Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

Influenza is an acute viral respiratory infection that has caused high morbidity and mortality *** A virus(IAV)has been found to activate multiple programmed cell death pathways,including *** is a novel form of programmed cell death in which the accumulation of intracellular iron promotes lipid peroxidation,leading to cell ***,little is known about how influenza viruses induce ferroptosis in the host *** this study,based on network pharmacology,we predicted the mechanism of action of Maxing Shigan decoction(MXSGD)in IAV-induced ferroptosis,and found that this process was related to biological processes,cellular components,molecular function and multiple signaling pathways,where the hypoxia inducible factor-1(HIF-1)signaling pathway plays a significant ***,we constructed the mouse lung epithelial(MLE-12)cell model by IAV-infected in vitro cell experiments,and revealed that IAV infection induced cellular ferroptosis that was characterized by mitochondrial damage,increased reactive oxygen species(ROS)release,increased total iron and iron ion contents,decreased expression of ferroptosis marker gene recombinant glutathione peroxidase 4(GPX4),increased expression of acyl-CoA synthetase long chain family member 4(ACSL4),and enhanced activation of hypoxia inducible factor-1α(HIF-1α),induced nitric oxide synthase(iNOS)and vascular endothelial growth factor(VEGF)in the HIF-1 signaling *** with MXSGD effectively reduced intracellular viral load,while reducing ROS,total iron and ferrous ion contents,repairing mitochondrial results and inhibiting the expression of cellular ferroptosis and the HIF-1 signaling ***,based on animal experiments,it was found that MXSGD effectively alleviated pulmonary congestion,edema and inflammation in IAV-infected mice,and inhibited the expression of ferroptosis-related protein and the HIF-1 signaling pathway in lung tissues.

We investigate the plastic deformation and constitutive behaviour of bulk metallic glasses （BMGs）. A dimensionless Deborah number DeiD = tr/ti is proposed to characterize the rate effect in BMGs, where tr is the structural relaxing characteristic time of BMGs under shear load, ti is the macroscopic imposed characteristic time of applied stress or the characteristic time of macroscopic deformation. The results demonstrate that the modified free volume model can characterize the strain rate effect in BMGs effectively.

The cell membrane is a critical barrier for cellular homeostasis,integral to signaling and intercellular communication,and vital for understanding cellular functions and disease *** its microenvironment is crucial for uncovering the molecular basis of physiological and pathological processes associated with the cell membrane,driving the development of bioanalytical toolkits capable of dynamically monitoring the cell surface *** the continuous advancement of functional nucleic acids and dynamic DNA nanotechnology,DNA nanodevices with controllable nanosized geometry,specific molecular recognition,and selective membrane-localization properties offer a versatile platform for probing the cell membrane *** this review,we summarize the current biosensing and membrane-anchoring mechanisms of DNA nanodevices and highlight their use in studying key cell membrane events,including membrane lipid dynamics,transmembrane transport,receptor dimerization,and signal ***,we discuss the challenges and potential future applications of DNA nanodevices in advancing cell membrane biology research and biomedical applications.

Hollow glass microspheres(HGMs)have great potential in building energy-saving and industrial *** TiO2-modified HGMs were prepared by a sol-gel method in acetic acid-ethanol *** electron microscopy,X-ray diffraction,zeta-potential measurements,nitrogen-sorption measurements,and Fourier-transform infrared and ultraviolet-visible-near-infrared diffuse reflectance spectroscopies showed that the alkali modification of the HGMs greatly influenced the loading and microstructure of the TiO2 *** TiO2 loading could be accurately controlled by ethanol addition and the TiO2 coating time.A mechanism for the TiO2 coating of the HGM surface is *** synergistic action of hydrogen bonding and electrostatic forces resulted in close contact between the HGMs and TiO2 sol at pH *** effects of different TiO2 loading rates on the reflective and thermal insulation properties were *** near-infrared reflectance of 15.9%TiO2 coated on HGMs was 96.27%,and the inner surface temperature of the composite pigment coated on aluminum board was reduced by 22.4℃.The TiO2/HGM composite pigments exhibited excellent solar reflective and thermal insulation properties,so have potential in the construction of exterior walls and roofs.

Commercial Cu and Al current collectors for lithium-ion batteries(LIBs)possess high electrical conductivity,suitable chemical and electrochemical ***,the relatively flat surface of traditional current collectors causes weak bonding strength and poor electrochemical contact between current collectors and electrode materials,resulting in potential detachment of active materials and rapid capacity degradation during extended ***,we report an ultrafast femtosecond laser strategy to manufacture hierarchical micro/nanostructures on commercial Al and Cu foils as current collectors for high-performance *** hierarchically micro/nanostructured current collectors(HMNCCs)with high surface area and roughness offer strong adhesion to active materials,fast electronic delivery of entire electrodes,significantly improving reversible capacities and cyclic stability of HMNCCs based ***,LiNi_(0.5)Co_(0.2)Mn_(0.3)O_(2)(NCM523)cathode with Al HMNCC generated a high reversible capacity after 200 cycles(25%higher than that of cathode with Al CC).Besides,graphite anode with Cu HMNCC also maintained prominent reversible capacity even after 600 ***,the full cell assembled by graphite anode with Cu HMNCC and NCM523 cathode with Al HMNCC achieved high reversible capacity and remarkable cycling stability under industrial-grade mass *** study provides promising candidate for achieving high-performance LIBs current collectors.
