metabolic health is highly dependent on intestinal and hepatic handling of dietary and endogenous lipids and *** of lipid and lipoprotein metabolism are commonly observed in patients with insulin resistant states such as obesity,metabolic syndrome,and type 2 ***-idence from both animal models and human studies indicates that a major underlying factor in metabolic or diabetic dysli-pidemia is the overproduction of hepatic and intestinal apoli-poprotein(apo)B-containing lipoprotein *** particles are catabolized down into highly proatherogenic remnants,which can be taken up into the arterial intima and promote plaque *** gut-derived peptides have been identiﬁed as key regulators of energy metabolism;one such peptide is the incretin hormone glucagon-like peptide(GLP)-*** laboratory has previously demonstrated that GLP-1 can signal both centrally and peripherally to reduce postprandial and fasting lipoprotein ***,we have demonstrated that GLP-1 receptor(GLP-1R)agonists can ameliorate diet-induced ***,we published evidence for a novel vagal neuroendocrine signalling pathway by which native GLP-1 may exert its anti-lipemic eﬀ***,we demonstrated a novel role for other gut-derived peptides in regulating intestinal lipoprotein ***,ample evidence supports a key role for GLP-1R on the portal vein aﬀerent neurons and nodose ganglion in modulating intestinal fat absorption and lipoprotein produc-tion and identiﬁes other gut-derived peptides as novel regula-tors of postprandial *** from these data may support identiﬁcation of potential drug targets and the development of new therapeutics targeting treatment of dia-betic dyslipidemia.

The pancreas exerts endocrine and exocrine functions in energy *** neural innervation and immune milieu are both crucial in supporting pancreatic *** neuronal network connects the pancreas with the central nervous system(CNS)and the enteric nervous system(ENS)and sustains metabolic *** nerves in the pancreas are categorized as spinal sensory afferent fibers,vagal sensory afferent nerves,autonomic fibers of both sympathetic and parasympathetic divisions,and fibers from the ENS and intrapancreatic *** innervate different regions and various cell types,which collectively determine physiological *** have established that the diverse pathological conditions,including pancreatitis,diabetes,and pancreatic tumor,are attributed to aberrant immune reactions;however,it is largely not clear how the neuronal network may influence the disease *** by the recent advances illuminating the organ-wide neuronal architecture and the dysfunctions in pancreatic disorders,this review will highlight emerging opportunities to explore the cellular interrelationship,particularly the neuroimmune components in pancreatic health and diseases.

metabolic dysfunction-associated steatotic liver disease(MASLD)is a growing cause of chronic liver diseases worldwide,with a global prevalence of 38%.1 Patients with MASLD are at risk of developing liver-related complications,such as cirrhosis,hepatocellular carcinoma,and extra-hepatic adverse events.1 In recent years,endocrine-disrupting chemicals(EDCs)have been linked to the development and progression of MASLD.

Diabetes has become a serious public health concern worldwide,and China is the epidemic *** China,the prevalence of type 2 diabetes(T2D)was 11.6%among adults aged≥18 years[1].Cardiovascular disease(CVD)occurs earlier and with greater severity in patients with T2D than in individuals without T2D[2].Thus,potential risk factors for screening T2D are needed to prevent such poor clinical outcomes.

A 61-kb biosynthetic gene cluster(BGC),which is accountable for the biosynthesis of hibarimicin(HBM)B from Microbispora rosea *** TP-A0121,was heterologously expressed in Streptomyces coelicolor M1154,which generated a trace of the target products but accumulated a large amount of shunt *** on rational analysis of the relevant secondary metabolism,directed engineering of the biosynthetic pathways resulted in the high production of HBM B,as well as new HBM derivates with improved antitumor *** results not only establish a biosynthetic system to effectively synthesize HBMs-a class of the largest and most complex Type-Ⅱpolyketides,with a unique pseudo-dimeric structure-but also set the stage for further engineering and deep investigation of this complex biosynthetic pathway toward potent anticancer drugs.

DeepSecE:A Deep-Learning-Based Framework for Multiclass Prediction of Secreted Proteins in Gram-Negative Bacteria

Proteins secreted by Gram-negative bacteria are tightly linked to the virulence and adaptability of these microbes to environmental *** identification of such secreted proteins can facilitate the investigations of infections and diseases caused by these bacterial ***,current bioinformatic methods for predicting bacterial secreted substrate proteins have limited computational efficiency and application scope on a genome-wide ***,we propose a novel deep-learning-based framework—DeepSecE—for the simultaneous inference of multiple distinct groups of secreted proteins produced by Gram-negative *** remarkably improves their classification from nonsecreted proteins using a pretrained protein language model and transformer,achieving a macro-average accuracy of 0.883 on 5-fold *** benchmarking suggests that DeepSecE achieves competitive performance with the state-of-the-art binary predictors specialized for individual types of secreted *** attention mechanism corroborates salient patterns and motifs at the N or C termini of the protein *** this pipeline,we further investigate the genome-wide prediction of novel secreted proteins and their taxonomic distribution across~1,000 Gram-negative bacterial *** present analysis demonstrates that DeepSecE has major potential for the discovery of disease-associated secreted proteins in a diverse range of Gram-negative *** online web server of DeepSecE is also publicly available to predict and explore various secreted substrate proteins via the input of bacterial genome sequences.

Precocious Puberty in a 5-Year-Old Girl with a Giant Hypothalamic Hamartoma Discovered Perinatally: Case Report

Background: Hypothalamic hamartoma is a rare non-neoplastic lesion, typically identified in early childhood during investigations for precocious puberty and/or gelastic seizures. However, cases of diagnosis even earlier or during fetal development have been documented. Case Presentation: A newborn girl was diagnosed with hydrocephalus during pregnancy. An MRI revealed a large oval hypothalamic process, which suggested a hypothalamic hamartoma. At the age of 2, she experienced alternating crying and laughing seizures, followed by a decrease in visual acuity. Due to involvement of the optic pathways, surgery was not performed and she underwent antiepileptic medicines and gamma knife radiotherapy. At the age of 5 years and 3 months, she presented with breast development and laboratory tests confirmed central precocious puberty. Quarterly injections of GnRH agonists have since been administered with favorable results. Conclusion: Early-diagnosed hypothalamic hamartomas require close monitoring, by an experienced multidisciplinary, to promptly detect and treat potential complications, especially precocious puberty, and prevent any undesirable impact on final height.

Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

Trimethylamine oxide supplementation differentially regulates fat deposition in liver, longissimus dorsi muscle and adipose tissue of growing-finishing pigs

Trimethylamine oxide(TMAO) is a microbiota-derived metabolite, and numerous studies have shown that it could regulate fat metabolism in humans and mice. However, few studies have focused on the effects of TMAO on fat deposition in growing-finishing pigs. This study aimed to investigate the effect of TMAO on fat deposition and intestinal microbiota in growing-finishing pigs. Sixteen growing pigs were randomly divided into 2 groups and fed with a basal diet with 0 or 1 g/kg TMAO for 149 d. The intestinal microbial profiles, fat deposition indexes, and fatty acid profiles were measured. These results showed that TMAO supplementation had a tendency to decrease lean body mass(P < 0.10) and significantly increased backfat thickness(P < 0.05), but it did not affect growth performance. TMAO significantly increased total protein(TP) concentration, and reduced alkaline phosphatase(ALP) concentration in serum(P < 0.05). TMAO increased the a diversity of the ileal microbiota community(P < 0.05), and it did not affect the colonic microbial community. TMAO supplementation significantly increased acetate content in the ileum, and Proteobacteria and Escherichia-Shigella were significantly enriched in the TMAO group(P < 0.05). In addition, TMAO decreased fat content, as well as the ratio of linoleic acid, n-6polyunsaturated fatty acids(PUFA), and PUFA in the liver(P < 0.05). On the contrary, TMAO increased intramuscular fat content of the longissimus dorsi muscle, whereas the C18:2n6c ratio was increased,and the n-6 PUFA:PUFA ratio was decreased(P < 0.05). In vitro, 1 mmol/L TMAO treatment significantly upregulated the expression of FASN and SREBP1 in C2C12 cells(P < 0.05). Nevertheless, TMAO also increased adipocyte area and decreased the CPT-1B expression in subcutaneous fat(P < 0.05). Taken together, TMAO supplementation regulated ileal microbial composition and acetate production, and regulated fat distribution and fatty acid composition in growing-finishing pigs. These results
