research on Spatial-Temporal Characteristics and Driving Factor of agricultural Carbon Emissions in China

Macroscopic grasp of agricultural carbon emissions status, spatial-temporal characteristics as well as driving factors are the basic premise in further research on China’s agricultural carbon emissions. Based on 23 kinds of major carbon emission sources including agricultural materials inputs, paddy ifeld, soil and livestock breeding, this paper ifrstly calculated agricultural carbon emissions from 1995 to 2010, as well as 31 provinces and cities in 2010 in China. We then made a decomposed analysis to the driving factors of carbon emissions with logarithmic mean Divisia index （LMDI） model. The results show：（1） The amount of agricultural carbon emissions is 291.1691 million t in 2010. Compared with 249.5239 million t in 1995, it increased by 16.69%, in which, agricultural materials inputs, paddy ifeld, soil, enteric fermentation, and manure management accounted for 33.59, 22.03, 7.46, 17.53 and 19.39%of total agricultural carbon emissions, respectively. Although the amount exist ups and downs, it shows an overall trend of cyclical rise; （2） There is an obvious difference among regions：the amount of agricultural carbon emissions from top ten zones account for 56.68%, while 9.84%from last 10 zones. The traditional agricultural provinces, especially the major crop production areas are the main source regions. Based on the differences of carbon emission rations, 31 provinces and cities are divided into ifve types, namely agricultural materials dominant type, paddy ifeld dominant type, enteric fermentation dominant type, composite factors dominant type and balanced type. The agricultural carbon emissions intensity in west of China is the highest, followed by the central region, and the east zone is the lowest; （3） Compared with 1995, efifciency, labor and structure factors cut down carbon emissions by 65.78, 27.51 and 3.19%, respectively;while economy factor increase carbon emissions by 113.16%.

High-Throughput Screening of Nanoparticle-Stabilizing Ligands:Application to Preparing Antimicrobial Curcumin Nanoparticles by Antisolvent Precipitation

Water-dispersible curcumin nanoparticles were prepared by bottom-up antisolvent precipitation approach. A new high-throughput screening technique was developed for selecting appropriate ligands stabilizing the nanoparticles in aqueous medium and improving their performance. The initial set of twenty-eight potential stabilizing ligands was evaluated based on their capacity to improve curcumin dispersibility in aqueous medium. The performance of four promising ligands(amino acid proline, polyphenol tannic acid, polycation Polyquaternium 10, and neutral polymer polyvinylpyrrolidone) was tested in ultrasound-aided antisolvent precipitation trials. Using the selected stabilizing ligands diminished the average particle size from ca. 1,200 to 170–230 nm, reduced their dispersity, improved stability, and allowed reaching curcumin concentration of up to 1.4 m M in aqueous medium. Storage stability of the aqueous nanodispersions varied from 2 days to 2 weeks, depending on stabilizing ligand. Studying the effects of ionic strength and pH on size and f-potential of the particles suggested that electrostatic forces and hydrophobic interactions could be the major factors affecting their stability. The ligand-protected nanoparticles showed minimal inhibitory concentration of 400 or500 μM toward Escherichia coli. We suggest that the presented screening approach may be useful for preparing nanoparticles of various poorly water-soluble bioactive materials.

SCFA Profile of Rice RS Fermentation by Colonic Microbiota, <i>Clostridium butyricum</i>BCC B2571, and <i>Eubacterium rectale</i>DSM 17629

Resistant starch type 3 (RS3) produced from high amylose food sources through retrogradation or enzymatic process is known to have physiological function as dietary fiber. Fermentation of RS3 by colonic microorganisms produced SCFA (acetate, propionate, and butyrate), maintained the health of colon, balance of gut microbiota, preventing inflammatory bowel diseases (IBD) and colon cancer. RS3 in this study was produced from IR-42 and Inpari-16 broken rice by enzymatic treatment (combination of amylase-pullulanase). The Resistant Starch was fermented for 12 and 24 h by colonic microbiota (extracted from healthy human subject), Clostiridium butyricum BCC-B2571, or Eubacterium rectale DSM 17629. SCFA produced was analyzed by gas chromatography. Treatment by amylase-pullulanase combination was advantageous to increase their RS3 content. The result showed that after enzymatic process, the RS3 content of IR-42 (41.13%) was not significantly different (p 0.05) from that of Inpari-16 (37.70%). High concentration of acetate (82.5 mM) and propionate (7.5 mM) were produced by colonic microbiota after 12 h fermentation and best concentration of butyrate (6.8 mM) was produced by colonic microbiota after 24 h fermentation. It is clear that utilization of colonic microbiota rather than single strain was better in the production of SCFA.

Soil Aggregate Stability and Aggregate-Associated Carbon Under Different Tillage Systems in the North China Plain

The inlfuences of tillage systems on soil carbon （C） stocks have been studied extensively, but the distribution of soil C within aggregate fractions is not well understood. The objective of this study was to determine the inlfuences of various tillage systems on soil aggregation and aggregate-associated C under wheat （Triticum aestivum L.） and corn （Zea mays L.） double cropping systems in the North China Plain. The experiment was established in 2001, including four treatments：moldboard plow （MP） with residue （MP＋R） and without residue （MP-R）, rotary tillage with residue （RT）, and no-till with residue （NT）. In 2007 soil samples were collected from the 0-5, 5-10, and 10-20 cm depths, and were separated into four aggregate-size classes （〉2 000, 250-2 000, 53-250, and〈53 μm） by wet-sieving method. Aggregate-associated C was determined, and the relationships between total soil C concentration and aggregation-size fractions were examined. The results showed that NT and RT treatments signiifcantly increased the proportion of macroaggregate fractions （〉2 000 and 250-2 000 μm） compared with the MP-R and MP＋R treatments. Averaged across all depths, mean weight diameters of aggregates （MWD） in NT and RT were 47 and 20% higher than that in MP＋R. The concentration of bulk soil organic C was positively correlated with MWD （r=0.98; P=0.024） and macroaggregate fraction （r=0.96; P=0.036） in the 0-5 cm depth. In the 0-20 cm depth, comparing with MP＋R, total C occluded in the〉2 000 μm fraction was increased by 9 and 6%under NT and RT, respectively. We conclude that adoption of conservation tillage system, especially no-till, can increase soil macro-aggregation and total C accumulation in macroaggregates, which may improve soil C sequestration in the intensive agricultural region of the North China Plain.

New Temperature Sensitive Genic Male Sterile Lines with Better Outcrossing Ability for Production of Two-Line Hybrid Rice

An investigation was carried out with three newly developed temperature sensitive genic male sterile （TGMS） lines for their floral traits, seed production potential and outcrossing ability in ten cross combinations. In the TGMS lines, fertile pollens had an average diameter of 0.89 mm while the sterile pollens was with 0.02 mm ***-29-150GY produced the biggest fertile pollens with 0.92 mm and other two lines produced relatively smaller pollens with 0.91 and 0.85 ram. Pollen fertility during the fertility reversion period was an average of 60.7%. TS-29-150GY had the maximum of 66.9% spikelet fertility whereas other two lines （TNAU18S and TNAU60S） had relatively lower spikelet fertility of 27.8% and 26.7%, respectively. Average of 17.00 g of seed yield was obtained in the TGMS lines during the fertility reversion period. TS-29- 150GY had the highest value of 21.20 g of seed yield while TNAU18S and TNAU60S produced 16.6 g and 13.2 g of seed yield, respectively. The low seed production ability of these three TGMS lines was attributed only to the environmental conditions prevailing during the period. All three TGMS lines had considerable outcrossing potential of 41.2%, 24.6% and 25.0%, respectively. The cross combinations viz. TNAU18S/IET21508 （36 g/plant）, TNAU18S/IET21044 （13 g/plant）, TNAU18S/IET21009 （26.5 g/plant）, TNAU60S/CB-09-106 （26.2 g/plant）, TNAU60S/IET21009 （14 g/plant） and TS29-150-GY/DRR 3306 （39.2 g/plant） showed perfect synchronization with acceptable hybrid seed yield, indicating suitability of TGMS system under Indian condition. Based on the outcrossing related traits viz. panicle exertion, angle of glume opening, stigma length and pollen size, TNAU18S was identified as the best, followed by TS-29-150GY.

Identification of Behavior of Voluntary Traceability and Analysis of Its Determinants: A Case Study of Hog Slaughtering and Processing Firms in Zhejiang Province,China

In recent years, the Chinese government has highlighted the importance of adopting hog safety/quality traceability, and a growing amount of research continues to entice firms to adopt traceability systems. In this study, a survey was conducted on a sample of pig slaughtering and processing firms in Zhejiang, China through personal interviews and emails. The aim of this study was to examine the determinants of firm behavior on the implementation of voluntary traceability systems with more stringent standards and controls than those of the mandatory system in China. The results revealed that motivation based on product quality improvement, capital ability and role perception （business type） had significantly positive relationships with a firm＇s voluntary traceability. Other incentives, such as operation improvement, recall risk reduction, reduced occurrence of safety issues, and technical strength were not found to be supportive in our study. This study provides an opportunity to better understand the determinants of firm behavior on voluntary traceability, particularly in light of the fact that some Chinese firms are facing the threat of criminal action for the use of illegal additives and the abuse of Clenbuterol. Policy recommendations on encouraging the implementation of pork safety voluntary traceability by hog slaughtering and processing firms are also discussed.

Localization of the Arabidopsis Senescence- and Cell Death-Associated BFN1 Nuclease： From the ER to Fragmented Nuclei

Plant senescence- or PCD-associated nucleases share significant homology with nucleases from different organisms. However, knowledge of their function is limited. Intracellular localization of the Arabidopsis senescence- and PCD-associated nuclease BFN1 was investigated. Analysis of BFN1-GFP localization in transiently transformed tobacco protoplasts revealed initial localization in filamentous structures spread throughout the cytoplasm, which then clustered around the nuclei as the protoplasts senesced. These filamentous structures were identified as being of ER origin. In BFN1- GFP-transgenicArabidopsis plants, similar localization of BFN1-GFP was observed in young leaves, that is, in filamentous structures that reorganized around the nuclei only in senescing cells. In late senescence, BFN1-GFP was localized with fragmented nuclei in membrane-wrapped vesicles. BFNI＇s postulated function as a nucleic acid-degrading enzyme in senescence and PCD is supported by its localization pattern. Our results suggest the existence of a dedicated compartment mediating nucleic acid degradation in senescence and PCD processes.

mRNA profiles of piRNA pathway genes in emerald ash borer Agrilus planipennis

Recent advances of Piwi-interacting RNAs （piRNAs） have enabled a better understanding of their putative roles in the protection of gerrn cells against selfish genetic elements. The germ plasm proteins namely, PIWI and VASA, are key players in regulation of piRNAs. However, the biogenesis of piRNAs remains poorly understood. We report the characterization and the transcript profiles of a PIWI and VASA gene in various tissues and developmental stages of the emerald ash borer （Agrilusplanipennis）, an exotic invasive insect pest that has attained a high impact status worldwide. The deduced amino acid sequences of both these genes （herein referred to ＂ApPIWI and ApVASA＂） revealed homology with similar proteins from other insect and non-insect taxa. Peak transcript levels for ApPIWI and ApVASA were observed in the ovaries （compared to the other tissues assayed） and in adults （compared to the other developmental stages assayed）. To our knowledge this is the first report of profiling piRNA pathway genes in A. planipennis. These results provide new insights into the putative function of the PIWI and VASA proteins in insects.

Deciphering Ascorbic Acid Regulatory Pathways in Ripening Tomato Fruit Using a Weighted Gene Correlation Network Analysis Approach

Genotype is generally determined by the co-expression of diverse genes and multiple regulatory pathways in plants. Gene co-expression analysis combining with physiological trait data provides very important information about the gene function and regulatory mechanism. L-Ascorbic acid （AsA）, which is an essential nutrient component for human health and plant metabolism, plays key roles in diverse biological processes such as cell cycle, cell expansion, stress resistance, hormone synthesis, and signaling. Here, we applied a weighted gene correlation network analysis approach based on gene expression values and AsA content data in ripening tomato （Solanum lycopersicum L.） fruit with different AsA content levels, which leads to identification of AsA relevant modules and vital genes in AsA regulatory pathways. Twenty- four modules were compartmentalized according to gene expression profiling. Among these modules, one negatively related module containing genes involved in redox processes and one positively related module enriched with genes involved in AsA biosynthetic and recycling pathways were further analyzed. The present work herein indicates that redox pathways as well as hormone-signal pathways are closely correlated with AsA accumulation in ripening tomato fruit, and allowed us to prioritize candidate genes for follow-up studies to dissect this interplay at the biochemical and molecular level.

Fermentation product of RS3 inhibited proliferation and induced apoptosis in colon cancer cell HCT-116

Cell free supernatant containing short chain fatty acid (SCFA) resulted from fermentation of resistant starch type three (RS3) by Clostridium butyricum BCC B2571 or Eubacterium rectale DSM 17629 were investigated for their ability to inhibit proliferation and induce apoptosis of human colon cancer cell line HCT-116. HCT-116 was cultured in complete medium and after 50% confluent, incubation was continued for another 48 hours in the absence or presence cell free supernatant containing SCFA mixture at butyrate levels up to 10 mM. The study revealed that the proliferation inhibition effect was higher (>80%) on HCT-116 treated with supernatant of C. butyricum BCC B2571 than that (<70%) of HCT-116 treated with supernatant of E. rectale DSM 17629. The cells were induced to undergo apoptosis by both supernatant. The apoptosis occured through mitochondrial pathway by changing the expression of gene Bcl-2 and Bax, thus incresed the Bax/Bcl-2 ratio by more than 3.5 fold. The protein caspase-3 was increased by more than 250% in the presence of the cell free supernatant.
