Non-Structural Protein 5 of Zika Virus Interacts with p53 in Human Neural Progenitor Cells and Induces p53-Mediated Apoptosis

Zika virus(ZIKV) infection could disrupt neurogenesis and cause microcephaly in neonates by targeting neural progenitor cells(NPCs). The tumor suppressor p53-mediated cell cycle arrest and apoptotic cell death have been suggested to be activated upon ZIKV infection, yet the detailed mechanism is not well understood. In the present study, we investigated the effects of ZIKV-encoded proteins in the activation of p53 signaling pathway and found that, among the ten viral proteins,the nonstructural protein 5(NS5) of ZIKV most significantly activated the transcription of p53 target genes. Using the immunoprecipitation-coupled mass spectrometry approach, we identified that ZIKV-NS5 interacted with p53 protein. The NS5-p53 interaction was further confirmed by co-immunoprecipitation and GST pull-down assays. In addition, the MTase domain of NS5 and the C-terminal domain of p53 were mapped to be responsible for the interaction between these two proteins. We further showed that ZIKV-NS5 was colocalized with p53 and increased its protein level in the nuclei and able to prolong the half-life of p53. Furthermore, lentivirus-mediated expression of ZIKV-NS5 in hNPCs led to an apparent cell death phenotype. ZIKV-NS5 promoted the cleavage of PARP1 and significantly increased the cell apoptosis of h *** together, these findings revealed that ZIKV-NS5 is a previously undiscovered regulator of p53-mediated apoptosis in hNPCs, which may contribute to the ZIKV-caused abnormal neurodevelopment.

Zebrafish（Danio rerio） is an ideal model for studying the mechanism of infectious disease and the interaction between host and *** a teleost,zebrafish has developed a complete immune system which is similar to ***,the easy acquirement of large amounts of transparent embryos makes it a good candidate for gene manipulation and drug *** a zebrafish infection model,all of the site,timing,and dose of the bacteria microinjection into the embryo are important factors that determine the bacterial infection of ***,we established a multi-site infection model in zebrafish larvae of 36 hours post-fertilization（hpf） by micro-injecting wild-type or GFP-expressing Staphylococcus aereus（***） with gradient burdens into different embryo sites including the pericardial cavity（PC）,eye,the fourth hindbrain ventricle（4V）,yolk circulation valley（YCV）,caudal vein（CV）,yolk body（YB）,and Duct of Cuvier（DC） to resemble human infectious *** the combination of GFP-expressing *** and transgenic zebrafish Tg（corola：eGFP;lyz：Dsred） and Tg（lyz：Dsred） lines whose macrophages or neutrophils are fluorescent labeled,we observed the dynamic process of bacterial infection by in vivo multicolored confocal fluorescence *** of zebrafish embryo survival, bacterial proliferation and myeloid cells phagocytosis show that the site- and dose-dependent differences exist in infection of different bacterial entry *** work provides a consideration for the future study of pathogenesis and host resistance through selection of multi-site infection *** interaction mechanisms between pathogenic bacteria virulence factors and the immune responses of zebrafish could be determined through zebrafish multi-site infection model.

The success of combined antiretroviral therapy （cART） has dramatically improved the clinical outcomes of HIV infection and made HIV infection a chronic disease. Nonetheless, cART alone cannot eliminate viral reservoirs and fully recover patients＇ health, so the patients have to receive this treatment for lifetime. Therefore, researchers have focused their efforts on the cure for HIV infection. In recent years, some intriguing and inspiring cases indicated that the cure of HIV infection （HIV cure） or durable alleviation of HIV infection may not be a dream any more. ＇Berlin Patient＇＇Mississippi Baby＇, and probably ＇Long Beach Baby＇ have brought great hope to the HIV cure. Many scientists believe that the HIV cure is just a question of time.

Clinical outcomes of patients with chronic hepatitis C after generic direct-acting antiviral treatments in Vietnam: A retrospective analysis

Objective:To evaluate the outcomes of generic direct-acting antiviral treatments for chronic hepatitis C in ***:The medical records of 522 patients(median 45 years;Female,25.3%)with chronic hepatitis C treated at a tertiary hospital in northern Vietnam in 2016 were retrospectively ***:Female patients were significantly older than male patients(median 52,IQR 41-59 vs.43 years,IQR:39-55;Pinfection with hepatitis B virus or human immunodeficiency virus was noted in 5.8%and 3.1%of patients,*** were treated with ledipasvir/sofosbuvir with or without ribavirin(70.9%),sofosbuvir/pegylated-interferon+ribavirin(13.2%),daclatasvir/sofosbuvir with or without ribavirin(12.5%),or sofosbuvir/ribavirin(3.4%),and 96.4%(n=503)completed the direct-acting antiviral *** patient discontinued treatment due to adverse event(s).A sustained virologic response 12 weeks after the end of the treatment(SVR12)was evaluated in 62.6%of *** sustained virologic response 12 weeks after the end of the treatment was 98.7%regardless of HCV genotypes or direct-acting antiviral *** severity of liver stiffness was significantly decreased from 10.2 to 6.3 kilopascals measured by transient elastography by the treatment(P<0.001).Among patients who completed the directacting antiviral treatment,17.7%returned for further follow-ups after ***:In Vietnam,the current generic direct-acting antiviral treatment for chronic hepatitis C was effective regardless of HCV genotypes and direct-acting antiviral regimens with the attenuation of liver *** is feasible to implement direct-acting antiviral treatment to cure chronic hepatitis C patients at any liver fibrosis stages in Vietnam.

Dear Editor,Glioblastoma(GBM)is a lethal primary brain cancer,with a median survival of less than 2 years.1 Immune checkpoint blockades(ICBs)have revolutionized cancer therapy in the last decade,but they have little clinical benefit in GBM.1 Genomic and transcriptomic analysis revealed a significant enrichment of PTEN mutations in GBM patients resistant to ICBs.2 PTEN deficiency activates the phosphatidylinositol 3-kinase(PI3K)-AKT pathway to shape an immunosuppressive microenvironment.
