Screening for Bioactive Compounds from Mulberry Leaves Combining Bio-Affinity Ultrafiltration with α-Glucosidase and Pancreatic Lipase

With the growing prevalence of diabetes and obesity worldwide, the role of α-glucosidase and lipase inhibitors has gained increasing concentration. Mulberry, a functional medicinal and edible plant, has been reported extensively in alleviating both diabetes and obesity. However, there is still a lack of refined research on its active ingredients and mechanisms. In our study, the inhibitory activities of mulberry leaf against α-glucosidase and pancreatic lipase reached 191.37 ± 14.46 and 775 ± 68.60 μg/mL, respectively. Meanwhile, it also promoted glucose consumption in the HepG2 cells model. Subsequently,four α-glucosidase ligands and four pancreatic lipase ligands were identified through bio-affinity ultrafiltration combined with liquid-chromatography-mass spectrometry(UF-HPLC–MS/MS). In vitro inhibition assays confirmed the inhibitory potential of these representative ligands. Molecular docking suggested that these possible ligands could be inserted into the active pocket of the target enzyme through multiple intermolecular forces. Our findings indicated that the polyphenols in mulberry leaves may contribute significantly to their hypoglycemic and hypolipidemic properties. Furthermore, our study provides valuable insights for the screening of active ingredients in other medicinal and edible plants, which could help in the development of new therapies for diabetes and obesity.

HIFs in hypoxic regulation of the extracellular matrix:focus on little-known player HIF-3

Highly efficient labelling of extracellular vesicles for enhanced detection on a microfluidic platform

Developing precise extracellular vesicles(EVs)labelling techniques with minimal disturbance is of great importance to the follow-up EVs detection and ***,currently available methods such as using probes to conjugate phospholipids or membrane proteins have certain limitations due to EV steric hindrance,dye aggregation,***,we present a microfluidic platform to enhance EVs’labelling efficiency and improve their *** platform provides excellent sample throughput and high-efficiency EV labelling at lower label concentrations with an optimized flowing *** cytometry analysis(FCM)and cellular uptake results show that EV labelling by utilizing this platform possesses the merits of a higher labelling efficiency with 64.1%relative improvement than conventional co-incubation method and a lower background ***,this technique maintains EVs’size,morphology and biological *** the recipient cells uptake the EVs treated by the microfluidic platform,the spatial and temporal distribution of EVs in the cells are clearly *** results demonstrate that our method holds great potential in efficient labelling of EVs,which is essential to subsequent EV quantification and analysis.

Peptide-drug conjugates have achieved considerable development and application as a novel strategy for targeted delivery of anticancer drugs. Bioactive peptides induced calcium deposition can irreversibly assist inhibition of tumors. However, active regulation of calcium level through signal transduction of bioactive substances has not been reported yet. In this study, novel neuropeptide-doxorubicin conjugates(NP-DOX) with lysosome-specific acid response were described for neuropeptide Y_1 receptor(Y_1R)-overexpressed triple-negative breast cancer. The delivery mechanism of NP-DOX was clarified that diverse pathways were involved, including intracellular and intercellular transport. Importantly, up-regulation of Y_1 R-mediated intracellular calcium level via second messenger inositol triphosphate was presented in NP-DOX treated MDA-MB-231 cells. In vivo antitumor efficacy demonstrated that NP-DOX showed less organ toxicity and enhanced tumor inhibition benefited from its controlled release and Y_1R-mediated calcium deposition, compared with free DOX. This bioconjugate is a proof-of-concept confirming that neuropeptide-mediated control of signaling responses in neuropeptide-drug conjugates enables great potential for further applications in tumor chemotherapy.
