Establishment and application of a surrogate model for human Ebola virus disease in BSL-2 laboratory

The Ebola virus(EBOV)is a member of the Orthoebolavirus genus,Filoviridae family,which causes severe hemorrhagic diseases in humans and non-human primates(NHPs),with a case fatality rate of up to 90%.The development of countermeasures against EBOV has been hindered by the lack of ideal animal models,as EBOV requires handling in biosafety level(BSL)-4 ***,accessible and convenient animal models are urgently needed to promote prophylactic and therapeutic approaches against *** this study,a recombinant vesicular stomatitis virus expressing Ebola virus glycoprotein(VSV-EBOV/GP)was constructed and applied as a surrogate virus,establishing a lethal infection in *** infection with VSV-EBOV/GP,3-week-old female Syrian hamsters exhibited disease signs such as weight loss,multi-organ failure,severe uveitis,high viral loads,and developed severe systemic diseases similar to those observed in human EBOV *** animals succumbed at 2–3 days post-infection(dpi).Histopathological changes indicated that VSV-EBOV/GP targeted liver cells,suggesting that the tissue tropism of VSV-EBOV/GP was comparable to wild-type EBOV(WT EBOV).Notably,the pathogenicity of the VSV-EBOV/GP was found to be species-specific,age-related,gender-associated,and challenge ***,equine anti-EBOV immunoglobulins and a subunit vaccine were validated using this ***,this surrogate model represents a safe,effective,and economical tool for rapid preclinical evaluation of medical countermeasures against EBOV under BSL-2 conditions,which would accelerate technological advances and breakthroughs in confronting Ebola virus disease.

Effects of simulated gut pH environment on bacterial composition and pheromone production of Dendroctonus valens

Bark beetles are an economically and ecologically important insect group,with aggregation behavior and thus host colonization success depends on pheromone-mediated *** some species,such as the major invasive forest pest in China,red turpentine beetle(Dendroctonus valens),gut microbiota participates in pheromone production by converting tree monoterpenes into pheromone ***,how variation in gut microenvironment,such as pH,affects the gut microbial composition,and consequently pheromone production,is *** this study,we fed wild caught *** with 3 different pH media(main host diet with natural pH of 4.7;a mildly acidic diet with pH 6 mimicking the beetle gut pH;and highly acidic diet with pH 4),and measured their effects on the gut pH,bacterial community and production of the main aggregation and anti-aggregation pheromone(verbenone).We further tested the verbenone production capacity of 2 gut bacterial isolates in different pH environments(pH 6 and 4).Compared to natural state or main host diet,feeding on less acidic diet(pH 6)diluted the acidity of the gut,whereas feeding on highly acidic diet(pH 4)enhanced *** changes in gut pH reduced the abundance of dominant bacterial genera,resulting in decreased verbenone ***,the highest pheromone conversion rate of the bacterial isolates was observed in pH mimicking the acidity in beetle *** together,these results indicate that changes in gut pH can affect gut microbiota composition and pheromone production,and may therefore have the potential to affect host colonization behavior.

Multiple isotopes reveal the driving mechanism of high NO_(3)^(-) level and key processes of nitrogen cycling in the lower reaches of Yellow River

The continuous increase of nitrate(NO_(3)^(-))level in rivers is a hot issue in the ***,the driving mechanism of high NO_(3)^(-)level in large rivers is still lacking,which has limited the use of river water and increased the cost of water *** this study,multiple isotopes and source resolution models are applied to identify the driving mechanism of high NO_(3)^(-)level and key processes of nitrogen cycling in the lower reaches of the Yellow River(LRYR).The major sources of NO_(3)^(-)were sewage and manure(SAM)in the low-flow season and soil nitrogen(SN)and chemical fertilizer(CF)in the high-flow *** was the most key process of nitrogen cycling in the ***,in the biological removal processes,denitrification may not occur *** temporal variation of contributions of NO_(3)^(-)sources were estimated by a source resolution model in the *** proportional contributions of SAM and CF to NO_(3)^(-)in the low-flow and high-flow season were 32.5%-52.3%,44.2%-46.2%and 36.0%-40.8%,54.9%-56.9%,*** driving mechanisms of high NO_(3)^(-)level were unreasonable sewage discharge,intensity rainfall runoff,nitrification and lack of nitrate removal *** control the NO_(3)^(-)concentration,targeted measures should be implemented to improve the capacity of sewage and wastewater treatment,increase the utilization efficiency of nitrogen fertilizer and construct ecological *** study deepens the understanding of the driving mechanism of high nitrate level and provides a vital reference for nitrogen pollution control in rivers to other area of the world.

A novel flow electrode capacitive deionization device with spindle-shaped desalting chamber

Flow-electrode capacitive deionization(FCDI)is an innovative technology in which an intermediate chamber plays an important role in the desalination ***,relatively few studies have been conducted on the structures of these intermediate *** this study,we propose a novel flow-electrode capacitive deionization device with a spindle-shaped inlet chamber(S-FCDI).The desalination rate of the S-FCDI under optimal operating conditions was 36%higher than that of the FCDI device with a conventional rectangular chamber(R-FCDI).The spindle-shaped chamber transferred 1.2μmol more ions than the rectangular chamber,based on energy per ***,we performed a detailed analysis of different inlet chamber shapes using computational fluid dynamics *** concluded that S-FCDI has a relatively low flow resistance and almost no stagnation *** provides unique insights into the development of intermediate *** study may contribute to the improvement of the desalination performance in industrial applications of FCDI.

The use of antibiotics in animal production raises great public safety concerns; therefore, there is an urgent need for the development of substitutes for antibiotics. In recent decades, plant-derived feed additives have been widely investigated as antibiotic alternatives for use in animal health and production because they exert multiple biological functions and are less likely to induce resistance *** review summarizes the research history and classification of phytogenic feed additives and their main functions, potential modes of action, influencing factors, and potential negative effects. Further, we highlight the challenges in developing sustainable, safe, and affordable plant-derived antibiotic alternatives for use in livestock production.

Integrated multi-omics reveals the relationship between growth performance,rumen microbes and metabolic status of Hu sheep with different residual feed intakes

Residual feed intake(RFI)is a metric that provides a more accurate measure of feed *** lower the RFI,the higher the feed *** changes in the host microbiome and metabolome contribute to the greater feed efficiency of low RFI(LRFI)*** aim of this study was to explore the differences in rumen microorganisms,rumen metabolites and plasma metabolites of Hu sheep with differing RFI through the microbiome and metabolome.A total of 80 Hu sheep were *** experiment consisted of a 15-d pretrial period and a 128-d experimental *** RFI in the experimental period was calculated for all sheep,and the sheep were screened into high RFI(HRFI,n=8)and LRFI(n=8)*** HRFI and LRFI sheep did not differ in their initial and final body weights,average daily gain and body measurements,but the dry matter intake of LRFI sheep was significantly decreased(28.4%,P<0.001).The sheep with LRFI had higher digestibility of crude protein(P=0.010)and ether extract(P=0.010)compared to HRFI *** concentrations of acetate(P=0.036),propionate(P=0.010),valerate(P=0.027)and total volatile fatty acids(P=0.048)in rumen of LRFI group were higher compared to HRFI *** results of 16S rDNA sequencing indicated that the sheep with LRFI had higher proportions of Prevotella genus in rumen liquid(P=0.031).The rumen metabolome and plasma metabolome results showed that the citrate cycle,pyruvate metabolism and alanine,aspartate and glutamate metabolism processes were more active for sheep in LRFI group,which provided more energy substrate such as malic acid,oxoglutaric acid and citric *** conclusion,sheep with LRFI can utilize feed more efficiently,and the more active energy metabolism pathway and the production of energy substances may account for the higher feed efficiency.

The intrinsic variability of memristor switching behavior can be used as a natural source of randomness,this variability is valuable for safe applications in hardware,such as the true random number generator(TRNG).However,the speed of TRNG is still be further ***,we propose a reliable Ag/SiNx/n-Si volatile memristor,which exhibits a typical threshold switching device with stable repeat ability and fast switching *** volatile-memristor-based TRNG is combined with nonlinear feedback shift register(NFSR)to form a new type of high-speed dual output ***,the bit generation rate reaches a high speed of 112 kb/*** addition,this new TRNG passed all 15 National institute of Standards and Technology(NIST)randomness tests without post-processing steps,proving its performance as a hardware security *** work shows that the SiNx-based volatile memristor can realize TRNG and has great potential in hardware network security.

On equivalence of state-based potential games

In this paper, we explore state-based potential games using the semi-tensor product of ***, applying the potential equation, we derive both a necessary and sufficient condition as well as a sufficient condition to verify whether a state-based game qualifies as a potential game. Next, we present two static equivalence conditions of state-based potential games. We then delve into dynamic equivalence. We propose a criterion that allows us to identify state-based games that are dynamically equivalent to state-based potential games and share similar dynamic properties. Ultimately, we introduce the concept of state-based networked evolutionary games. We provide a necessary and sufficient condition to ensure that a state-based networked evolutionary game can be classified as a state-based potential game.

Background Excessive backfat deposition lowering carcass grade is a major concern in the pig industry,especially in most breeds of obese type *** mechanisms involved in adipogenesis and fat accumulation in pigs remain *** 2-hydroxyisobutyrylation(Khib),is a novel protein post-translational modification(PTM),which play an important role in transcription,energy metabolism and metastasis of cancer cells,but its role in adipogenesis and fat accumulation has not been *** In this study,we first analyzed the modification levels of acetylation(Kac),Khib,crotonylation(Kcr)and succinylation(Ksu)of fibro-adipogenic progenitors(FAPs),myogenic precursors(Myo)and mesenchymal stem cells(MSCs)with varied differentiation potential,and found that only Khib modification in FAPs was significantly higher than that in ***,in parallel with its regulatory enzymes lysine acetyltransferase 5(KAT5)and histone deacetylase 2(HDAC2)protein levels,the Khib levels increased quadratically(P<0.01)during adipogenic differentiation of ***5 knockdown in FAPs inhibited adipogenic differentiation,while HDAC2 knockdown enhanced adipogenic *** also demonstrated that Khib modification favored to adipogenic differentiation and fat accumulation by comparing Khib levels in FAPs and backfat tissues both derived from obese-type pigs(Laiwu pigs)and lean-type pigs(Duroc pigs),***,the expression patterns of KAT5 and HDAC2 matched well to the degree of backfat accumulation in obese-and lean-type *** From the perspective of protein translational modification,we are the first to reveal the role of Khib in adipogenesis and fat deposition in pigs,and provided new clues for the improvement of fat accumulation and distribution as expected via genetic selection and nutritional strategy in obese-type pigs.

Influences of lauric acid addition on performance,nutrient digestibility and proteins related to mammary gland development in dairy cows

Lauric acid(LA)has the possibility to improve milk production in dairy cows by improving mammary gland development,however,the mechanism by which it might regulate mammary gland development is *** influence of LA on milk production,nutrient digestibility and the expression of proteins related to mammary gland development in dairy cows were *** primiparous Holstein dairy cows were divided into 4 groups in a randomized block *** treatments included the control(0 g/d LA per cow),low-LA(100 g/d LA per cow),medium-LA(200 g/d LA per cow),and high-LA(300 g/d LA per cow).Yields of milk,fat-corrected milk,and energy-corrected milk quadratically increased(P<0.05),and yield and content of milk fat linearly increased(P<0.05)with LA *** of C12:0,C18:1 and C20:1 fatty acids in milk fat linearly increased(P<0.05),but that of C16:0 fatty acid linearly decreased(P=0.046).Supplementation of LA led to a linear and quadratical increase(P<0.05)in digestibility of dry matter,organic matter,neutral detergent fibre and acid detergent fibre,and ruminal total volatile fatty acid concentration but a linear reduction(P=0.018)in the ratio of acetate to *** enzymatic activities of ruminal pectinase,xylanase,andα-amylase,and populations of total bacteria and anaerobic fungi increased linearly(P<0.05),while populations of total protozoa and methanogens decreased linearly(P<0.05)with increased LA *** LA addition,blood glucose,triglyceride,estradiol,prolactin,and insulin-like growth factor 1 concentrations increased linearly(P<0.05)and albumin and total protein concentrations increased quadratically(P<0.05).Moreover,addition of 200 g/d LA promoted(P<0.05)the expression of protein involved in mammary gland development and fatty acids *** results suggested that LA addition enhanced milk production and fatty acids synthesis by stimulating nutrient digestion,the expression of proteins associated with milk fat synth
