DNA methylation controlling abscisic acid catabolism responds to light to mediate strawberry fruit ripening

Phytohormones,epigenetic regulation and environmental factors regulate fruit ripening but their interplay during strawberry fruit ripening remains to be *** this study,bagged strawberry fruit exhibited delayed ripening compared with fruit grown in normal light,correlating with reduced abscisic acid(ABA) *** of the key ABA catabolism gene,ABA 8′-hydroxylase FaCYP707A4,was induced in bagged *** light exclusion whole genome DNA methylation levels were up-regulated,corresponding to a delayed ripening process,while DNA methylation levels in the promoter of FaCYP707A4 were suppressed,correlating with increases in transcript and decreased ABA *** indicated FaCRY1,a blue light receptor repressed in bagged fruit and FaAGO4,a key protein involved in RNA-directed DNA methylation,could bind to the promoter of *** interaction between FaCRY1 and FaAGO4,and an increased enrichment of FaAGO4 directed to the FaCYP707A4 promoter in fruit grown under light suggests FaCRY1 may influence FaAGO4 to modulate the DNA methylation status of the FaCYP707A4 ***,transient overexpression of FaCRY1,or an increase in FaCRY1 transcription by blue light treatment,increases the methylation level of the FaCYP707A4 promoter,while transient RNA interference of FaCRY1 displayed opposite *** findings reveal a mechanism by which DNA methylation influences ABA catabolism,and participates in light-mediated strawberry ripening.

Increased glacier melt enhances future extreme floods in the southern Tibetan Plateau

Mountainous areas are of special hydrological concern because topography and atmospheric conditions can result in large and sudden floods,posing serious risks to water-related safety in neighbouring *** Yarlung Zangbo(YZ)River basin is the largest river basin on the Tibetan Plateau(TP),but how floods will discharge in this basin and how the role of glacier melt in floods will change throughout the 21st-century under shared socioeconomic pathways scenarios(SSP2-4.5 and SSP5-8.5)remain ***,we comprehensively address this scientific question based on a well-validated large-scale glacier-hydrology *** results indicate that extreme floods was projected to increase in the YZ basin,and was mainly reflected in increased duration(4–10 d per decade)and intensity(153–985 m^(3)s^(−1)per decade).Glacier runoff was projected to increase(2–30 mm per decade)throughout the 21st-century,but there was also a noticeable decrease or deceleration in glacier runoff growth in the late first half of the century under the SSP2-4.5,and in the latter half of the century under the *** melt was projected to enhance the duration(12%–23%)and intensity(15%–21%)of extreme floods under both SSPs,which would aggravate the impact of future floods on the socioeconomics of the YZ *** effect was gradually overwhelmed by precipitation-induced floods from glacier areas to YZ *** study takes the YZ basin as a projection framework example to help enrich the understanding of future flood hazards in basins affected by rainfall-or meltwater across the TP,and to help policy-makers and water managers develop future plans.

Dear Editor,Utilization of heterosis is fundamental to modern agricultural improvement and contributes significantly to food security and agricultural development(Ouyang et al.,2022).However,heterosis of F,hybrids cannot be stably ***,or asexual reproduction through seeds,offers the potential for the fixation of heterosis in F,hybrids and the formation of clonal offspring,which could have considerable implications for plant breeding and agriculture(Schmidt,2020).But naturally occurring apomixis has never been found in major ***,synthetic apomixis has been successfully generated by inducing incomplete meiotic development combined with parthenogenesis development to produce cloned rice(Oryza sativa)seeds(Khanday et al.,2019;Wang et al.,2019;Vernet et al.,2022;Wei et al.,2023).

Human monoamine oxidase B(hMAO-B)has emerged as a pivotal therapeutic target for Parkinson's *** to adverse effects and shortage of commercial drugs,there is a need for novel,highly selective,and reversible hMAO-B inhibitors with good blood-brain barrier *** this study,a high-throughput at-line nanofractionation screening platform was established with extracts from Chuanxiong Rhizoma,which resulted in the discovery of 75 active compounds,including phenolic acids,volatile oils,and phthalides,two of which were highly selective novel natural phthalide hMAO-B inhibitors that were potent,selective,reversible and had good blood‒brain *** docking and molecular dynamics simulations elucidated the inhibition ***(IC_(50)=103 nmol/L;SI=645)and neocnidilide(IC_(50)=131 nmol/L;SI=207)demonstrated their excellent potential as hMAO-B *** offset the limitations of deactivating enzymes associated with irreversible hMAO-B inhibitors such as *** SH-SY5Y cell assays,sedanolide(EC_(50)=0.962μmol/L)and neocnidilide(EC_(50)=1.161μmol/L)exhibited significant neuroprotective effects,comparable to the positive drugs rasagiline(EC_(50)=0.896μmol/L)and safinamide(EC_(50)=1.079μmol/L).These findings underscore the potential of sedanolide as a novel natural hMAO-B inhibitor that warrants further development as a promising drug candidate.

Mitochondrial toxicity induced by therapeutic drugs is a major contributor for cardiotoxicity,posing a serious threat to pharmaceutical industries and patients’***,mitochondrial toxicity testing is not incorporated into routine cardiac safety screening *** accurately model native human cardiomyocytes,we comprehensively evaluated mitochondrial responses of adult human primary cardiomyocytes(h PCMs)to a nucleoside analog,remdesivir(RDV).Comparison of their response to human pluripotent stem cell-derived cardiomyocytes revealed that the latter utilized a mitophagy-based mitochondrial recovery response that was absent in h ***,action potential duration was elongated in h PCMs,reflecting clinical incidences of RDV-induced QT *** a screen for mitochondrial protectants,we identified mitochondrial ROS as a primary mediator of RDV-induced *** study demonstrates the utility of h PCMs in the detection of clinically relevant cardiac toxicities,and offers a framework for h PCM-based high-throughput screening of cardioprotective agents.

The development and application of the small-grain rice sterile line Zhuo201S(Z201S)has demonstrated its potential for mechanized hybrid rice seed production,leading to significant cost ***,the molecular mechanism responsible for the small-grain size characteristic of Z201S remains *** this study,we conducted a genetic analysis using near-isogenic lines constructed from Z210S,a small-grain rice sterile line,and R2115,a normal-grain *** results revealed that the small-grain trait in Z201S is governed by a single partially dominant gene which also enhances grain *** mapping,we localized the causal gene to the short arm of chromosome 2,within a 113 kb physical region delimited by the molecular markers S2-4-1 and *** analysis and gene expression assays indicated LOC_Os02g14760 as the most likely candidate gene,suggesting that the small-grain size trait of Z201S is controlled by a novel locus that has not been previously identified.

Co-occurrence of per- and polyfluoroalkyl substances, heavy metals and polycyclic aromatic hydrocarbons and their composite impacts on microbial consortium in soil: A field study

This is the first study to report the co-occurrence of per-and polyfluoroalkyl substances(PFASs),heavy metals,and polycyclic aromatic hydrocarbons(PAHs)and their impacts on the native microbial consortium in soil due to the long-term *** PFASs,heavy metals,and PAHs were detected in soil samples collected at 2–6 m below the ground surface at different sampling locations in a steel-making *** total concentrations of PFASs varied from 6.55 to 19.79 ng g^(-1),with perfluorooctane sulfonate(PFOS),perfluorobutane sulfonate,and 6:2 chlorinated polyfluorinated ether sulfonate(alternative of PFOS)being the predominant *** concentrations of arsenic,cadmium,and lead were detected in the ranges of 4.40–1270.00,0.01–8.67,and 18.00–647.00 mg kg^(-1),respectively,and the concentration of total PAHs was detected in the range of 1.02–131.60 mg kg^(-1).The long-term exposure to mixed contaminants of PFASs,heavy metals,and PAHs led to lower richness and diversity of microbial communities in *** soil bacterial communities were mainly composed of Pseudomonas,norank_p_GAL15,Leptothrix,norank_o_Rokubacteriales,and *** between soil environmental factors and microbial communities indicated that cation exchange capacity and total phosphorus were two key factors in shaping the composition of native microbial ***,Arthrobacter,Leptothrix,and Sphingobium were found to be significantly positively correlated with PFAS concentrations,indicating that these genera could tolerate the stress exerted by PFASs,along with the stress imposed due to the presence of heavy metals or/and PAHs.

Near-infrared(NIR)transparent inverted all-inorganic perovskite solar cells(PSCs)are excellent top cell candidates in tandem *** essential challenge is the replacement of metal contacts with transparent conductive oxide(TCO)electrodes,which requires the introduction of a buffer layer to prevent sputtering *** this study,we show that the conventional buffers(i.e.,small organic molecules and atomic layer deposited metal oxides)used for organic-inorganic hybrid perovskites are not applicable to all-inorganic perovskites,due to non-uniform coverage of the vulnerable layers underneath,deterioration upon ion bombardment and moisture induced perovskite phase transition,A thin film of metal oxide nanoparticles by the spin-coating method serves as a non-destructive buffer layer for inorganic ***-inorganic inverted near-infrared-transparent PSCs deliver a PCE of 17.46%and an average transmittance of 73.7%between 780 and 1200 *** combination with an 18.56%Cu(In,Ga)Se_(2) bottom cell,we further demonstrate the first all-inorganic perovskite/CIGS 4-T tandem solar cell with a PCE of 24.75%,which exhibits excellent illumination stability by maintaining 86.7%of its initial efficiency after 1400 *** non-destructive buffer lays the foundation for efficient and stable NIR-transparent inverted inorganic perovskite solar cells and perovskite-based tandems.

Knockdown of ATRX enhances radiosensitivity in glioblastoma

Background Glioblastoma are highly malignant type of primary brain tumors. Treatment for glioblastoma multiforme (GBM) generally involves surgery combined with chemotherapy and radiotherapy. However, the development of tumoral chemo- and radioresistance induces complexities in clinical practice. Multiple signaling pathways are known to be involved in radiation-induced cell survival. However, the role of alpha-thalassemia X-linked mutant retardation syndrome (ATRX), a chromatin remodeling protein, in GBM radioresistance remains *** In the present study, the ATRX mutation rate in patients with glioma was obtained from The Cancer Genome Atlas, while its expression analyzed using bioinformatics. Datasets were also obtained from the Gene Expression Omnibus, and ATRX expression levels following irradiation of GBM were determined. The effects of ATRX on radiosensitivity were investigated using a knockdown *** The present study demonstrated that the ATRX mutation rate in patients with GBM was significantly lower than that in patients with low-grade glioma, and that patients harboring an ATRX mutation exhibited a prolonged survival, compared with to those harboring the wild-type gene. Single-cell RNA sequencing demonstrated that ATRX counts increased 2 days after irradiation, with ATRX expression levels also increasing in U-251MG radioresistant cells. Moreover, the results of in vitro irradiation assays revealed that ATRX expression was increased in U-251MG cells, while ATRX knockdown was associated with increased levels of *** High ATRX expression levels in primary GBM may contribute to high levels of radioresistance. Thus ATRX is a potential target for overcoming the radioresistance in GBM.
