＜正＞Flutamide,a non - steroidal androgen receptor antagonist,has been used as an adjunct after trauma -hemorrhage for restoring immune and cardiovascular function and for decreasing the mortality from subsequent *** stroke resembles septic shock in many *** hypothesized that heat - induced multiple organ dysfunction syndrome（MODS） and lethality can be reduced by flutamide preconditioning and/or post - heat stroke *** major groups of mice were designated for experiments:a) mice treated with vehicle once 3 h before whole body heating（WBH）;b) mice treated with vehicle solution once daily for 3 consecutive days after WBH;c) mice treated with flutamide（50 mg/kg,S.C.） once 3 h before WBH;and d) mice treated with flutamide（50 mg/kg,S.C.） once daily for consecutive 3 days after *** WBH -treated animals were exposed to heat stress（ambient temperature 41.2℃for 1 h） and then allowed to recover at room temperature（24℃）.Mice that survived on day 4 of WBH were considered *** and biochemical parameters were monitored.2.5 h after *** evaluated the effect of flutamide in heatstroke mice and showed that flutamide significantly（i） attenuated hypothermia,（ii） reduced the numbers of the apoptotic cells in the hypothalamus,the spleen,the liver,and he kidney,（iii） attenuated plasma index of the toxic oxidizing radicals like nitric oxide metabolites and hydroxyl radicals,（iv） diminished plasma index of the organ injury like lactate dehydrogenase,（v） attenuated plasma systemic inflammation response molecules like tumor necrosis factor-α,and interleukin-6,（vi） reduced an index of infiltration of polymorphonuclear neutrophils in the lung like myeloperoxidase activity,and（vii） promoted the fractional survival to threefold compared with the vehicle - treated heatstroke ***,flutamide could be a novel agent for the treatment of heatstroke animals or patients in the early stage.

The discovery of Severe Acute Respiratory Syndrome-Coronavirus-2 (SARS-CoV-2) in Wuhan, hubei province, China, in December 2019 raised global health warnings. Quickly, in 2020, the virus crossed borders and infected individuals across the world, evolving into the COVID-19 pandemic. Notably, early signs of the virus’s existence were observed in various countries before the initial outbreak in Wuhan. As of 12th of April, the respiratory disease had infected over 762 million people worldwide, with over 6.8 million deaths recorded. This has led scientists to focus their efforts on understanding the virus to develop effective means to diagnose, treat, prevent, and control this pandemic. One of the areas of focus is the isolation of this virus, which plays a crucial role in understanding the viral dynamics in the laboratory. In this study, we report the isolation and detection of locally circulating SARS-CoV-2 in Kenya. The isolates were cultured on Vero Cercopithecus cell line (CCL-81) cells, RNA extraction was conducted from the supernatants, and reverse transcriptase-polymerase chain reaction (RT-PCR). Genome sequencing was done to profile the strains phylogenetically and identify novel and previously reported mutations. Vero CCL-81 cells were able to support the growth of SARS-CoV-2 in vitro, and mutations were detected from the two isolates sequenced (001 and 002). Genome sequencing revealed the circulation of two isolates that share a close relationship with the Benin isolate with the D614G common mutation identified along the S protein. These virus isolates will be expanded and made available to the Kenya Ministry of Health and other research institutions to advance SARS-CoV-2 research in Kenya and the region.

To the Editor:In the last decade,neoadjuvant chemotherapy(NAC)has become a well-accepted treatment option for breast cancer,although few detailed description of NAC in China has yet been reported.[1]A previous study found that among patients with clinically node-negative(cN0)breast cancer,97.7%(432/442)with breast pathologic complete response(bpCR)had ypN0(absence of metastases in the axillary lymph nodes);and 71.6%(882/1232)without bpCR achieved ypN0(P<0.001).As for human epidermal growth factor receptor 2(HER2)positive or triple-negative breast cancer(TNBC)that achieved bpCR.

Correlation of serum NGAL and OPG contents with the change of carotid plaques in diabetic patients

Objective:To investigate the correlation of serum NGAL and OPG contents with the change of carotid plaques in diabetic ***: A total of 99 patients with type 2 diabetes mellitus combined with carotid plaque who were treated in our hospitals between February 2016 and March 2018 were selected as group A and 86 patients with carotid plaque alone were selected as group B. The serum contents of NGAL OPG, inflammatory markers and lipid metabolism indexes were compared between the two groups, and Pearson test was used to evaluate the inner link of serum NGAL and OPG contents with carotid plaque properties in diabetic patients with carotid ***: Serum NGAL content of group A was higher than that of group B whereas OPG content was lower than that of group B;serum inflammatory markers MMP-9, IL-1β, IL-17 and CRP contents were higher than those of group B;serum lipid metabolism indexes ApoB and Lp-PLA2 contents were higher than those of group B whereas ApoAⅠcontent was lower than that of group B. Correlation analysis showed that serum NGAL and OPG contents of patients with diabetes mellitus combined with carotid plaque were directly correlated with the content of inflammatory markers and lipid metabolism ***: Serum NGAL content abnormally increases whereas OPG content abnormally decreases in patients with diabetes mellitus combined with carotid plaque, and they are directly correlated with the stability of carotid plaque.

Investigation and experimental validation of curcumin-related mechanisms against hepatocellular carcinoma based on network pharmacology

Growth of embryo and gene expression of nutrient transporters in the small intestine of the domestic pigeon(Columba livia)

The objective of this study was to investigate the relationship between gene expression of nutrient（amino acid, peptide, sodium and proton） transporters in the small intestine and embryonic growth in domestic pigeons（Columba livia）. One hundred and twenty-five fertilized eggs were randomly assigned into five groups and were incubated under optimal conditions（temperature of 38.1 °C and relative humidity of 55%）. Twenty embryos/birds from each group were sacrificed by cervical dislocation on embryonic day（E） 9, 11, 13, 15 and day of hatch（DOH）. The eggs, embryos（without yolk sac）, and organs（head, brain, heart, liver, lungs, kidney, gizzard, small intestine, legs, and thorax） were dissected, cleaned, and weighed. Small intestine samples were collected for RNA isolation. The m RNA abundance of intestinal nutrient transporters was evaluated by real-time reverse transcription-polymerase chain reaction（RT-PCR）. We classified these ten organs into four types according to the changes in relative weight during embryonic development. In addition, the gene expression of nutrient transporters was differentially regulated by embryonic day. The m RNA abundances of b^0,＋AT, EAAT3, y^＋LAT2, Pep T1, LAT4, NHE2, and NHE3 increased linearly with age, whereas m RNA abundances of CAT1, CAT2, LAT1, EAAT2, SNAT1, and SNAT2 were increased to higher levels on E9 or E11 and then decreased to lower levels until DOH. The results of correlation analysis showed that the gene expressions of b^0,＋AT, EAAT3, Pep T1, LAT4, NHE2, NHE3, and y^＋LAT2 had positive correlations with body weight（0.71〈correlation coefficient（CC）〈0.82, P〈0.0001）, while CAT1, CAT2, EAAT2, SNAT1, and SNAT2 had negative correlations with body weight（-0.86〈CC〈-0.64, P〈0.0001）. The gene expressions of b^0,＋AT, EAAT3, LAT4, Pep T1, NHE2, NHE3, and y^＋LAT2 showed positive correlations with intestinal weight（0.80〈CC〈0.91, P〈0.0001）, while CAT1, CAT2, and EAAT2 showed negative correlations with intestinal weight（-0.84〈CC〈-0.67, P〈0.0

Effects of Ligustrum robustum on gut microbes and obesity in rats

AIM: To investigate the anti-obesity and antibacterial effects of Ligustrum robustum(L. robustum) in vivoand in vitro and its possible mechanisms. METHODS: The effects of L. robustum aqueous extract(LR) on various gut bacteria in vitro were evaluated. The effects of LR on high-fat diet-fed(HFD) rats in vivo were also assessed. Culture methods,quantitative polymerase chain reaction,and terminalrestriction fragment length polymorphism were used to analyze the effects of LR on gut bacteria. Biochemical tests were also performed to detect the changes in obesity-related indicators after LR treatment. RESULTS: LR treatment lowered adipose weight and decreased Lee's index,blood glucose,total cholesterol,and lipid in the tested groups relative to control(P control,and the effects of L. robustum might be mediated by the regulation of gut bacteria.

Sequence Analyses on N Gene of Rabies Viruses Isolated from Guangdong Province

[ Objective] To find the dominant rabies virus strains that are epidemic in Guangdong Province and thus to better prevent and control rabies. [ Method] Canine brains and salivary glands collected from several regions of Guangdong Province were detected by direct imrnunofluorescence assay and nested RT-PCR. The isolates were further identified by intracranial inoculation in suckling mice. The N gene was amplified by RTPCR and the sequences were aligned with those of prevalent strains in Guangxi region and vaccine strains. [ Result] Two strains were isolated from specimens collected from Guangdong Province. Their N gene sequences had a high similarity of 97.9% -99.4% to those of Guangxi strains and also had a similarity of 87.7% -88.1% to those of vaccine strains of rabies virus. [ Conclusion] Apparently healthy dogs may carry rabies virus, and the prevalence of rabies virus shows obvious regional differences.
