A novel live attenuated vaccine candidate protects chickens against subtype B avian metapneumovirus

Avian metapneumovirus(aMPV) is a highly contagious pathogen that causes acute upper respiratory tract diseases in chickens and turkeys, resulting in serious economic losses. Subtype B aMPV has recently become the dominant epidemic strain in China. We developed an attenuated aMPV subtype B strain by serial passaging in Vero cells and evaluated its safety and efficacy as a vaccine candidate. The safety test showed that after the 30th passage, the LN16-A strain was fully attenuated, as clinical signs of infection and histological lesions were absent after *** LN16-A strain did not revert to a virulent strain after five serial passages in chickens. The genomic sequence of LN16-A differed from that of the parent wild-type LN16(wtLN16) strain and had nine amino acid mutations. In chickens, a single immunization with LN16-A induced robust humoral and cellular immune responses, including the abundant production of neutralizing antibodies, CD4^(+) T lymphocytes, and the Th1(IFN-γ) and Th2(IL-4 and IL-6)cytokines. We also confirmed that LN16-A provided 100% protection against subtype B aMPV and significantly reduced viral shedding and turbinate inflammation. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype B aMPV.

Introduction:Coxsackievirus A6(CVA6)has emerged as a significant pathogen responsible for severe cases of hand,foot,and mouth disease(HFMD).This study aims to delineate the demographic characteristics and analyze the viral evolution of severe HFMD associated with CVA6,thereby assisting in its surveillance and ***:In this investigation,74 strains of CVA6 were isolated from samples collected from severe HFMD cases between 2012 and *** VP1 gene sequences of CVA6 were amplified and analyzed to assess population historical dynamics and evolutionary characteristics using BEAST,DnaSP6,and ***:A significant portion(94.4%)of severe CVA6-associated HFMD cases(51 out of 54,with 20 lacking age information)were children under 5 years *** the 74 CVA6 strains analyzed,72 belonged to the D3a sub-genotype,while only two strains were D2 *** average genetic distance between VP1 sequences prior to 2015 was 0.027,which increased to 0.051 when compared to sequences *** population dynamics analysis indicated three significant population expansions of severe CVA6-associated HFMD during 2012-2013,2013-2014,and 2019-2020,resulting in the formation of 65 distinct *** with the MCC tree findings,transitioning between regional haplotypes required multiple base substitutions,showcasing an increase in population diversity during the evolutionary process(from 14 haplotypes in 2013 to 55 haplotypes over the subsequent decade).Conclusions:CVA6,associated with severe HFMD,is evolving and presents a risk of outbreak ***,enhanced surveillance of severe HFMD is imperative.

KIF5B-mediated internalization of FMDV promotes virus infection

Pasteurella multocida (PM) has been infecting a variety of hosts for a long time,causing sustained economic losses worldwide;however,there have been limited studies on its extensive adaptability (Aktories et al.2012).Analysis of strains data collected in our laboratory revealed that PM typically acquires foreign genes through transformation and conjugation,rather than transformation and *** and conjugative elements (ICEs)are a crucial mechanism that leads to abrupt changes in niche preferences and enhances environmental adaptability for bacteria,with their number far exceeding the number of plasmids and phages (Wozniak and Waldor 2010;Johnson and Grossman 2015;Botelho and Schulenburg 2021).Previous research has shown that the European bovine-sourced strain Pm36950 contains the experimentally transferable resistant ICEPmu1,which could integrate into the host chromosome or forming a circular intermediate (Michael et al.2012).Additionally,a few studies have suggested that PM strains isolated from European bovine and Asian swine might contain ICEs,but these claims lack experimental verification (Klima et al.2014;Moustafa et al.2015;Kadlec et al.2017;Peng et al.2017;Beker et al.2018;Schink et al.2022).Currently,there is no data available on ICE-carrying PM strains isolated from hosts outside of European cattle or Chinses *** letter presents a report on a novel ICE identified in the hypervirulent and multidrug-resistant PM HN141014 strain isolated from Chinese *** ICE was specifically analyzed for its resistance genes,transferable capacity and host diversity.

Activation of skeletal carbohydrate-response element binding protein(ChREBP)-mediated de novo lipogenesis increases intramuscular fat content in chickens

The intracellular lipids in muscle cells of farm animals play a crucial role in determining the overall intramuscular fat(IMF)content,which has a positive impact on meat ***,the mechanisms underlying the deposition of lipids in muscle cells of farm animals are not yet fully *** purpose of this study was to determine the roles of carbohydrate-response element binding protein(ChREBP)and fructose in IMF deposition of *** virus-mediated ChREBP overexpression in tibialis anterior(TA)muscle of chickens,seven 5-d-old male yellow-feather chickens were *** 10 d after virus injection,the chickens were slaughtered to obtain TA muscles for *** fructose administration trial,sixty 9-wk-old male yellow-feather chickens were randomly divided into 2 groups,with 6 replicates per group and 5 chickens per *** chickens were fed either a basal diet or a basal diet supplemented with 10%fructose(purity≥99%).At 4 wk later,the chickens were slaughtered,and breast and thigh muscles were collected for *** results showed that the skeletal ChREBP mRNA levels were positively associated with IMF content in multiple species,including the chickens,pigs,and mice(P<0.05).ChREBP overexpression increased lipid accumulation in both muscle cells in vitro and the TA muscles of mice and chickens in vivo(P<0.05),by activation of the de novo lipogenesis(DNL)***,activation of ChREBP by dietary fructose administration also resulted in increased IMF content in mice and notably chickens(P<0.05).Furthermore,the lipidomics analysis revealed that ChREBP activation altered the lipid composition of chicken IMF and tented to improve the flavor profile of the *** conclusion,this study found that ChREBP plays a pivotal role in mediating the deposition of fat in chicken muscles in response to fructose-rich diets,which provides a novel strategy for improving meat quality in the livestock industry.
