the mRNA expression patterns of tumor necrosis factor-a and TNFR-I in some vital organs after thermal injury

AIM: To investigate changes of tumor necrosis factor-α (TNFα) and TNFR-Ⅰ expression in vital organs and their significance in the pathogenesis of multiple organ damage associated with endogenous endotoxin following major ***: Wistar rats subjected to a 35 % full-thickness scald injury were sacrificed at 12 h, 24 h, 48 h, and 72 h postburn, respectively. Meanwhile, eight rats were taken as normal controls. Tissue samples from liver, spleen, kidney,lung and intestine were collected to assay tissue endotoxin levels and measure TNF-α and TNFR-Ⅰ expression, In addition, blood samples were obtained for the determination of organ function ***: Endotoxin levels in liver, spleen and lung increased markedly after thermal injury, with the highest level in liver. the gene expression of TNF-α in liver, lung and kidney was up-regulated after thermal injury, while the TNFR-Ⅰ mRNA expression in liver, lung, kidney and intestine was shown decreased throughout the observation period. thus, the mRNA expression ratio of TNF-α to TNFRⅠ was significantly increased postburn, particularly in pulmonary tissue (67-fold). In addition, the significant correlations between the expression of TNFR-Ⅰ or the expression ratio of TNF-α/TNFR mRNA in liver tissue and serum aspartate aminotransferase levels were noted (P ＜0.05-0.01). Similar results were also obtained between pulmonary TNF-α mRNA expression and myeloperoxidase activities (P＜0.01), whereas there was a highly negative correlation between levels of renal TNFR-Ⅰ mRNA expression and serum ***: Burn injury could result in the translocation of gut-derived endotoxin that was mainly distributed in the liver, spleen and lung. the translocated endotoxin then made the expression of TNF-α and TNFR-Ⅰ mRNA up-regulated and down-regulated respectively in various organs, which might be involved in the pathogenesis of multiple organ damage following burns.

Role of basic fibroblast growth factor combined with mesenchymal stem cells in promoting wound healing

To investigate the role of basic fibroblast growth factor(bFGF) in inducing the differentiation of mesenchymal stem cells (MSCs) to skin tissues in vivo and discuss its effect on skin wound *** MSCs were isolated from porcine marrow,purified and cultured with 5-bromodeoxyuridine (5-BrdU) as a label in ***-eight skin wounds on the six procines were divided into bFGF+MSCs(Group A),MSCs (Group B),bFGF (Group C) and control group randomly and treated *** closing index (WCI),cell morphology and histological changes in the defected regions were evaluated 3,7,14,21 and 42 days after *** endothelium cells derived from MSCs in the granulation tissues were assayed by immuno-staining of 5-BrdU and Factor Ⅷ in the same *** Healing speed of defected regions in Group A was faster than that of the other *** scores of histopathology on the specimens of Group A were also higher than that of the other *** dual-positive stained cells by BrdU and Factor Ⅷ in Group A were more than those in Group *** bFGF can induce MSCs differentiating into vessel endothelium cell and hence promote the repair of skin wounds.9 refs,3 figs,5 tabs.

Hepatic angiomyolipoma-misdiagnosis as hepatocellular carcinoma:A report of 14 cases

Angiomyolipoma (AML) is a rare benign mesenchymal tumor of the liver, composed of a varying heterogeneous mixture of three tissue components: blood vessels, smooth muscle, and adipose cells. It has recently been proposed that the perivascular epithelial cell (PEC) is the common progenitor[1，2] Since its first description by Ishak in 1976[3], there have been more than 100 cases reported in the English literature[4-6]. With the advance of radiological techniques, many more tumors are being diagnosed by the means. But radiological findings of AML may only be suggestive of the lesion; its definitive diagnosis requires histological confirmation[9-19]. Some authors regard renal and hepatic AMLs, pulmonary and soft tissue lymphangiomyomatosis[2], pulmonary and pancreatic clear cell “sugar” tumor, and cardiac rhabdomyoma as closely related groups of tumors, based on their morphologic overlap and common immunoreactivity for HMB-45[l]. they show different microscopic appearances, however, according to their organ of origin. the goals of this study were to highlight more subtle morphology and to gain possible insights into the differential diagnosis that could provide important information about this disease.
