Site-specific N-glycosylation characterization and allergenicity analysis of globulin-1 S allele from wheat

N-glycans in many proteins are of great concern because of their strong association with food allergies. Triticum aestivum(bread wheat), a major food crop, is known as one of the “Big Eight” allergenic groups. However, little research has been done about N-glycans in wheat glycoproteins. In this study, a soluble wheat glycoprotein was purified from wheat and further identified as globulin-1 S allele(GSA). The wheat GSA displayed significant IgE-binding activity. Moreover, one N-glycosylation site and 6 kinds of N-glycans were identified by mass spectrometry, including 3 high mannose types and 3 complex types. Furthermore, the IgE-binding activity of wheat GSA is proved to be reduced by the removal of N-glycan, thermal treatment(temperatures > 80 ℃), and strong acidic treatment(pH 3.0). These findings would provide a better understanding of the effects of N-glycosylation, thermal treatment, and acidic treatment on the molecular characteristics of GSA, and further provide new insights into the development of hypoallergenic wheat products.

Seaweed Complex Preparation （SCP） is a clinical traditional Chinese medicine preparation which is composed of seven traditional Chinese herbs, and it has been used for treatment of lung cancer, liver cancer and digestive cancer. However, little infor- mation is available about the pharmacodynamic basis. The antitumor, immunomodulatory and free radical scavenging effects of SCP were evaluated in this study. Transplanted tumor in vivo method was used to determine the antitumor effect. The effects on spleno- cyte proliferation and phagocytosis of macrophages in tumor-bearing mice were measured by the MTT method and the phagocytizing cock red blood cell （CRBC） method respectively. The scavenging activities of SCP on DPPH and hydroxyl radicals in vitro were investigated. It was found that the medium-dose and high-dose of SCP could significantly inhibit the growth of transplanted hepatic tumor of murine hepatocarcinoma cell line H22, and promote proliferation of splenocytes and phagocytosis of macrophages. SCP possessed noticeable scavenging activities on DPPH and hydroxyl radicals. The antiturnor effects of SCP might be achieved by im- proving immune system and scavenging free radicals, which is in accordance with the viewpoint of traditional Chinese medicine in promoting the body resistance and eliminating pathogenic factors for cancer treatment.

Pyropia haitanensis polysaccharide(LP)have been found for having many excellent functions such as *** Caenorhabditis elegans models,we evaluated the anti-aging activity of LP by observing the lifespan,reproduction,pharyngeal pumping,stress response,quantitative fluorescence of polyglutamic acid,and nuclear localization of DAF-16 of *** results reveal that LP could extend the adult lifespan of wild-type and polyQ nematodes,indicating a connection of its anti-aging benefit with the toxicity-suppressing *** number of polyglutamic acid aggregates in high concentration groups decreased by 24.39%(P<0.05)to the *** high-dose group strongly induced DAF-16 nuclear translocation over intermediate and cytosolic localizations compared with the control(P<0.001).Therefore,we believe that LP could extend the lifespan and reduce the protein aggregation in *** through nuclear DAF-16∷GFP expression.

Purification and Refolding of a Novel β-Agarase from Inclusion Body of E. coli

β-agarase AgaB appears to represent a new family of glycoside hydrolase; it is structurally and functionally different from other known agarases. In the present study, AgaB was expressed with a temperature-inducible expression system in E. coli BL21 (DE3) as a fusion protein bearing a C-terminal hexahistidine tag. The protein existed mainly in the form of inclusion body. After being washed and solubilized, AgaB in inclusion body was denatured and purified to electrophoretic purity by immobilized metal affinity chromatography. The purified AgaB was then refolded using a simple pulse dilution method, and the refolded AgaB showed a high specific hydrolysis activity of about 1600 units /mg protein. Forty milligrams of refolded pure protein were obtained from 1L of culture.

To improve the light environment and welfare of the turtle cultured indoors,the effects of lighting mode on growth performance,cortisol level,and oxidative stress of juvenile Chinese three-keeled pond turtle,Chinemys reevesii,were investigated in this *** experimental turtles with an initial weight of 5.61±0.09 g were reared in tanks under four different lighting modes:three groups with light(lighting the basking area and water area,LBW;lighting the water area only,LW;lighting the basking area only,LB)and control group(no light,NL).The experiment was conducted for more than six months,with each group having three *** 203 d of the experiment,the turtle in the LW group exhibited higher weight gain rate(WGR)and a specific growth rate(SGR,%/d)compared to other ***,results showed that the final body weight of the turtle exposed to LW was higher than that exposed to other *** the physiological level,serum cortisol level in turtles exposed to LW was significantly lower than that in other *** oxidative stress,the level of catalase(CAT)in turtles exposed to LW and LB was significantly lower than that exposed to LBW and *** malonaldehyde(MDA)activity in turtles exposed to LW was significantly lower than other *** on the growth performance and health status,it is suggested that lighting the water area only is the optimal lighting mode for the juvenile threekeeled pond turtle cultured indoors.
