Effects of long-term straw incorporation on nematode community composition and metabolic footprint in a rice–wheat cropping system

Soil nematode communities can provide valuable information about the structure and functions of soil food webs,and are sensitive to agricultural practices,including short-term straw ***,currently,such effects under longterm straw incorporation conditions at different fertility levels are largely ***,we conducted a 13-year ongoing experiment to evaluate the effects of long-term straw incorporation on the structure and functions of the soil food web in low and high fertility soils through analyzing its effects on nematode communities,food web indices and metabolic *** treatments were included:straw removal(–S)under non-fertilized(–NPK)or fertilized(+NPK)conditions;and straw incorporation(+S)under–NPK or+NPK *** samples from a 0–20 cm depth layer were collected when wheat and rice were *** with straw removal,straw incorporation increased the abundances of total nematodes,bacterivores,plant-parasites and omnivores-predators,as well the relative abundances of omnivores-predators with increases of 73.06,89.29,95.31,238.98,and 114.61%in–NPK soils and 16.23,2.23,19.01,141.38,and 90.23%in+NPK soils,*** of sampling times and fertilization effects,straw incorporation increased the diversity and community stability of nematodes,as indicated by the Shannon-Weaver diversity index and maturity *** and structure index did not show significant responses to straw incorporation,but a slight increase was observed in the structure *** analysis of nematode metabolic footprints showed that straw incorporation increased the plant-parasite footprint and structure footprint by 97.27 and 305.39%in–NPK soils and by 11.29 and 149.56%in+NPK soils,but did not significantly influence enrichment,bacterivore and fungivore *** conclusion,long-term straw incorporation,particularly under a low fertility level,favored the soil nematodes and regulated the soil food web mainly via a top-down

Sperm cryopreservation is the best modality to ensure future fertility for males diagnosed with cancer. The extent to which cryopreserved sperm is actually used for impregnation, the fertility treatment options that are available and the success rates of these treatments have not been investigated in depth. The medical records of 682 patients who cryopreserved sperm cells due to cancer treatment were analyzed. Seventy of these patients withdrew their frozen sperm for fertility treatments over a 20-year period （most within the first 4 years after cryopreservation）. Sperm quality of different malignancies and outcomes of assisted reproduction treatment （ART） for pregnancy achievement in relation to the type of treatment and the type of malignancy were evaluated. The results showed that the rate of using cryo-thawed sperm from cancer patients for fertility treatments in our unit was 10.3%. Sperm quality indices differed between different types of malignancies, with the poorest quality measured in testicular cancer. Conception was achieved in 46 of the 184 ART cycles （25%）, and resulted in 36 deliveries. The use of intracytoplasmic sperm injection （ICSI） methodology yielded a significantly higher pregnancy rate （37.4%） than intrauterine insemination （IUI; 11.5%） and was similar to other groups of infertile couples using these modalities. In vitro fertilization （IVF） failed to produce pregnancies. In conclusion, the rate of use of cryopresseved sperm in cancer patients is relatively low （10.3%）. Achievement of pregnancies by ICSI presents the best option but when there are enough stored sperm samples and adequate quality, I UI can be employed. Cryopreservation is nevertheless the best option to preserve future fertility potential and hope for cancer patients.

For men with severe oligozoospermia, sperm cryopreservation can preserve surgically obtained sperm. How to cryopreserve single sperm in men is still a hot topic in assisted reproduction technology. Aim to analyze the laboratory and pregnancy outcomes of single sperm cryopreservation group, we retrospectively selected 38 cycles underwent single sperm cryopreservation and thawing as the study group and 618 cycles underwent conventional sperm cryopreservation and thawing as the control group, which were performed in the reproductive medicine center of the Sixth Affiliated Hospital, Sun Yatsen University, from April 2014 to October 2023. All the sperm came from microdissection testicular sperm extraction (micro-TESE), and performed intracytoplasmic sperm injection (ICSI) for fertilization. Zygotes were cultured to Day 3 embryo, which were freshly transferred to female uterus. Surplus embryos were cultured to blastosphere and cryopreserved. There was no statistical difference in female/male age, female BMI, infertility duration and female basal sex hormone (FSH, LH E2, AMH), No. of oocytes retrieved per cycle, No. of ICSI oocytes per cycle and No. of embryos transferred per cycle between the two groups (P > 0.05). No significant difference was found in two-pronuclear oocyte fertilization rate (59.23% VS 58.84%), Day 3 available embryo rate (61.81% VS 63.55%), Day 3 good-quality embryo rate (45.73% VS 50.27%), blastocyst formation rate (47.83% VS 49.46%), the implantation rate (47.37% VS 52.16%), clinical pregnancy rate (36.84% VS 47.18%), miscarriage rate (14.29% VS 12.68%) and live birth rate (85.71% VS 81.70%) between two groups (P > 0.05). In conclusion, single-sperm cryopreservation was the optimal method to preserve sperm after micro-TESE. It can increase the utilization of each sperm and lead to clinical pregnancy.

Long-term excessive application of mineral fertilizer has led to soil acidification and phosphorus(P) accumulation, increasing the risk of P loss and environmental pollution, and cessation of fertilization is widely considered as a cost-effective management strategy to relieve this situation;however, how such cessation influences P speciation and concentrations in a bulk soil and colloidal fractions and whether decreasing P concentration might maintain soil fertility remain unclear. In this study, the effects of long-term fertilization(ca. 40 years) and short-term cessation of fertilization(ca. 16 months) on inorganic, organic,and colloidal P in lime concretion black soil were investigated using P sequential fractionation and31P nuclear magnetic resonance spectroscopy. After long-term fertilization, available P, dicalcium phosphate, iron-bound P, orthophosphate monoesters, and orthophosphate diesters increased significantly, but soil p H decreased by ca. 2.8 units, indicating that long-term fertilization caused soil acidification and P accumulation and changed P speciation markedly. In contrast, short-term fertilization cessation increased soil p H by ca. 0.8 units and slightly reduced available and inorganic P. Available P after fertilization cessation was 22.9–29.8 mg kg-1, which was still sufficient to satisfy crop growth requirements. Additionally, fertilization cessation increased the proportions of fine colloids(100–450 nm, including nontronite and some amorphous iron oxides) and drove a significant release of iron/aluminum oxide nanoparticles(1–100 nm) and associated P with orthophosphate and pyrophosphate species. In summary, short-term fertilization cessation effectively alleviated soil acidification and inorganic P accumulation, while concomitantly maintaining soil P fertility and improving the potential mobilization of P associated with microparticles.

Importin proteins were originally characterized for their central role in protein transport through the nuclear pores, the only intracellular entry to the nucleus. This vital function must be tightly regulated to control access by transcription factors and other nuclear proteins to genomic DNA, to achieve appropriate modulation of cellular behaviors affecting cell fate. Importin-mediated nucleocytoplasmic transport relies on their specific recognition of cargoes, with each importin binding to distinct and overlapping protein subsets. Knowledge of importin function has expanded substantially in regard to three key developmental systems： embryonic stem cells, muscle cells and the germ line. In the decade since the potential for regulated nucleocytoplasmic transport to contribute to spermatogenesis was proposed, we and others have shown that the importins that ferry transcription factors into the nucleus perform additional roles, which control cell fate. This review presents key findings from studies of mammalian spermatogenesis that reveal potential new pathways by which male fertility and infertility arise. These studies of germline genesis illuminate new ways in which importin proteins govern cellular differentiation, includ ng v a d rect ng proteins to d st nct ntrace ular compartments and by determining cellular stress responses.
