BACKGROUND Endoscopic ultrasound(EUS)-guided transluminal drainage is an advanced technique used to treat pancreatic fluid collections(PFCs). However, gastric varices and intervening vessels may be associated with a high risk of bleeding and are, therefore, listed as relative contraindications. Herein, we report two patients who underwent interventional embolization before EUS-guided *** SUMMARY Two 32-year-old males developed symptomatic PFCs after acute pancreatitis and came to our hospital for further treatment. One patient suffered from intermittent abdominal pain and vomiting, and computed tomography(CT) imaging showed an encapsulated cyst 7.93 cm × 6.13 cm in size. The other patient complained of a mass inside the abdomen, which gradually became enlarged. Gastric varices around the ideal puncture site were detected by EUS when we evaluated the possibility of endoscopic drainage in both patients. Interventional embolization was recommended as the first procedure to decrease the risk of bleeding. After that, EUS-guided transluminal drainage was successfully conducted, without vascular rupture. No postoperative complications occurred during hospitalization, and no recurrence was detected at the last follow-up CT scan performed at 1 *** Interventional embolization is a safe, preoperative procedure that is performed before EUS-guided drainage in PFC patients with gastric varices or at high risk of bleeding.

Cachexia,which affects 50–80%of cancer patients,is a debilitating syndrome that leads to 20%of cancer-related deaths.A key feature of cachexia is adipose tissue atrophy,but how it contributes to the development of cachexia is poorly ***,we demonstrate in mouse models of cancer cachexia that white adipose tissue browning,which can be a characteristic early-onset manifestation,occurs prior to the loss of body weight and skeletal muscle *** analysing the proteins differentially expressed in extracellular vesicles derived from cachexia-inducing tumours,we identified a molecular chaperone,Glucose-regulated protein 75(GRP75),as a critical mediator of adipocyte ***,GRP75 binds adenine nucleotide translocase 2(ANT2)to form a GRP75–ANT2 ***,stabilized ANT2 enhances its interaction with uncoupling protein 1,leading to elevated expression of the latter,which,in turn,promotes adipocyte *** with withanone,a GRP75 inhibitor,can reverse this browning and alleviate cachectic phenotypes in ***,our findings reveal a novel mechanism by which tumour-derived GRP75 regulates white adipose tissue browning during cachexia development and suggest a potential white adipose tissue-centred targeting approach for early cachexia intervention.

Objective N6-methyladenosine（m6A） is a common epigenetic modification in eukaryotes. In this study, we explore the potential impact of m6A-associated single nucleotide polymorphisms（m6A-SNPs）on heart failure（HF）.Methods Data from genome-wide association studies（GWAS） investigating HF in humans and from m6A-SNPs datasets were used to identify HF-associated m6A-SNPs. Their functions were explored using expression quantitative trait locus（e QTL）, gene expression, and gene enrichment analyses. Mediation protein quantitative trait locus（p QTL）-Mendelian randomization（MR） was used to investigate the potential mechanism between critical protein levels and risk factors for *** We screened 44 HF-associated m6A-SNPs, including 10 m6A-SNPs that showed e QTL signals and differential expressions in HF. The SNP rs1801270 in CDKN1A showed the strongest association with HF(P = 7.75 × 10-6). Additionally, MR verified the genetic association between the CDKN1A protein and HF, as well as the mediating effect of blood pressure（BP） in this pathway. Higher circulating level of CDKN1A was associated with a lower risk of HF（odds ratio [OR] = 0.82, 95% confidence interval [CI]: 0.69to 0.99）. The proportions of hypertension, systolic BP, and diastolic BP were 48.10%, 28.94%, and18.02%, respectively. Primary peritoneal carcinoma(PPC) is a type of rare malignant epithelial tumor. Metastasis from PPC to breast has been rarely reported. PPC originates de novo from the peritoneal tissues rather than invasion or metastasis from adjacent or remote *** have been implicated in many cases of carcinomas of unknown primary origin. It is similar to ovarian cancer(Ov Ca),because it shares the same common embryonic origin, the coelomic epithelium(mesodermal origin). The mechanism of oncogenesis remains elusive. In this article, we report a rare case of PPC in a patient 10 years after total abdominal hysterectomy and bilateral salpingooophorectomy for uterine leiomyoma, which was widely spread in the abdomen and metastasized to the colon, liver and distant organs including breast. The treatment is similar to that of primary ovarian cancer. We also reviewed the primary peritoneal cancer metastatic to breast and discuss the possible mechanisms and biology of primary peritoneal cancer,using experimental and animal model.

Objective:To observe the changes of symptoms,Chinese medicine(CM)syndrome,and lung inflammation absorption during convalescence in patients with coronavirus disease 2019(COVID-19)who had not totally recovered after hospital discharge and whether CM could promote the improvement ***:This study was designed as a prospective cohort and nested case-control study.A total of 96 eligible patients with COVID-19 in convalescence were enrolled from Beijing Youan Hospital and Beijing Huimin Hospital and followed up from the hospital discharged *** were divided into the CM(64 cases)and the control groups(32 cases)based on the treatment with or without CM and followed up at 14,28,56,and 84 days after *** the CM group,patients received the 28-day CM treatment according to two types of CM *** in clinical symptoms,CM syndrome,and absorption of lung inflammation were ***:All the 96 patients completed the 84-day follow-up from January 21 to March 28,*** the 84th day of follow-up,respiratory symptoms were less than 5%.There was no significant difference in the improvement rates of symptoms,including fatigue,sputum,cough,dry throat,thirst,and upset,between the two groups(P>0.05).Totally 82 patients(85.42%)showed complete lung inflammation absorption at the 84-day *** day 14,the CM group had a significantly higher absorption rate than the control group(P<0.05)and the relative risk of absorption for CM *** group was 3.029(95%confidence interval:1.026-8.940).The proportions of CM syndrome types changed with time prolonging:the proportion of the pathogen residue syndrome gradually decreased,and the proportion of both qi and yin deficiency syndrome gradually ***:Patients with COVID-19 in convalescence had symptoms and lung inflammation after hospital discharge and recovered with time *** could improve lung inflammation for early *** types of CM syndrome can be transformed wi

Adult tendon stem/progenitor cells(TSPCs)are essential for tendon maintenance,regeneration,and repair,yet they become susceptible to senescence with age,impairing the self-healing capacity of *** this study,we employ a recently developed deep-learning-based efficacy prediction system to screen potential stemness-promoting and senescence-inhibiting drugs from natural products using the transcriptional signatures of *** top-ranked candidate,prim-O-glucosylcimifugin(POG),a saposhnikovia root extract,could ameliorate TPSC senescent phenotypes caused by long-term passage and natural aging in rats and humans,as well as restore the self-renewal and proliferative capacities and tenogenic potential of aged *** vivo,the systematic administration of POG or the local delivery of POG nanoparticles functionally rescued endogenous tendon regeneration and repair in aged rats to levels similar to those of normal ***,POG protects TSPCs against functional impairment during both passage-induced and natural aging by simultaneously suppressing nuclear factor-κB and decreasing mTOR signaling with the induction of ***,the strategy of pharmacological intervention with the deep learning-predicted compound POG could rejuvenate aged TSPCs and improve the regenerative capacity of aged tendons.
