Transferrin receptor-targeted immunostimulant for photodynamic immunotherapy against metastatic tumors through β-catenin/CREB interruption

The immunosuppressive phenotype of tumor cells extensively attenuates the immune activation effects of traditional treatments. In this work, a transferrin receptor (TfR) targeted immunostimulant (PTI) is fabricated for photodynamic immunotherapy against metastatic tumors by interrupting beta-catenin signal pathway. To synthesize PTI, the photosensitizer conjugated TfR targeting peptide moiety (PalmiticK(PpIX)-HAIYPRH) is unitized to encapsulate the transcription interrupter of ICG-001. On the one hand, the recognition of PTI and TfR can promote drug delivery into tumor cells to destruct primary tumors through photodynamic therapy and initiate an immunogenic cell death with the release of tumor-associated antigens. On the other hand, PTI will interrupt the binding between beta-catenin and cAMP response element-binding protein (CREB), regulating the gene transcription to downregulate programmed death ligand 1 (PD-L1) while upregulating C-C motif chemokine ligand 4 (CCL4). Furthermore, the elevated CCL4 can recruit the dendritic cells to present tumor-specific antigens and promote T cells activation and infiltration, and the downregulated PD-L1 can avoid the immune evasion of tumor cells and activate systemic anti-tumor immunity to eradicate lung metastasis. This work may inspire the development of antibody antibody-free strategy to activate systemic immune response in consideration of immunosuppressive conditions. (C) 2024 The Authors. Symmetrization of quasi-regular patterns with periodic tilting of regular polygons

Computer-generated aesthetic patterns arewidely used as design materials in various fields. Themost common methods use fractals or dynamicalsystems as basic tools to create various patterns. Toenhance aesthetics and controllability, some researchershave introduced symmetric layouts along with thesetools. One popular strategy employs dynamical systemscompatible with symmetries that construct functionswith the desired symmetries. However, these aretypically confined to simple planar symmetries. Theother generates symmetrical patterns under theconstraints of tilings. Although it is slightly moreﬂexible, it is restricted to small ranges of tilingsand lacks textural variations. Thus, we proposed anew approach for generating aesthetic patterns bysymmetrizing quasi-regular patterns using general kuniformtilings. We adopted a unified strategy toconstruct invariant mappings for k-uniform tilings thatcan eliminate texture seams across the tiling ***, we constructed three types of symmetriesassociated with the patterns: dihedral, rotational, andreﬂection symmetries. The proposed method can beeasily implemented using GPU shaders and is highlyefficient and suitable for complicated tiling with regularpolygons. Experiments demonstrated the advantages of our method over state-of-the-art methods in terms ofﬂexibility in controlling the generation of patterns withvarious parameters as well as the diversity of texturesand styles.

The ECT System for RAON＇S Cavities

The ECT （eddy current test） system is in use for Nb surface quality control in many laboratories. This system can inspect Nb surthce quickly using high resolution probe. The ECT system for RAON＇s cavity was made with these features： it has a 3-axis acting probe movenlent system, it can inspect a big size of Nb sheet, which is 1,200 mm × 365 mm and it contains the analysis program that can show the result as a 2D and 3D images as well as the relative figure of the surface level. The standard sample was made with various sizes of defects using the same Nb sheet that was used to make RISP （Rare Isotope science Project） cavity. The ECT system conditioning was carried out to optimize the ECT operation on the frequency of which the range is from 300 KHZ to 2 MHz. The result of 900 KHZ showed the strongest signal. The conditioning experiment on other parameter will be carried out in the near future.

Heterogeneous Task Allocation Model and Algorithm for Intelligent Connected Vehicles

With the development of vehicles towards intelligence and connectivity, vehicular data is diversifying and growing dramatically. A task allocation model and algorithm for heterogeneous Intelligent Connected Vehicle (ICV) applications are proposed for the dispersed computing network composed of heterogeneous task vehicles and Network Computing Points (NCPs). Considering the amount of task data and the idle resources of NCPs, a computing resource scheduling model for NCPs is established. Taking the heterogeneous task execution delay threshold as a constraint, the optimization problem is described as the problem of maximizing the utilization of computing resources by NCPs. The proposed problem is proven to be NP-hard by using the method of reduction to a 0-1 knapsack problem. A many-to-many matching algorithm based on resource preferences is proposed. The algorithm first establishes the mutual preference lists based on the adaptability of the task requirements and the resources provided by NCPs. This enables the filtering out of un-schedulable NCPs in the initial stage of matching, reducing the solution space dimension. To solve the matching problem between ICVs and NCPs, a new many-to-many matching algorithm is proposed to obtain a unique and stable optimal matching result. The simulation results demonstrate that the proposed scheme can improve the resource utilization of NCPs by an average of 9.6% compared to the reference scheme, and the total performance can be improved by up to 15.9%.

Recombinant sTRAIL induces cell death of tumor cell lines as well as primary cancer cells

Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a new member of TNF family. It was reported that TRAIL could induce apoptosis of tumor cells but not normal cells in tissue culture system. To further study the biological activity and potential clinical significance, a recombinant soluble TRAIL (rsTRAIL) has been expressed stably in E. coli after transformation of pET28b vector containing the extracellular domain of TRAIL. The yield of rsTRAIL is approximately as high as 60% of whole bacterial proteins. The rsTRAIL, purified by Ni^+ -agarose affinity chromatography, could remarkably trigger apoptosis at the concentrations of 0.1—1 μg/mL in all 7 tumor cell lines tested in vitro. However, this killing activity has not been observed in mouse fibroblast cell line (NIH3T3) as normal control. Further investigation shows that the rsTRAIL could also kill primary tumor cells isolated freshly from patients with cardiac cancer, breast cancer and malignant thymoma, while the normal human

Role of brown adipose tissue in metabolic syndrome, aging, and cancer cachexia

Brown adipose tissue （BAT） plays a fundamental role in maintaining body temperature by producing heat. BAT that had been know to exist only in mammals and the human neonate has received great attention for the treatment of obesity and diabetes due to its important function in energy metabolism, ever since it is recently reported that human adults have functional BAT. In addition, beige adipocytes, brown adipocytes in white adipose tissue （WAT）, have also been shown to take part in whole body metabolism. Multiple lines of evidence demonstrated that transplantation or activation of BAT or/and beige adipocytes reversed obesity and improved insulin sensitivity. Furthermore, many genes involved in BAT activation and/or the recruitment of beige cells have been found, thereby providing new promising strategies for future clinical application of BAT activation to treat obesity and metabolic diseases. This review focuses on recent advances of BAT function in the metabolic aspect and the relationship between BAT and cancer cachexia, a pathological process accompanied with decreased body weight and increased energy expenditure in cancer patients. The underlying possible mechanisms to reduce BAT mass and its activity in the elderly are also discussed.
