LncRNA HOTAIR promotes DNA damage repair and radioresistance by targeting ATR in colorectal cancer

Long non-coding RNAs(lncRNAs)have been implicated in cancer progression and drug resistance ***,there is evidence that lncRNA HOX transcript antisense intergenic RNA(HOTAIR)is involved in colorectal cancer(CRC)*** present study aimed to examine the functional role of lncRNA HOTAIR in conferring radiotherapy resistance in CRC cells,as well as the underlying *** relative expression levels of HOTAIR were examined in 70 pairs of CRC tumor and para-cancerous tissues,as well as in radiosensitive and radioresistant *** correlations between HOTAIR expression levels and clinical features of patients with CRC were assessed using the Chi-square *** assays such as cell proliferation,colony formation and apoptosis assays were conducted to determine the radiosensitivity in CRC cells with HOTAIR silencing after treatment with different doses of *** pull-down assay andﬂuorescence in situ hybridization(FISH)were used to determine the interaction between HOTAIR and DNA damage response mediator ataxia-telangiectasia mutated-and Rad3-related(ATR).HOTAIR was signiﬁcantly upregulated in CRC tumor tissues,especially in radioresistant tumor *** elevated expression of HOTAIR was correlated with more advanced histological grades,distance metastasis and the poor prognosis in patients with *** HOTAIR suppressed the proliferation and promoted apoptosis and radiosensitivity in CRC *** knockdown also inhibited the tumorigenesis of CRC cells and enhanced the sensitivity to radiotherapy in a mouse xenograft ***,the data showed that HOTAIR could interact with ATR to regulate the DNA damage repair signaling *** HOTAIR impaired the ATR-ATR interacting protein(ATRIP)complex and signaling in cell cycle ***,the present results indicate that lncRNA HOTAIR facilitates the DNA damage response pathway and promotes radioresistance in CRC cells by targeting ATR.

Lactobacillus delbrueckii subsp. bulgaricus(L. bulgaricus) and Streptococcus thermophilus(S. thermophilus) are commonly used starters in milk fermentation. Fermentation experiments revealed that L. bulgaricus-S. thermophilus interactions(LbSt I) substantially impact dairy product quality and production. Traditional biological humidity experiments are time-consuming and labor-intensive in screening interaction combinations, an artificial intelligence-based method for screening interactive starter combinations is necessary. However,in the current research on artificial intelligence based interaction prediction in the field of bioinformatics, most successful models adopt supervised learning methods, and there is a lack of research on interaction prediction with only a small number of labeled ***, this study aimed to develop a semi-supervised learning framework for predicting LbSt I using genomic data from 362 isolates(181per species). The framework consisted of a two-part model: a co-clustering prediction model(based on the Kyoto Encyclopedia of Genes and Genomes(KEGG) dataset) and a Laplacian regularized least squares prediction model(based on K-mer analysis and gene composition of all isolates datasets). To enhance accuracy, we integrated the separate outcomes produced by each component of the two-part model to generate the ultimate LbSt I prediction results, which were verified through milk fermentation experiments. Validation through milk fermentation experiments confirmed a high precision rate of 85%(17/20; validated with 20 randomly selected combinations of expected interacting isolates). Our data suggest that the biosynthetic pathways of cysteine, riboflavin, teichoic acid, and exopolysaccharides, as well as the ATP-binding cassette transport systems, contribute to the mutualistic relationship between these starter bacteria during milk fermentation. [Objectives]To investigate the synergistic effect of Arnebiae Radix after processing.[Methods]The effects of raw Arnebiae Radix and milk processed Arnebiae Radix on hypothermia in yeast-induced febrile rats were compared.[Results]The processed and unprocessed Arnebiae Radix at high,medium and low doses all had a certain effect on inhibiting the rise of body temperature in *** high dose unprocessed group,the medium dose processed group and the high dose processed group had the best inhibitory effect on body temperature,the low dose processed group could delay the fever time,and the low dose unprocessed group had poor inhibitory effect on fever.[Conclusions]The prepared Arnebiae Radix has enhanced drug effect,and milk processed Arnebiae Radix can be used to replace common Arnebiae Radix to reduce the dosage of Arnebiae Radix and save Arnebiae Radix resources.

OBJECTIVE To investigate the effects and mechanism of a new chemical synthetic drug TPh on the proliferation and apoptosis of Hela cel s. METHODS MTT assay was performed to detect the effect of TPh on the proliferation of He La cel s at different times. The impact of each dose on the cycle of Hela cells was analyzed by flow cytometry; Annexin-Ⅴ FITC/PI staining was used to detect the effect of TPh on the apoptosis of He La cells;JC-1 staining was made to observe the effect of TPh on the mitochondrial membrane potential of Hela cell. The levels of apoptosis-related protein caspase-3, caspase-8,caspase-9 were evaluated in TPh-treated cel s by Western blotting. RESULTS TPh could inhibit the proliferation of Hela cells within a certain concentration range, and the effect is proportional to time and concentration. Comparing with the blank control group, the proportion of cells in the G0/G1 phase gradually increased and the proportion of cells in the G2/M phase decreased in all of the groups with TPh. Besides, apoptotic He La cells increased in a dose-dependent manner after treated with different concentrations of TPh, mitochondrial membrane potential reduced in different degrees, the protein expression of caspase-3,caspase-8 and caspase-9 were increased in a dose-dependent manner under TPh treatment. CONCLUSION TPh could inhibit the proliferation of He La cel s in a dose-dependent manner; it blocks He La cells in the G0/G1 phase and inhibits the conversion of Hela cells in the G0/G1 phase to S and G2/M phase. Besides, it could induces apoptosis of He La cells by reducing the mitochondrial membrane potential and promoting the expression of caspase-3, caspase-8, caspase-9.

Promotion of reactive oxygen species activated by nanosilver surface engineering for resistant bacteria-infected skin tissue therapy

Nanosilver has been regarded as a promising alternative to traditional antibiotics for fighting pathogenassociated infections due to its efficacy toward a broad spectrum of ***,bacterial resistance to nanosilver has emerged *** this contribution,a surface engineering strategy based on N-halamine chemistry to address bacterial resistance to nanosilver was *** 1,3-dichloro-5,5-dimethylhydantoin(DCDMH)as an N-halamine source,AgCI nanodots were deposited on the surface of Ag nano wires(Ag NWs)via in situ redox reaction to prepare AgCl-on-Ag *** in vitro and in vivo tests,AgCl-on-Ag NWs effectively inactivated two antibiotic-resistant bacteria,ampicillinresistant Escherichia coli(AREC)and methicillin-resistant Staphylococcus aureus(MRSA)with the minimum bactericidal concentration(MBC)as low as 10μg·ml~(-1)and exhibited good biosafety against normal *** experimental and theoretical tests demonstrated that AgCl-onAg NWs worked on AREC and MAS A by generating high level of reactive oxygen species under visible light irradiation,coupled with the sustained Ag ion ***,the antibacterial mechanism of AgCl-on-Ag NWs against MRSA was verified at the gene level by transcriptome analysis(RNA sequencing).Moreover,the fullthickness defect model verified that AgCl-on-Ag NWs reduced inflammatory cell infiltration and dramatically accelerated wound *** work provides a synergistic mechanism based on nanosilver surface engineering to eradicate the resistant bacteria that can alleviate drug resistance and develop an innovative approach for the treatment of bacterial infections.
