Development and characterization of 3D-printed electroconductive pHEMA-co-MAA NP-laden hydrogels for tissue engineering

tissue engineering(tE)continues to be widely explored as a potential solution to meet critical clinical needs for diseased tissue replacement and tissue *** this study,we developed a poly(2-hydroxyethyl methacrylate-co-methacrylic acid)(pHEMA-co-MAA)based hydrogel loaded with newly synthesized conductive poly(3,4-ethylene-dioxythiophene)(PEDOt)and polypyrrole(PPy)nanoparticles(NPs),and subsequently processed these hydrogels into tissue engineered constructs via three-dimensional(3D)*** presence of the NPs was critical as they altered the rheological properties during ***,all samples exhibited suitable shear thinning properties,allowing for the development of an optimized processing window for 3D *** were 3D printed into pre-determined disk-shaped configurations of 2 and 10 mm in height and diameter,*** observed that the NPs disrupted the gel crosslinking efficiencies,leading to shorter degradation times and compressive mechanical properties ranging between 450 and 550 *** conductivity of the printed hydrogels increased along with the NP concentration to(5.10±0.37)×10^(−7)S/*** vitro studies with cortical astrocyte cell cultures demonstrated that exposure to the pHEMA-co-MAA NP hydrogels yielded high cellular viability and proliferation ***,hydrogel antimicrobial studies with staphylococcus epidermidis bacteria revealed that the developed hydrogels affected bacterial *** together,these materials show promise for various tE strategies.

Ray tracing Particle Image Velocimetry(Rt-PIV)is an optical technique for high resolution velocity measurements in challenging optical systems,such as transparent packed beds,that uses ray tracing to correct for distortions introduced by transparent geometries in the light *** ray tracing based correction is a post processing step applied to the raw PIV particle images before classical PIV *** this study,Rt-PIV is performed in the top layer of a body centred cubic(bcc)sphere packing with gaseous flow,where optical access is obtained by the use of transparent N-BK7 glass balls with a diameter of d=40 ***-PIV introduces new experimental and numerical challenges,for example a limited field of view,illumination difficulties,a very large required depth of field and high sensitivity to geometric parameters used in the ray tracing *** challenges and their implications are the main scope and discussed in the present ***,the validation of the ray tracing reconstruction step is presented and examples for the obtained corrected vector fields in a packed bed are *** results show the strength of the method in reconstructing velocity fields behind transparent spheres that would not have been accessible by optical measurement techniques without the ray tracing correction.

On-the-fly precision spectroscopy with a dual-modulated tunable diode laser and Hz-level referencing to a cavity

Advances in laser spectroscopy have enabled many scientific breakthroughs in physics, chemistry,biology, and astronomy. Optical frequency combs pushed measurement limits with ultrahigh-frequency accuracy and fast-measurement speed, while tunable-diode-laser spectroscopy is used in scenarios that require high power and continuous spectral coverage. Despite these advantages of tunable-diode-laser spectroscopy, it is challenging to precisely determine the instantaneous laser frequency because of fluctuations in the scan speed. Here, we demonstrate a simple spectroscopy scheme with a frequencymodulated diode laser that references the laser on-the-fly to a fiber cavity. the fiber cavity's free spectral range is on-the-fly calibrated with sub-10-Hz frequency precision. We achieve a relative precision of the laser frequency of 2 × 10-8for an 11-tHz frequency range at a measurement speed of 1 tHz∕s. this is an improvement of more than 2 orders of magnitude compared to existing diode-laser-spectroscopy *** scheme provides precise frequency calibration markers, while simultaneously tracking the instantaneous scan speed of the laser. We demonstrate the versatility of our method through various applications, including dispersion measurement of a fiber, ultrahigh-Q microresonators, and spectroscopy of a hydrogen fluoride gas cell. the simplicity, robustness, and low cost of this spectroscopy scheme are valuable for out-of-the-lab applications like lidar and environmental monitoring.

Experimental and numerical investigation to elucidate the fluid flow through packed beds with structured particle packings

the present paper presents an experimental and numerical investigation of the dispersion of the gaseous jet flow and co-flow for the simple unit cell(SUC)and body-centred cubic(BCC)configuration of particles in packed *** experimental setup is built in such a way that suitable and simplified boundary conditions are imposed for the corresponding numerical framework,so the simulations can be done under very similar conditions as the ***,a porous plate is used for the co-flow to achieve the uniform velocity and the fully developed flow is ensured for the jet *** SUC and BCC particle beds consist of 3D-printed spheres,and the non-isotropy near the walls is mostly eliminated by placing half-spheres at the channel *** flow velocities are analysed directly at the exit of the particle bed for both beds over 36 pores for the SUC configuration and 60 pores for the BCC configuration,for particle Reynolds numbers of 200,300,and *** particle image velocimetry is experimentally arranged in such a way that the velocities over the entire region at the exit of the packed bed are obtained *** numerical method consists of a state-of-the-art immersed boundary method with adaptive mesh *** paper presents the pore jet structure and velocity field exiting from each pore for the SUC and BCC packed particle *** numerical and experimental studies show a good agreement for the SUC configuration for all flow *** the BCC configuration,some differences can be observed in the pore jet flow structure between the simulations and the experiments,but the general flow velocity distribution shows a good overall *** axial velocity is generally higher for the pores located near the centre of the packed bed than for the pores near the *** addition,the axial velocities are observed to increase near the peripheral pores of the packed *** behaviour is predominant for the BCC configuration as compared to t

Converting tREx-RGB green-channel data to 557.7 nm auroral intensity:Methodology and initial results

the recently deployed transition Region Explorer(tREx)-RGB(red-green-blue)all-sky imager(ASI)is designed to capture“true color”images of the aurora and *** the 557.7 nm green line is usually the brightest emission line in visible auroras,the green channel of a tREx-RGB camera is usually dominated by the 557.7 nm *** this rationale,the tREx mission does not include a specific 557.7 nm imager and is designed to use the RGB green-channel data as a proxy for the 557.7 nm *** this study,we present an initial effort to establish the conversion ratio or formula linking the RGB green-channel data to the absolute intensity of 557.7 nm auroras,which is crucial for quantitative uses of the RGB *** illustrate two approaches:(1)through a comparison with the collocated measurement of green-line auroras from the tREx spectrograph,and(2)through a comparison with the modeled green-line intensity according to realistic electron precipitation flux measurements from low-Earth-orbit satellites,with the aid of an auroral transport *** demonstrate the procedures and provide initial results for the tREx-RGB ASIs at the Rabbit Lake and Lucky Lake *** RGB response is found to be *** conversion ratios or formulas between RGB green-channel data and the green-line auroral intensity are given and can be applied immediately by tREx-RGB data *** methodology established in this study will also be applicable to the upcoming SMILE ASI mission,which will adopt a similar RGB camera system in its deployment.

DPY19L3 promotes vasculogenicmimicry by its C-mannosyltransferase activity

C-mannosylation is a post-translational modification that occurs intracellularly in the endoplasmic *** humans,biosynthesis of C-mannosylation in proteins containing thrombospondin type 1 repeat is catalyzed by the DPY19 family;nonetheless,biological functions of protein C-mannosylation are not yet fully understood,especially in tumor *** mimicry(VM)is the formation of fluid-conducting channels by highly invasive and genetically deregulated tumor cells,enabling the tumors to form matrix-embedded vasculogenic structures,containing plasma and blood cells to meet the metabolic demands of rapidly growing *** this study,we focused on DPY19L3,a C-mannosyltransferase,and aimed to unravel its role in *** of DPY19L3 inhibited the formation of VM in Ht1080 human fibrosarcoma ***-expression of wild-type DPY19L3 recovered VM formation;however,DPY19L3 isoform2,an enzymatic activity-defect mutant,did not restore it,suggesting that the C-mannosyltransferase activity of DPY19L3 is crucial to its ***,the knockdown of DPY19L3 in MDA-MB-231 breast cancer cells hindered its network formation ***,our findings suggest that DPY19L3 is required for VM formation and stipulate the relevance of C-mannosylation in oncogenesis.
