Understanding host-graft crosstalk for predicting the outcome of stem cell transplantation

Mesenchymal stromal cells(MSCs)hold great promise for tissue regeneration in debilitating disorders.Despite reported improvements,the short-term outcomes of MSC transplantation,which is possibly linked to poor cell survival,demand extensive investigation.Disease-associated stress microenvironments further complicate outcomes.This debate underscores the need for a deeper understanding of the phenotypes of transplanted MSCs and their environment-induced fluctuations.Additionally,questions arise about how to predict,track,and comprehend cell fate post-transplantation.In vivo cellular imaging has emerged as a critical requirement for both short-and long-term safety and efficacy studies.However,translating preclinical imaging methods to clinical settings remains challenging.The fate and function of transplanted cells within the host environment present intricate challenges,including MSC engraftment,variability,and inconsistencies between preclinical and clinical data.The study explored the impact of high glucose concentrations on MSC survival in diabetic environments,emphasizing mitochondrial factors.Preserving these factors may enhance MSC survival,suggesting potential strategies involving genetic modification,biomaterials,and nanoparticles.Understanding stressors in diabetic patients is crucial for predicting the effects of MSC-based therapies.These multifaceted challenges call for a holistic approach involving the incorporation of large-scale data,computational disease modeling,and possibly artificial intelligence to enable deterministic insights.

The construction industry is a major contributor to environmental pollution.The effect of the construction industry on the environment may be mitigated using eco-friendly construction materials,such as biocomposites.Once developed,biocomposites may offer a viable alternative to the current materials in use.However,biocomposites are lagging in terms of adoption and eventual use in the construction industry.This article provides insights into the steps for biocomposites to become a product that is ready to use by the construction industry in a structural role.The development and the adoption of such a material is tackled with the use of two concepts,i.e.,technology readiness level and roadmapping,and explored in a case study on the“liquid wood”.Furthermore,interviews in the construction industry are carried out to identify the industry’s take on biocomposites.A customized roadmap,which underlines a mostly nontechnical perspective concerning this material,has emerged.Additionally,the adoption and diffusion issues that the“liquid wood”may encounter are outlined and complemented with further recommendations.

Assessment of recharge capacity potential of groundwater using comparative multi-criteria decision analysis approaches

Groundwater resources have always been some of the most valuable resources of human settlements.Climate changes and ever-increasing water demands registered in the last century have led to diminishing levels of groundwater reserves,as well as reduced recharging potential.Therefore,in order to use groundwater aquifers in a sustainable manner,it is required to identify areas with higher replenishing potential.The current study addresses the issue of generating a map for identifying differently ranked groundwater recharging potential values,in the aquifers of the Moldavian Plain region,Romania.For the purpose of conducting the analysis,maps were created through GIS based multi-criteria Analytic Hierarchy Process(AHP)and Catastrophe Theory(CT),with seven relevant,thematic,spatial layers:precipitation distribution,lithological strata,soil texture,declivity,drainage density,land use and the distribution of groundwater level tendencies.The results of the two methods of analysis are similar.Prediction differences are of maximum 3%,in the case of extreme classes(very bad and very good)and in the case of middle classes the deviation is not greater than 0.4%.Following the validation of the results generated by the two methods that were applied,it was observed that the predictions offered by CT are more accurate.This aspect can be based on the fact that the main factors that contribute to the prediction are different.This type of workflow emphasizes the necessity of implementing appropriate groundwater management plans for mitigating reservoir scarcity/depletion,and recommending sustainable solutions for future groundwater exploitation practices.

BACKGROUND Colon cancer is one the most common forms of cancer in both sexes.Due to important progress in the field of early detection and effective treatment,colon and rectal cancer survivors currently account for 10%of cancer survivors worldwide.However,the effects of anti-cancer treatments,especially oxaliplatinbased chemotherapy,on the quality of life(QoL)have been less evaluated.Although the incidence of severe chemotherapy-induced neuropathy(CIPN)in clinical studies is below 20%,data from real-world studies is scarce,and CIPN is probably under-reported due to patient selection and the patients’fear that reporting side-effects might lead to treatment cessation.AIM To determine the impact of CIPN on QoL in colorectal cancer patients with a recent history of oxaliplatin-based chemotherapy.METHODS We performed a prospective cross-sectional study in two major Romanian oncology tertiary hospitals—the Regional Institute of Oncology Ia?i(Iasi,Romania)and the Fundeni Clinical Oncology Institute(Bucharest,Romania).All consecutive patients with colon or rectal cancer,undergoing Oxaliplatin-based chemotherapy that consented to enroll in the study,were assessed by means of two questionnaires—the EORTC QQ-CR29(quality of life in colon and rectal cancer patients)and the QLQ-CIPN20(assessment of neuropathy).Several demographical,social,clinical and treatment data were also collected.Statistical analysis was performed by means of SPSS v20.The student t test was used to assess the relationship between the QLQ-CIPN20 and QLQ-CR29 results.Kaplan Meyer-curves were used to report 3-year progression-free survival(PFS)in patients that discontinued chemotherapy vs those that completed the recommended course.RESULTS Of the 267 patients that fulfilled the inclusion criteria in the pre-specified time frame,101(37.8%)agreed to participate in the clinical study.At the time of the enrolment in the study,over 50%of the patients had recently interrupted their oxaliplatin-based chemotherapy,most often due to neuropathy.Almost 85%of the resp

A Comparative Analysis of Binary Logistic Regression and Analytical Hierarchy Process for Landslide Susceptibility Assessment in the Dobrovǎt River Basin,Romania

A correct assessment of the landslide susceptibility component is extremely useful for the diminution of associated potential risks to local economic development, particularly in regard to land use planning and soil conservation. The purpose of the present study was to compare the usefulness of two methods, i.e., binary logistic regression(BLR) and analytical hierarchy process(AHP), for the assessment of landslide susceptibility over a 130-km^2 area in the Moldavian Plateau(eastern Romania) region, where landslides affect large areas and render them unsuitable for agriculture. A large scale inventory mapping of all types of landslides(covering 13.7% of the total area) was performed using orthophoto images, topographical maps, and field surveys. A geographic information system database was created, comprising the nine potential factors considered as most relevant for the landsliding process. Five factors(altitude, slope angle, slope aspect, surface lithology, and land use) were further selected for analysis through the application of a tolerance test and the stepwise filtering procedure of BLR. For each predictor, a corresponding raster layer was built and a dense grid of equally spaced points was generated, with an approximately equal number of points inside and outside the landslide area, in order to extract the values of the predictors from raster layers. Approximately half of the total number of points was used for model computation, while the other half was used for validation. Analytical hierarchy process was employed to derive factor weights, with several pair-wise comparison matrices being tested for this purpose. The class weights, on a scale of 0 to 1, were taken as normalized landslide densities. A comparison of results achieved through these two approaches showed that BLR was better suited for mapping landslide susceptibility, with 82.8% of the landslide area falling into the high and very high susceptibility classes. Adipose tissue(AT)is recognized as a complex organ involved in major homeostatic body functions,such as food intake,energy balance,immunomodulation,development and growth,and functioning of the reproductive organs.The role of AT in tissue and organ homeostasis,repair and regeneration is increasingly recognized.Different AT compartments(white AT,brown AT and bone marrow AT)and their interrelation with bone metabolism will be presented.AT-derived stem cell populations-adipose-derived mesenchymal stem cells and pluripotentlike stem cells.Multilineage differentiating stress-enduring and dedifferentiated fat cells can be obtained in relatively high quantities compared to other sources.Their role in different strategies of bone and fracture healing tissue engineering and cell therapy will be described.The current use of AT-or AT-derived stem cell populations for fracture healing and bone regenerative strategies will be presented,as well as major challenges in furthering bone regenerative strategies to clinical settings.

A knowledge-driven approach is proposed for assessing the music influence on university students.The proposed method of modeling and conducting the interactive pilot study can be useful to convey other surveys,interviews,and experiments created in various phases of the user interface(UI)design processes,as part of a general human-computer interaction(HCI)methodology.Benefiting from existing semantic Web and linked data standards,best practices,and tools,a microservice-oriented system is developed as a testbed platform able to generate playlists in a smart way according to users’music preferences.This novel approach could bring also benefits for user interface adaptation by using semantic Web techniques.Statistical analysis based on the ANOVA method and post-experiment survey data led to the conclusion that music listened has a significant impact on students’cognitive abilities in various contexts.All obtained results were semantically enhanced by using different conceptual models in order to create a knowledge graph providing support for automated reasoning.Also,a knowledge-based persona Web smart editor was implemented in order to include music preferences for certain categories of the potential users operating a specific interactive system.

Exact solution for the rotational flow of a generalized second grade fluid in a circular cylinder

This paper deals with the rotational flow of a generalized second grade fluid, within a circular cylinder, due to a torsional shear stress. The fractional calculus approach in the constitutive relationship model of a second grade fluid is introduced. The velocity field and the resulting shear stress are determined by means of the Laplace and finite Hankel transforms to satisfy all imposed initial and boundary conditions. The solutions corresponding to second grade fluids as well as those for Newtonian fluids are obtained as limiting cases of our general solutions. The influence of the fractional coefficient on the velocity of the fluid is also analyzed by graphical illustrations.

Unsteady flow of viscoelastic fluid between two cylinders using fractional Maxwell model

The unsteady flow of an incompressible fractional Maxwell fluid between two infinite coaxial cylinders is studied by means of integral transforms. The motion of the fluid is due to the inner cylinder that applies a time dependent tor- sional shear to the fluid. The exact solutions for velocity and shear stress are presented in series form in terms of some generalized functions. They can easily be particularized to give similar solutions for Maxwell and Newtonian fluids. Fi- nally, the influence of pertinent parameters on the fluid motion, as well as a comparison between models, is highlighted by graphical illustrations.
