Genetic Diversity Analysis of Badnaviruses Infecting Banana in Burkina Faso

Badnaviruses are serious plant pararetroviruses affecting banana and causes serious economic losses to banana production worldwide. This study aims to examine the variability of BsV and sCBV nature infecting banana in Burkina Faso. Polymerase Chain Reaction (PCR) used the Badna FP/RP specific primers for the RT/Rnase H regions present in badnaviruses. The PCR yielded about 579 bp amplicons from banana infected by BsV and sCBV. The 38 BsV isolates recorded low nucleotide identity ranging from 58.9% - 98.1%. Based on percentage nucleotide sequence identity and phylogenetic analyse, BsV isolates were categorized into four groups: 1, 2, 3 and 4. Group 4 shared 76.9% - 100% identity with BsOL species. However, Groups 1 and 3 recorded a low identity ranging, from 76.8% - 79.2%, 68.8% - 79.7% with BsCV, and 72.8% - 79.0% between Group 2 and BsOLV. Groups 1, 2 and 3 were assigned to a potentially new BsV species. The two sCBV isolates recorded a low nucleotide identity of 68% among themselves indicating high diversity. In addition, sCBV_Cd and sCBV_CE showed high nucleotide identity 91.3% and 58.7% with sCBV_C and sCBV, when they were compared to all published sCBV genotypes. In addition, phylogenetic analysis revealed the segregation of sCBV isolates into two genotypes, sCBV_Cd in C and sCBV_CE segregated in a new genotype namely Z. Recombination analyses showed weak signatures of recombination among some of the BsV and sCBV sequences.

AIM: This study was designed to characterize the en-dothelin pathway in an immortalized human adenocarcino-ma-derived alveolar epithelial cell line (A549) and human umbilical vein endothelial cell line (HUVEC). METHODs: The release of BT-1 and big-ET-1 was measured in the incubation medium of both cell lines. The expression of mRnAs coding for the endothelin iso-forms (hppET-1, -2, -3), the endothelin converting enzymes (hECE-la, b, c, and d) and the hETA and hETB receptors was investigated using RT-PCR. The expression of ECE-1 mRnA in various human tissues and in A549 cells was investigated by northern blot analysis and the subcellular localization of ECE-1 in A549 cells was investigated by immunoblotting using a polyclonal antibody. REsULTs: Under control conditions, HUVEC release both ET-1 and big-ET-1 (ratio 5 to 1) while in A549 cells the big-ET-1 levels were below the threshold of detection. Banana streak virus (BsV) and sugarcane bacilliform virus (sCBV) are two badnaviruses commonly found in all banana growing areas of the world. It is a threat to the production and improvement of Musa germplasm. In Burkina Faso, the presence of badnaviruses was reported in banana producing regions. The objective of this study was to determine the prevalence of BsV and sCBV in banana production areas of Burkina Faso. A survey followed by a symptomatologic study was conducted in banana plantations in 27 localities of the nine main banana producing regions from July to October 2018 and september to december 2020. In all, 251 leaf samples were collected and analysed for BsV and sCBV infection by Indirect Antigen Coated Plate Assay-ELIsA followed by amplification of the RT/Rnase H region using Polymerase chain reaction with Badna FP/RP and sCBV F/R primers, respectively. A variety of symptoms were observed on almost all plant organs which were revealed due to BsV by symptomatologic study. The results of serological and molecular diagnosis revealed a high overall prevalence of BsV in 80.48% of the samples tested. BsV was distributed in seven survey regions out of nine with prevalence ranging from 10% to 100% in north, Centre, Centre West, Hauts Bassins, Cascades, Centre East and Boucle of Mouhoun regions. Very low prevalence was recorded for sCBV in Cascades and East Centre region with 4.35 and 12.5%, respectively. species detection using specific primers to each species revealed three main species: Banana streak Obino l’ewaï virus (BsOLV), Goldfinger virus (BsGFV) and Imové virus (BsIMV) in the samples tested, respectively in the proportions of 23%, 8% and 0.8%. Co-infection between BsV species was also detected.

Morphological Identification of the Main Fungi Associated with sesame in Burkina Faso

several fungal species are responsible for diseases that damage sesame crop and cause production losses. These seed-borne fungi cause serious damage by reducing seed germination. To identify the fungi associated with sesame seeds in Burkina Faso, 72 sesame seed samples were collected from 24 localities in the main production areas (sudanian zone, sudano-sahelian zone, sahelian zone). These samples were submitted for seed health analysis using the blotting paper incubation method. The results obtained revealed that the seed samples were contaminated by various fungi, the main ones being: Fusarium spp. Link: Fr. infecting 97.22% of the samples, Macrophomina phaseolina (Tassi) Goid. (90.28%), Alternaria sesamicola E. Kawamura (83.33%), Curvularia lunata (Wakker) Boedijn (76.39%), Cercospora sesami A. Zimmerm. (73.61%), Phoma sorghina Boerema, Dorenbosch, & Van Kesteren (73.61%), Aspergillus flavus Link: Fr. (69.44%), Cladosporium sphaerospermum Penz. (56.94%), Rhizopus sp. Ehrenb. (40.28%) and A. niger Tiegh. (36.11%). In each seed sample, 0.25% to 94% of the seeds host one or more fungal species. Regarding the climatic zones, the results revealed that A. sesamicola and A. flavus were more represented in the sudanian zone than in the sudano-sahelian and sahelian zones;while P. sorghina, and C. sphaerospermum were more encountered in the sahelian zone than in the other two zones. This study showed that the majority of sesame seeds produced and used by farmers in Burkina Faso host one or more species of fungi at high levels.

Toward an Intelligent system for Taurine Cattle Recognition

Unlike zebus, taurine cattle have the natural ability to resist trypanosomosis, a parasitic disease endemic to the humid areas of West Africa. However, repeated crossbreeding between zebus and taurine cattle is jeopardizing the genetic heritage of the Taurines and their ability to resist trypanosomosis. To strengthen protection and conservation efforts, it is essential to accurately distinguish purebred taurines from crossbreds. In this study, five Machine Learning models were built using morphological data collected from 1968 cattle. These models were trained to determine whether a given individual is purebred taurine or not. The classifiers yielded promising results. The random forest model and RBF Kernel sVM performed the best with up to 86% and 85% accuracy respectively. Moreover, the study of the correlation coefficients and the feature importance scores allowed us to define the most discriminating morphological traits.

AIM: To determine in the hamster cheek pouch whether or not the changes in plasma extravasation induced by vascular endothelial growth factor (VEGF) could be affected by fructose diet. METHODs: Hamsters were subjected to control drinking water or to water containing fructose (10％) for 18 weeks. REsULTs: The fructose diet induced a small but significant increase in glycemia (0.80±0.11 and 1.09±0.15, n=8 and 9 for control and fructosetreated animals, respectively, Pnin-induced plasma extravasation was not affected by the fructose diet while the effects of VEGF were markedly increased (maximal number of leakage sites: 76±20 and 126±55, n=8 and 9 for control and fructose-treated animals, respectively, PnCLUsIOn: Even moderate changes in glycemic levels can produce profound alteration in the VEGF response.

Monitoring Forest Recovery in Protected Forests of northern Côte d’Ivoire Using Landsat Imagery and Intensity Change Analysis

In this paper, the initiatives of reforestation of the national forests of the north of the Côte d’Ivoire were examined using geomatics and the analysis of change of intensity by taking the case of the protected Forest of Badénou (PFB). A spatial analysis based on multi-spectral and multi-temporal Landsat imagery was carried out to assess land cover changes in the (PFB) over the past two decades and determine whether patterns of change in terms of the intensity of gains/losses of each of the land cover classes were active or dormant between the period before (2000-2013) and after (2013-2019) the reforestation initiative. Five main classes were identified: forest (dry deciduous and gallery forests), tree savannah, shrub/grassy savannah (including agricultural lands), bare lands (bare soils and degraded land areas), and water course. All classes were satisfactorily classified, with an excellent producer’s and user’s and overall accuracies and very good Kappa coefficients. The results showed that between 2000 and 2019, the forest cover in the PFB increased from 7778 ha to 5054 ha, a decrease was marked between 2000 and 2013 of approximately 60% compared to its size in 2000, while a slight increase between 2013 and 2019 (4645 ha to 5054 ha) i.e. around 9%) certainly due to the reforestation since 2016. As for the annual intensities of change for each class in both study periods, changes (gain or loss) in forest and tree savanna were relatively dormant after reforestation, while annual bare land gain was relatively active and marked, indicating that degradation of forests remains a threat to the sustainability of the PFB. Forest degradation has occurred mainly in the eastern parts of the PFB, while the central parts have regained more tree cover. These results can help identify conservation and restoration priorities and improve the overall management of the PFB.

Influence of Mango Feed-Based Diets on the Production and Economic Profitability of Montbeliarde Cows’ Milk at the End of Lactation in Burkina Faso

The study evaluated the effects of a diet incorporating a mango-based feed on the technical-economic performance of milk production of Montbeliard cows at the end of lactation in a dairy farm in Bobo-Dioulasso, Burkina Faso. To do so, 14 cows were subjected to 3 diets, including an experimental diet incorporating mango (ED) in comparison to a balanced control diet (BCD) and to the feeding practice of the farmer (FCD) during 60 days. Results show that the average amount of milk milked per day from ED (10.93 ± 0.17 L) was significantly higher (p n BCD (8.80 ± 0.19 L) and RTA (5.39 ± 0.14 L). The conversion index of FPD (4.24 ± 0.14) was significantly higher (p n that of BCD (1.75 ± 0.19) and ED (1.26 ± 0.16). Average feed costs per liter of milk (FC/L) were significantly different (p (125.95 ± 4.56 FCFA), BCD (83.67 ± 6.36 FCFA) and ED (60.06 ± 5.49 FCFA). The average gross profit margins (GPM) were significantly higher and decreasing (p ,953 ± 5031 FCFA), BCD (167,157 ± 5831 FCFA) and FCD (74,188 ± 4185 FCFA). Mango feed can be used in the diet of dairy cows. Its use contributes to the reduction of production costs and the generation of substantial profits. The valorization of mango residues is an alternative for a greater availability of feed at lower cost for the animals. It could contribute to a better management of processing residues that pollute the environment and to the reduction of fruit flies that affect orchard yields through reinfestation. The increased use of mango in dairy cattle feed should be encouraged.

Parasitic nematodes have been reported as one of the major constraints to soybean production worldwide. The majority of nematodes are so-called “free-living” and feed mainly on bacteria, fungi, protozoa and other nematodes. In Burkina Faso, the presence of parasitic nematodes has been reported in crops such as rice and sorghum. The objective of this study was to identify the genera and species of nematodes associated with soybean production in Burkina Faso. Investigations were carried out on 24 genotypes of a medium-maturity group of soybean at the Farako-Ba research station. soil samples were taken from the trial soil before its installation. At harvest, soil samples with roots were taken from each genotype in the elementary plots. The composite sample is represented by 4 lots of soil samples with roots. Extractions and counts of nematodes were performed on the different lots. In total, 7 genera of plant-parasitic nematodes associated with soybean were identified. Among these genera, Pratylenchus (100% of infected genotypes), Helicotylenchus (97.28%) and scutellonema (94.44%) were the most prevalent in terms of frequency and abundance. some soybean genotypes were less susceptible to the genus Pratylenchus is known to be highly pathogenic in soybean. These were mainly the genotypes TGX2025-10E, TGX2023-3E and TGX2025-14E.
