Dear Editor,Two distinct pathways,non-homologous end joining(NHEJ)and homologous recombination(HR)repair,have evolved to repair DNA double-strand breaks(DSBs),the most deleterious type of DNA *** NHEJ and HR are often associated with a high incidence of tumorigenesis and the early onset of aging.A number of methods and tools have been developed to help elucidate the regulatory mechanisms of the two repair pathways in the *** vitro biochemical assays employ 32P-labeled broken DNA fragments and cell extracts to evaluate DSB repair *** DSB sites,several DSB repair factors form foci,which can be visualized through immunofluorescence and *** them,phosphorylated H2AX at the Ser139 residue is often considered a DSB marker,and its formation and removal at different time points post the induction of DSBs can be used to reflect the repair capacity and genomic *** addition,by quantifying the kinetics of the recruitment of specific NHEJ and HR factors such as 53BP1,DNA-PKcs,MRE11,RPA2,and RAD51,one would be able to make judgments on whether NHEJ,HR,or both pathways is/are affected and sometimes at which steps DSB repair pathways are *** alternative approach for kinetic analysis is to quantify the microirradiation-induced recruitment of repair ***,comet assays are also utilized to measure DNA damage-induced genomic stability at the single-cell ***,with these assays,it is still difficult to measure NHEJ and HR efficiency in a relatively less time-consuming and more quantitative manner.

Objective:To determine distribution of arthropods in processed rice products such as rice flour and rice cereal-based infant ***:Random samples of rice flour and rice cereal-based infant food purchased from commercial outlets were examined for the presence of arthropods using a modified Berlese Tullgren Funnel *** were mounted prior to identification and weevils were directly ***:For non-expired products, infestation was found in 6.7%of rice flour and none was found in rice cereal-based infant food *** arthropods found in the flour samples were Cheyletus spp.,Suidasia pontifica(S. pontifica),Tarsonemus spp.,Tyrophagus putrescentiae(***),Sitophilus granarius(S. granarius) and Sitophilus oryzae(***).Others which cannot be identified were Oribatid and Prostigmatid *** most common mites in rice flour were Tarsonemus spp.(69.1%),followed by ***(18.2%).For expired products,only one sample of rice cereal-based infant food was infested and the infestation was by mites of the family ***:This study demonstrates the presence of 4 allergenic species of ***,***,*** and *** in rice *** arthropods can contribute to the incidence of anaphylaxis upon consumption by atopic *** was no infestation of arthropods in rice cereal-based infant food surveyed except for an expired product in a moderate rusty tin container.

In vitro and in vivo bactericidal activity of Vitex negundo leaf extract against diverse multidrug resistant enteric bacterial pathogens

Objective:To investigate in vitro and in vivo antibacterial potentials of Vitex negundo(V. negundo) leaf extracts against diverse enteric ***:Water and methanol extracts of *** leaves were evaluated against enteric bacterial pathogens by using standard disc diffusion,viable bacterial cell count methods,determination of minimum inhibitory concentrations(MIC) and minimum bactericidal concentrations(MBC).Results:Methanol extract of *** leaves showed potent antibacterial activity(inhibition zone:9.9- 22.6 mm,MIC: 200-3 200μg/mL,MBC:200-6 400μg/mL) against all the pathogenic enleric bacteria(Vibrio cholerae,Vibrio parahaemolyticus,Vibrio mimicus,Echerichia *** spps.,and Aeromonas spps) *** extract of *** leaves showed potent bactericidal activity both in vitro laboratory conditions(MBC,200-400μg/mL) and in the intestinal environment(Dose,1-2 mg/mL) of infant mice against pathogenic Vibrio cholerae,the major causative agent of cholera. Furthermore,assays using the mice cholera model showed that *** methanol extract can protect mice from Vibrio cholerae infection and significantly decrease the mortality rate(P<0.0001).Conclusioas:For the first time we showed that methanol extract of *** leaves exhibited strong vibriocidal activity both in vitro and in vivo ***,it will be useful to identify and isolate the active compounds of this extract that could be a good alternative of antibiotics to treat cholera.

Background Lower exercise tolerance is an important component of asthma and the possible effects of non-invasive ven-tilation on exercise capacity in individuals with severe therapy-resistant asthma(STRA)are *** study aimed to evaluate the immediate effect of continuous positive airway pressure(CPAP)on exercise tolerance in children with *** We performed a controlled,randomized,crossover clinical trial including subjects aged 6 to 18 years old diagnosed with ***,anthropometric and lung function data were *** participants in the intervention group(IG)used CPAP(PEEP 10cmH2O and FiO20.21)for a period of 40 *** in the control group(CG)used CPAP with mini-mum PEEP at 1 cmH20 also for 40 ***,subjects from both groups underwent cardiopulmonary exercise testing(CPET).After a 15-day washout period,on a subsequent visit,subjects participated in the opposite group to the initial *** Thirteen subjects with a mean age of 12.30±1.7 years were *** variables of peak expiratory flow(PEF)and forced expiratory volume in the first second(FEV1)before using CPAP and after performing CPET did not show sig-nificant *** CPET results,there was no significant difference(P=0.59)between groups at peak exercise for oxygen consumption—VO2(CG:33.4±6.3 and IG:34.5±5.9,mL kg^(-1)min^(-1)).However,the IG(12.4±2.1)presented a total test time(min)significantly(P=0.01)longer than the CG(11.5±1.3).Conclusion The results suggest that the use of CPAP before physical exercise increases exercise duration in children and adolescents with STRA.

BACKGROUND We describe the treatment strategy for a patient who was found to have a partial hydatidiform mole and coexisting fetus(PHMCF)during the second *** patient was a 38-year-old Chinese woman who had become pregnant following in vitro fertilization and embryo *** wanted to determine the safest therapeutic strategy to terminate the PHMCF during the second *** SUMMARY In this case,we present a patient who was found to have a PHMCF complicated with serious continuous vaginal bleeding and pre-eclampsia during the second *** careful evaluation,the pregnancy was considered to be unsustainable and was terminated via caesarean section(CS).An infant with weak vital signs and a partially cystic placenta measuring 110 mm×95 mm×35 mm were delivered by *** patient was discharged after 4 *** serum levels ofβ-human chorionic gonadotropin decreased to within a normal range 5 wk after the operation,and no evidence of persistent trophoblastic disease or lung metastases was noticed at the 6-mo *** CS termination of PHMCF during the second trimester may be a relatively safe therapeutic strategy.
