNA gene sequences. However, the amino acid sequences of actin I and α-tubulin yielded different topologies. The main results are: 1) in all phylogenetic trees, the genus Oxytricha was paraphyletic; 2) Uroleptus was sister to a subset of Urostyla and Holosticha, albeit with low supporting values; 3) Halteria grandinella was separated distantly from the Oxytrichidae in trees inferred from actin I amino acid sequences but clustered with oligotrichids in the α-tubulin analysis. The inconsistency among the trees inferred from these different molecular markers may be caused by rapidly accumulated genetic characterizations of ciliates. Further studies with additional molecular markers and sampling of more taxa are expected to better address the relationships among sporadotrichids.

Community structure and biodiversity of free-living marine nematodes in the northern South China Sea

A quantitative study on the community structure and biodiversity of free-living marine nematodes and their relationship with environmental factors in the northern South China Sea were carried out based on the samples collected at five stations in the deep sea （from 313 to 1 600 m） and one station in shallowwaters （87 m） during the cruise in September, 2010. Results showed that the abundance of marine nematodes ranged from 224 to 1 996 ind./（10 cm2）. A total of 69 free-living marine nematode genera, belonging to 26 families and three orders, were identified. The most dominant genera were Sabatieria, Linhystera, Aegialoalaimus and Daptonema according to SIMPER analysis. Results of CLUSTER analysis revealed four types of marine nematode community （or station groups） in the sampling area. In terms of trophic structure, non-selective deposit feeders （1B） and selective deposit feeders （1A） were the dominant trophic types with highest genera numbers and abundances, which implied that organic detritus was the main food source of marine nematodes in the northern South China Sea. The percentage of male nematode was low, ranging from 2.22% to 17.81%, while those of juvenile individuals ranged from 36.99% to 82.09%. For genera level diversity of marine nematodes, Shannon-Wiener diversity indices （H＇） ranged from 3.76 to 4.57 and had highly significant negative correlation with water depth. In general, diversity indices H＇ at the five stations in deep sea （over 200 m） were lower than that at the station in shallow waters （87 m）. BIOENV analysis showed that the most important environmental factor controlling marine nematode communities was water denth.

Improvement of Nannochloropsis oceanica growth performance through chemical mutation and characterization of fast growth physiology by transcriptome profiling

Nannochloropsis oceanica promises to be an industrial-level producer of polyunsaturated fatty acids. In this study, the fastest and slowest growing N. oceanica mutants were selected through N-methyl-N＇-nitro-N-nitrosoguanidine mutation, and two mutant strains and the wild type （WT） subjected to transcriptome profiling. It was found that the 0D680 reads at stationary growth phase of both WT and its mutants were proportional to their cell density, thus indicating their division rate and growth speed during culture. This chemical mutation was effective for improving growth performance, and the fast strain divided faster by upregulating the expression of genes functioning in the cell cycle and downregulating genes involved in synthesis of amino acids, fatty acids, and sugars as well as the construction of ribosome and photosynthetic machinery. However, the relationship among the effected genes responsible for cell cycle, metabolism of fatty and amino acids, and construction of ribosome and photosynthetic machinery remained unclear. Further genetic studies are required for clarifying the genetic/metabolic networks underpinning the growth performance ofN. oceanica. These findings demonstrated that this mutation strategy was effective for improving the growth performance of this species and explored a means ofmicroalgal genetic improvement, particularly in species possessing a monoploid nucleus and asexual reproduction.

Structural Variation Analysis of Mutated Nannochloropsis oceanica Caused by Zeocin Through Genome Re-Sequencing

Zeocin can cause double strand breaks of DNA and thus may be employed as a mutagen. In this study, two strains of Nannochloropsis oceanica, the wild and the Zeocin-tolerant strains, were re-sequenced to verify such function of Zeocin, The results showed that Zeocin can mutate the N. oceanica genome and cause the structural variation. Zeocin either swept away or selected the alleles of genes functioning in ubiquitin-mediated proteolysis, alpha-linolenic acid metabolism, ascorbate and aldarate metabolism, ribosome biogenesis, and circadian rhythm, indicating that N. oceanica may have adjusted its metabolic performances for protein, carbohydrate, and lipid, and changed its ribosome biosynthesis and living rhythm to survive in Zeocin containing medium. In addition, Zeocin caused mutation may have influenced the expression of a set of tanscription factors. It was concluded that Zeocin effectively caused the structural variation of the genome of N. oceanica, and forced the microalgae to select out the alleles of a set of genes around these variations in order to adapt to Zeocin containing medium. Further studies on the genetic basis of the phenotypic adaptation of this haploid and asexual microalga and the application of Zeocin to its genetic improvement are very important.
