Inhibition of the cGAS–STING pathway:contributing to the treatment of cerebral ischemia-reperfusion injury

The cGAS–STING pathway plays an important role in ischemia-reperfusion injury in the heart,liver,brain,and kidney,but its role and mechanisms in cerebral ischemia-reperfusion injury have not been systematically ***,we outline the components of the cGAS–STING pathway and then analyze its role in autophagy,ferroptosis,cellular pyroptosis,disequilibrium of calcium homeostasis,inflammatory responses,disruption of the blood–brain barrier,microglia transformation,and complement system activation following cerebral ischemia-reperfusion *** further analyze the value of cGAS–STING pathway inhibitors in the treatment of cerebral ischemia-reperfusion injury and conclude that the pathway can regulate cerebral ischemia-reperfusion injury through multiple *** of the cGAS–STING pathway may be helpful in the treatment of cerebral ischemia-reperfusion injury.

A matrix metalloproteinase-responsive hydrogel system controls angiogenic peptide release for repair of cerebral ischemia/reperfusion injury

Hydrogen sulfide reduces oxidative stress in Huntington's disease via Nrf2

The pathophysiology of Huntington's disease involves high levels of the neurotoxin quinolinic acid. Quinolinic acid accumulation results in oxidative stress, which leads to neurotoxicity. However, the molecular and cellular mechanisms by which quinolinic acid contributes to Huntington's disease pathology remain unknown. In this study, we established in vitro and in vivo models of Huntington's disease by administering quinolinic acid to the PC12 neuronal cell line and the striatum of mice, respectively. We observed a decrease in the levels of hydrogen sulfide in both PC12 cells and mouse serum, which was accompanied by down-regulation of cystathionine β-synthase, an enzyme responsible for hydrogen sulfide production. However, treatment with NaHS(a hydrogen sulfide donor) increased hydrogen sulfide levels in the neurons and in mouse serum, as well as cystathionine β-synthase expression in the neurons and the mouse striatum, while also improving oxidative imbalance and mitochondrial dysfunction in PC12 cells and the mouse striatum. These beneficial effects correlated with upregulation of nuclear factor erythroid 2-related factor 2 expression. Finally, treatment with the nuclear factor erythroid 2-related factor 2inhibitor ML385 reversed the beneficial impact of exogenous hydrogen sulfide on quinolinic acid-induced oxidative stress. Taken together, our findings show that hydrogen sulfide reduces oxidative stress in Huntington's disease by activating nuclear factor erythroid 2-related factor 2,suggesting that hydrogen sulfide is a novel neuroprotective drug candidate for treating patients with Huntington's disease.

Effects of Acupuncture at 7-9 am and 3-5 pm on Plasma Thromboxane and Prostaglandin in Patients with Ischemic Cerebrovascular Disease

Objective:To observe the effects of acupuncture at different times on plasma thromboxane(TXB2) and prostaglandin 6-Keto-PGF1a(6-K-P) in patients with ischemic cerebrovascular ***:Totally 90 patients were randomly divided into a group acupunctured at 7-9 am,a group acupunctured at 3-5 pm and a drug control group,with 30 cases in each *** contents of plasma TXB2 and 6-K-P from venous blood before treatment were compared with those 15 days after ***:The plasma TXB2 levels of the two acupuncture groups were obviously lower than those before treatment(P<0.05,P<0.01) ,but the 6-K-P levels of both the acupuncture groups were remarkably higher than those before treatment(P<0.05,P<0.01) . And the TXB2 level in the 3-5 pm acupuncture group was obviously lower than that in the 7-9 am acupuncture group(P<0.05) ,and the 6-K-P level of the former was obviously higher than that of the latter(P<0.05) .Conclusion:Acupuncture can promote functional recovery in patients with ischemic cerebrovascular disease and enhance their survival quality.

Deciphering the molecular mechanisms of Simiaowan in the treatment of hyperuricemia: in vivo and in silico approaches

Clinical effect of Jiutengzhuyu tablets on promoting blood circulation in women with oviducal obstruction

OBJECTIVE： To test the clinical curative effect of Jiutengzhuyu tablets, a patented Chinese vine-de- rived drug used for promoting blood circulation, on women with oviducal obstruction. METHODS Patients with tubal infertility were di- vided randomly into two groups： 58 patients in the treatment group and 57 in a control group. The treated patients took Jiutengzhuyu tablets orally for 25 days. The control group received an intrauter- ine infusion of 5 mg dexamethasone sodium phos- phate, 4000 units of chymotrypsin, 80 000 units of gentamicin sulfate dissolved in 20 mL of normal sa- line at I mL/min. After 3 months of treatment, the curative effect on tubal patency was assessed and a l-year follow-up visit was used to document any pregnancies. RESULTS The total effective rate was 78% in the treatment group and 32% in the control group with a statistically significant difference （x2=24.57, P〈0.01）.CONCLUSION： Jiutengzhuyu was effective in treat- ing infertility caused by tubal infertility with a Traditional Chinese Medicine diagnosis of blood stasis.
