The life-history trait trade-offs mediated by reproduction and immunity in the brown planthopper,Nilaparvata lugens Stål

Reproduction and immune defense are costly functions,and they are expected to tradeoff with each other to drive evolution.The brown planthopper(BPH),Nilaparvata lugens Stål(Hemiptera,Delphacidae),is a global superpest that mostly damages rice crops.Yeast-like symbionts(YLS)exist in the abdominal fat body tissue and are tightly associated with the development,growth,and reproduction of BPH.Our previous research demonstrated that mating behavior promotes the release of YLS from the fat body into the hemolymph in the BPH,thereby triggering an immune response.Additionally,the fitness costs related to life-history traits of BPH(such as survival rate)have a strong dependence on the relative abundance of YLS.However,the possible relationship between reproduction and the immune response in BPH has not been identified.In this study,an omics-based approach was used to analyze the transcriptome of fat body tissues in mated and unmated BPH at 72 h post-eclosion,from which two antimicrobial peptide genes,NlDefensin A(NlDfA)and NlDefensin B(NlDfB),were selected since they were highly expressed in mated BPH.Subsequently,the full-length cDNA sequences of the NlDfA and NlDfB genes were cloned and analyzed.qPCR results showed up-regulation of the NlDfA and NlDfB genes in mated BPH when compared to unmated BPH.Spatial-temporal expression analysis indicated that the NlDfA and NlDfB genes were expressed in all tissues and developmental stages,and they were most highly expressed in the fat body at 24 h post-eclosion.Moreover,the symbionts in BPH were significantly inhibited by the in vitro expression of the NlDfA and NlDfB proteins.Furthermore,RNA interference(RNAi)-mediated suppression of NlDfA and NlDfB dramatically increased the relative abundance of YLS in the fat body,while YLS in the hemolymph decreased significantly.These BPHs also displayed some fitness disadvantages in survival,fecundity,hatchability,and possibly the vertical transmission of YLS from hemolymph to egg.Our results indicated that mating could heighten the immuni

Genetic variability of half-smooth tongue sole Cynoglossus semilaevis populations using microsatellite markers

Ten highly variable microsatellite loci were performed in order to evaluate the genetic variability of wild and hatchery samples of half-smooth tongue sole Cynoglossu＇s semilaevis. A group of 200 genotypes belonging to four wild samples, Laizhou （LZ）, Weihai （WH）, Qingdao （QD）, Rizhao （RZ） and one hatchery sample, Mingbo （MB）, were screened. All of the： ten microsatellite loci screened in this study showed marked polymorphism. A total of 70 different alleles were observed over all loci. The number of alleles per locus ranged from 4.2 to 1006 the average of expected and observed heterozygosity ranged from 0.67 to 0.82, and from 0：77 to0.87 respectively. A total of 10, 16, 10, 11 and 5 unique alleles each were found in LZ, WH~ QD：RZ and MB populations. The effective number of alleles varied from 2.87 for HSTS_7 to 6.83 for HSTS_h. The number of average genotypes ranged from 6.0 for HSTS a to 15.4 for HSTS_h： As compared with the wild populations, the hatchery population, MB, showed significant genetic changes such as fewer alleles per locus （P〈0.05）, a smaller number of low frequency alleles （P〈0.05）, a small number of unique alleles and a small number of genotypes （P〈0.05）, all indicative of a reduction in genetic diversity.

Comparative study of the effects of Tartary buckwheat seed and sprout consumption on the physiological indices and gut microbiota of C57BL/6J mice

Tartary buckwheat(Fagopyrum tataricum)is a well-known pseudocereal for its health and economic value.However,abundant antinutritional factors(ANFs)reduces its health benefits.As reported,germination can improve the nutritional profile of grains.In this study,we systematically evaluate the safety of Tartary buckwheat seeds(TB)and Tartary buckwheat sprouts(TBS)used as high active ingredients.After evaluating nutrition levels,bioactive compounds and ANFs in TBS during germinating,5^(th)-day TBS were selected as the raw material.C57BL/6J mice were gavaged daily with distilled water,TB,or TBS for 6 weeks.The physiological indices related to ANFs were determined.Results showed that the TB intake tends to generate negative effects on the gut microbiota,and organs.Additionally,upon TB intake,the Fe^(3+)content in serum,trypsin activity in pancreas and jejunum decreased,while the cytokine,IgE,and histamine levels in serum,water content in faeces,cytokine levels in liver and jejunum increased.Conversely,TBS did not induce any obvious negative effects on the above relevant indices and showed better lipid-lowering effect.Altogether,TBS are safer and more effective as a raw material to produce the functional food for long-term consumption with the intention of preventing and treating hyperlipidaemia.

A novel low molecular weight compound polysaccharides from Chinese herbal medicines: purification， characterization and anti-tumor activities in vitro and in vivo

A novel low molecular weight polysaccharide (LMW-CPS) was identified from a specific combination of Chinese herb ingredients. The monosaccharide composition of LMW-CPS was consisted of single arabinose， which had an α-L-furanose configuration with an average molecular weight of 2.06 kDa. NMR spectra and monosaccharide constitution analyses revealed that it had a backbone of →5)-α-L-Araf-(1→ with α-L-Araf-(1→ as the terminal residue. In vitro experiments found that it could lead to apoptosis and inhibit hepatocellular carcinoma cell proliferation by arresting them in the S phase. In vivo experiments showed that it protected immune organs such as the thymus and spleen， enhanced immune cell activities， stimulated cytokine release， augmented the abundance of CD8， CD3， CD4， and CD19 positive lymphocytes， and markedly impeded solid hepatocellular carcinoma progression in mice. H&E staining and cell cycle examination also indicated that LMW-CPS arrested hepatocellular carcinoma cells at the S phase to induce apoptosis. These findings indicated its promising potential for the treatment of hepatocellular carcinoma.

A FAS approach for stabilization of generalized chained forms: part 1. Discontinuous control laws

In this paper, a type of general nonholonomic systems is proposed, which contains both the Brockett’s two example systems, and their extended n-dimensional chained forms, as special cases. For the stabilization of such systems, a stabilizing controller is proposed based on the fully actuated system(FAS)approach, which is discontinuous at the origin but time-invariant when the open-loop system is time-invariant,and drives the feasible trajectories of the system to the origin exponentially. Furthermore, the proposed FAS approach is also extended to the sub-normal system case and the time-delay system case.

Noncovalent cross-linked engineering hydrogel with low hysteresis and high sensitivity for flexible self-powered electronics

In this study,the hydrogel network was reinforced by covalent-like hydrogen bonding,and the strong binding ability of boron-nitrogen coordination served as the main driving force.Among them,acrylamide(AM)and 3-acrylamidophenylboronic acid(AAPBA)were the main body,and the numerous hydroxyl groups in the trehalose(Treh)molecule and other polymer groups formed strong hydrogen bonding interactions to improve the mechanical properties of the PAM/PAAPBA/Treh(PAAT)hydrogel and ensured the simplicity of the synthesis process.The hydrogel possessed high strain at break(1239%),stress(64.7 kPa),low hysteresis(100%to 500%strain,corresponding to dissipation energy from 1.37 to 7.80 kJ/m^(3)),and outstanding cycling stability(retained more than 90%of maximum stress after 200 ten-sile cycles).By integrating carbon nanotubes(CNTs)into PAAT hydrogel(PAATC),the PAATC hydrogel with excellent strain response performance was successfully constructed.The PAATC conductive hydro-gel exhibited high sensitivity(gauge factor(GF)=10.58 and sensitivity(S)=0.304 kPa^(-1)),wide strain response range(0.5%-1000%),fast response time(450 ms),and short recovery time(350 ms),excellent fatigue resistance,and strain response stability.Furthermore,the PAATC-based triboelectric nanogener-ator(TENG)displayed outstanding energy harvesting performance,which shows its potential for appli-cation in self-powered electronic devices.

Interaction Between Ustilaginoidea virens and Rice and Its Sustainable Control

Ustilaginoidea virens is a common rice pathogen that can easily lead to a decline in rice quality and the production of toxins pose potential risks to human health.In this review,we present a comprehensive literature review of research since the discovery of rice false smut.We provide a comprehensive and,at times,critical overview of the main results and findings from related research,and propose future research directions.Firstly,we delve into the interaction between U.virens and rice,including the regulation of transcription factors,the process of U.virens infecting rice panicles,and the plant immune response caused by rice infection.Following that,we discuss the identification and characterization of mycotoxins produced by the pathogenic fungus,as well as strategies for disease management.We emphasize the importance of comprehensive agricultural prevention and control methods for the sustainable management of U.virens.This knowledge will update our understanding of the interaction between U.virens and rice plants,offering a valuable perspective for those interested in U.virens.

Prescribed-time leader-following consensus of linear multi-agent systems by bounded linear time-varying protocols

This paper considers the prescribed-time leader-following consensus problem of input-constrained linear multi-agent systems under generally directed communication topology in two cases:the Laplacian matrix related to the entire communication topology between agents is either known or unknown.In particular,the consensus problem for the former case is solved by a novel bounded linear time-varying(LTV) protocol,where the feedback gain is formulated by the parametric Lyapunov equation and the knowledge of the Laplacian matrix.Moreover,by utilizing a distributed observer,a fully bounded LTV protocol is proposed for the latter case.It should be noted that,compared with the existing results,the system under consideration is more general,the designed protocols are linear,and the consensus problem is accomplished even in a fully distributed manner.Finally,the effectiveness of the proposed approach is verified by a numerical example.

Prescribed time control based on the periodic delayed sliding mode surface without singularities

In this paper, nonsingular prescribed-time control is studied based on periodic delayed sliding mode surfaces. Different from the existing sliding mode control, where either singularity problems may appear or the convergence time depends on the initial state, the proposed sliding mode control approaches can achieve prescribed-time convergence without singularity. The proposed nonsingular sliding mode control approaches can be applied to both second-order and high-order nonlinear systems with prescribed-time convergence. As applications of the proposed sliding mode control approaches, the control of hypersonic vehicle systems is revisited. Numerical simulations on the nonlinear model of the hypersonic vehicle system show the effectiveness of the proposed methods.

Reduction of the oxidative damage to H_(2)O_(2)-induced HepG2 cells via the Nrf2 signalling pathway by plant flavonoids Quercetin and Hyperoside

Hyperoside and quercetin are similar in molecular structures.In this study,the antioxidant regulatory targets of hyperoside and quercetin are mainly in the nuclear factor(erythroid-2-derived)-related factor 2(Nrf2)pathway predicted by network pharmacology.And the antioxidant effect and mechanism of hyperoside and quercetin were measured and compared in H_(2)O_(2)-induced Hep G2 cells and Caenorhabditis elegans.The findings indicated that quercetin was more effective than hyperoside in reducing oxidative damage,which was proved by improved cell viability,decreased reactive oxygen species(ROS)production,decreased cellular apoptosis,and alleviated mitochondrial damage.In addition,quercetin was more efficient than hyperoside in enhancing the expression of Nrf2-associated m RNAs,increasing the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and catalase(CAT),and reducing the cellular malondialdehyde(MDA)content.Quercetin was superior to hyperoside in prolonging the lifespan of worms,decreasing the accumulation of lipofuscin,inhibiting ROS production,and increasing the proportion of skn-1 in the nucleus.With the Nrf2 inhibitor ML385,we verified that quercetin and hyperoside primarily protected the cells against oxidative damage via the Nrf2 signalling pathway.Furthermore,molecular docking and dynamics simulations demonstrated that the quercetin-Kelch-like ECH-associated protein 1(Keap1)complex was more stable than the hyperoside-Keap1 complex.The stable structure of the complex might hinder the binding of Nrf2 and Keap1 to release Nrf2 and facilitate its entry into the nucleus to play an antioxidant role.Overall,quercetin had a better antioxidant than hyperoside.
