Spatial transcriptomics combined with single-nucleus RNA sequencing reveals glial cell heterogeneity in the human spinal cord

Relationship between parenting stress and behavioral and emotional problems in preschool children: A mediation effect analysis

BACKGROUND Emotional reactions,such as anxiety,irritability,and aggressive behavior,have attracted clinical attention as behavioral and emotional problems in preschool-age *** To investigate the current status of family rearing,parental stress,and behavioral and emotional problems of preschool children and to analyze the mediating effect of the current status of family rearing on parental stress and behavioral/emo-tional *** We use convenience sampling to select 258 preschool children in the physical examination center of our hospital from October 2021 to September *** children and their parents were evaluated using a questionnaire ***'s correlation was used to analyze the correlation between child behavioral and emotional problems and parental stress and family rearing,and the structural equation model was constructed to test the mediating *** The score for behavioral/emotional problems of 258 preschool children was(27.54±3.63),the score for parental stress was(87.64±11.34),and the score for parental family rearing was(31.54±5.24).There was a positive correlation between the behavioral and emotional problems of the children and the“hostile/mandatory”parenting style;meanwhile,showed a negative correlation with the“support/participation”parenting style(all Pchildren's behavior problems was 29.89%.CONCLUSION Parental family upbringing has a mediating effect between parental stress and behavioral and emotional problems of *** paying attention to the behavioral and emotional problems of preschool-age children,clinical medical staff should provide correct and reasonable parenting advice to their parents to promote the mental health of preschool-age children.

Critical reflection on the role of theater nurses in a multidisciplinary team for perioperative care in China

Objective: The aim of this article is to reflect on the role of theater nurses in a multidisciplinary team, understand the factors that have influenced theater nurses' practice, and improve the authors' clinical practice ***: The author used Smyth's model to guide the process of reflection on the practice issue. Critical reflection, critical emancipatory theory, reflexivity, and critical social theory were used to help the author analyze the factors that have affected theater nurses' practice in the ***: There are gaps between the espoused and enacted theories. A theater nurse's practice is determined by multiple factors, such as political, structural, social, historical, cultural issues, and so on. The hierarchy of the health context could hinder possible changes in theater nurses' practice. To better understand our practice and implement transformation, we should shape a supportive environment,bear in mind the practice motto of "patient-centered" care, and improve our knowledge and reflection ***: Reflection plays a significant role in the advancing of practice among theater nurses and needs to be combined with clinical practice. To provide the best service of care to perioperative patients, a theater nurse should have an insightful understanding of the factors that have influenced her/his behaviors historically, socially, and culturally. By improving their critical reflection skills,practitioners could gain knowledge from experience.

Influencing factors of the neurodevelopment of high-risk infants

Background High-risk infants refer to newborns exposed to high-risk factors in the prenatal, natal or postnatal period. High-risk infants are at high risk of developmental retardation, and early identification of developmental abnormalities plays a vital role in improving high-risk infants' quality of *** To describe the neurodevelopment of high-risk infants aged less than 1 year old, and to analyse the incidences and influencing factors of neurodevelopmental abnormalities in order to provide a basis for neurodevelopment monitoring and management of highrisk *** High-risk infants born between January 2016 and December 2016 in the maternity and infant health hospitals of three districts in Shanghai were followed *** Gesell Developmental Scale was used to assess the neurodevelopmental level at the time of recruitment(0-2 months) and at 9 months. Univariate and multivariate analyses of the influencing factors were *** 484 high-risk infants(male 51 %, female 49%)with an average gestation age of 36.5±2.2 weeks were recruited. At the time of recruitment, the average age was2.1(0.8) months, and the developmental quotient(DQ)scores of full-term high-risk infants in motor(t=3.542,p=0.001), cognitive(t=3.125, p=0.002), language(t=3.189, p=0.002) and social(t=3.316, p=0.001) areas were higher than those of preterm infants. The incidences of developmental abnormalities of full-term high-risk infants in motor(χ~2 =9.452, p=0.002), cognitive(χ~2=6.258, p=0.012), language(χ~2 =12.319, p =0.001) and social(χ~2 =6.811, p=0.009) areas were lower than the preterm infants. At 9 months, there was no difference in the DQ scores and incidences of developmental abnormalities in four areas between full-term and preterm high-risk infants, and the incidence of developmental abnormalities was around 10%.Conclusion The incidence of neurodevelopmental abnormalities in high-risk infants aged less than 1 year old is high. Epidemiological Characteristics of Plague in the Meriones unguiculatus Plague Focus--inner mongolia Autonomous Region,China,1950−2019

From 1950 to 2019,a total of 267 plague cases in humans were reported in the inner mongolia Autonomous Region with 133 deaths and 10,710 Yersinia pestis isolates;21 of these cases were reported from the Meriones unguiculatus(***)plague focus with 6 deaths and 6,771 *** to the plague situation in the inner mongolia Autonomous Region and the implementation of local preventive measures,the prevalence of plague in humans in the *** plague focus of the inner mongolia Autonomous Region could be divided into four *** first stage was the initial stage of plague prevention and control(1950–1959),with 0.80 cases annually and a case fatality rate of 37.50%.The second stage was the plague eradication stage(1960–1979),with 0.15 cases annually and a case fatality rate of 33.33%.The third stage was the plague surveillance stage(1980-1999),with 0.25 cases annually and a case fatality rate of 20.00%.The fourth stage is the comprehensive prevention and control stage under the emergency system(2000-2019),with 0.25 cases annually and a case fatality rate of 20.00%.The surveillance of rodent density(1981-2019)and studies on plague-related factors among *** have shown that the higher the *** density,the higher the nocturnal rodent capture rate(r=0.670,p<0.05)and the higher the indirect hemagglutination assay(IHA)positive rate of ***(r=0.344,p<0.05);the higher the percentage of hosts infected,the lower the *** density(r=-0.361,p<0.05)and the lower the IHA positive rate of ***(r=-0.337,p<0.05);the higher the percentage of nests infected with fleas,the lower the IHA positive rate of ***(r=-0.348,p<0.05).Together,these results suggest that it is necessary to simultaneously monitor the pathogens,serology,and vector index of *** to accurately reflect the plague prevalence among local *** bubonic plague is the main plague type of the *** plague focus,s

Sevoflurane and nitric oxide synthase expression in rat cochlea

Sevoflurane exhibits anesthetic action by inhibiting the auditory cortex, brain stem nitric oxide synthase activity, and reducing nitric oxide （NO）, thereby interfering with the hearing process. However, the influence of sevoflurane on peripheric receptor （cochlea） NO remains poorly understood. Results from the present study showed that sevoflurane downregulated cochlear inducible NO synthase, endothelial NO synthase and neuronal NO synthase expression in a dose dependent manner. This suggests that sevoflurane can decrease cochlear NO synthase expression in a dose dependent manner.

Extracts from rabbit skin inflamed by the vaccinia virus attenuate bupivacaine-induced spinal neurotoxicity in pregnant rats

Extracts from rabbit skin inflamed by the vaccinia virus can relieve pain and promote repair of nerve injury. The present study intraperitoneally injected extracts from rabbit skin inflamed by the vaccinia virus for 3 and 4 days prior to and following intrathecal injection of bupivacaine into pregnant rats. The pain threshold test after bupivacaine injection showed that the maximum possible effect of tail-flick latency peaked 1 day after intrathecal injection of bupivacaine in the extract-pretreatment group, and gradually decreased, while the maximum possible effect in the bupivacaine group continued to increase after intrathecal injection of bupivacaine. Histological observation showed that after 4 days of intrathecal injection of bupivacaine, the number of shrunken, vacuolated, apoptotic and caspase-9-positive cells in the dorsal root ganglion in the extract-pretreatment group was significantly reduced compared with the bupivacaine group. These findings indicate that extracts from rabbit skin inflamed by the vaccinia virus can attenuate neurotoxicity induced by intrathecal injection of bupivacaine in pregnant rats, possibly by inhibiting caspase-9 protein expression and suppressing nerve cell apoptosis.

Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. Our results suggest that NESTS is feasible and cost-effective as a first-tier NBS program, which will change the status of current clinical practice of NBS in China.

Optimization and application of a dried blood spot-based genetic screening method for thalassemia in Shenzhen newborns

Background To optimize and apply an approach suitable for large-scale neonatal thalassemia genetic screening in China,thalassemia genotypes were determined by polymerase chain reaction-reverse dot blot using DNA extracted from dried blood spots(DBS)obtained from newborn screening *** Firstly,the most suitable commercial DNA extraction kit for DBS was ***,the appropriate amount of DBS required for the automated high-throughput DNA extraction system was ***,the thalassemia prevalence and genotype spectrum in Shenzhen were investigated in 2028 newborns using the optimized screening *** The Magentec extraction kit was best suited for the automated DBS DNA extraction system using eight 3-mm DBS *** neonatal thalassemia prevalence in Shenzhen was 9.12%;6.31%α-thalassemia,2.37%β-thalassemia,and 0.44%α-/β-*** Genetic screening based on DBS can precisely identify the thalassemia ***α-andβ-thalassemia are widely distributed in Shenzhen *** genetic screening is important for establishing a comprehensive thalassemia prevention program and for public education.
