Proteomic Analysis of Cucumber Defense Rresponses Induced by Propamocarb

Propamocarb is an agricultural chemical that has been widely used to protect cucumber plants from downy mildew. To understand the mechanisms of cucumber defense responses to propamocarb, we investigated the physiological and proteomic responses of the cucumber line D0351 with propamocarb application. We found that after treatment with propamocarb, the activities of detoxifying enzymes （glutathione reductase, GR; glutathione S-tramsferase, GST） and soluble sugar content of cucumber fruit were signiifcantly increased, but malonaldehyde （MDA） content was signiifcantly reduced. To identify components of propamocarb responsive signaling, we compared the high resolution two-dimensional gel electrophoresis （2-DE） protein proifles of control and propamocarb-treated fruits, and identiifed 18 differentially expressed （13 up-regulated and 5 down-regulated） proteins induced by propamocarb which were determined by matrix-assisted laser desorption/ionization time-of-lfight mass spectrometry （MALDI-TOF-MS）. The majority of the proteins had functions related to detoxication, energy and transport, protein biosynthesis, regulating reactions and defending against stresses. A real-time quantitative reverse transcriptional-polymerase chain reaction （qRT-PCR） was used to compare transcript and protein accumulation patterns for 18 candidate proteins, and the expression of 14 was consistent at both transcript and protein levels. The responses of cucumber proteome to propamocarb seemed complex; the identified proteins may play an important role in regulating adaptation activities following exposure to propamocarb. Data presented herein may shed light on understanding cucumber fruit defense responses under propamocarb treatment.

Chinese leek（Allium tuberosum Rottler ex Sprengel） is a common vegetable in China. In our previous study, Chinese leek in rotation was found to have significant antifungal and nematicidal activity. This study＇s aim was to investigate the potential antifungal and nematicidal activity associated with rhizosphere or endophytic microbes of Chinese leek. Thus, a total of 79 261 high-quality sequences were obtained from Chinese leek rhizosphere soil, leaf and root samples. In the rhizosphere soil, the bacterial community comprised five dominant phyla： Proteobacteria（37.85%）, Acidobacteria（10.99%）, Bacteroidetes（8.24%）, Cyanobacteria（7.79%） and Planctomycetes（7.1%）. The leaf and root bacterial communities comprised two dominant phyla： Cyanobacteria（83.42% in leaf and 75.44% in root） and Proteobacteria（14.75% in leaf and 21.04% in root）. Microbial diversity, richness and evenness in the rhizosphere soil bacterial community were higher than that in the endophytic bacterial communities. The rhizosphere bacterial community was significantly different from the endophytic bacterial communities. The endophytic bacterial communities from the leaf and the root were slightly, but not significantly different from each other. This study＇s findings would contribute to the isolation and identification of nematicidal and antifungal bacterial communities in Chinese leek.

Either arbuscular mycorrhizal fungi （AMF） or polyamines （PAs） may change root system architecture （RSA） of plants, whereas the interaction of AMF and PAs on RSA remains unclear. In the present study, we studied the interaction between AMF （Paraglomus occultum） and exogenous PAs, including putrescine （Put）, spermidine （Spd） and spermine （Spin） on mycorrhizal development of different parts of root system, plant growth, RSA and carbohydrate concentrations of 6-m-old citrus （Citrus tangerine Hort. ex Tanaka） seedlings. After 14 wk of PAs application, PA-treated mycorrhizal seedlings exhibited better mycorrhizal colonization and numbers of vesicles, arbuscules, and entry points, and the best mycorrhizal status of taproot, first-, second-, and third-order lateral roots was respectively found in mycorrhizal seedlings supplied with Put, Spd and Spm, suggesting that PAs might act as a regulated factor of mycorrhizal development through transformation of root sucrose more into glucose for sustaining mycorrhizal development. AMF usually notably increases RSA traits （taproot length, total length, average diameter, projected area, surface area, volume, and number of first-, second-, and third-order lateral roots） of only PA-treated seedlings. Among the three PA species, greater positive effects on RSA change and plant biomass increment of the seedlings generally rank as Spd〉Spm〉Put, irrespective of whether or not AMF colonization. PAs significantly changed the RSA traits in mycorrhizal but not in non-mycorrhizal seedlings. It suggests that the application of PAs （especially Spd） to AMF plants would optimize RSA of citrus seedlings, thus increasing plant growth （shoot and root dry weight）.

ζ-Carotene desaturase(ZDS)is an important enzyme in carotenoid ***,the Brassica oleracea *** ZDS(Boa ZDS)gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction(RT-PCR)and deposited in Gen Bank(accession number KY662297).The Boa ZDS gene contains an open reading frame of 1 686 bp that encodes a 561-amino acid *** analysis indicates that the ZDS protein is apparently conserved during plant evolution and is most closely related to *** *** and *** promoter sequence of the Boa ZDS gene was predicted to harbor several cis-acting elements that are related to light and phytohormone *** RT-PCR analysis showed that Boa ZDS expression varied among different developmental stages and *** ZDS expression remained stable during germination and seedling stages and rapidly increased at the mature leaf *** leaves showed the highest ZDS expression levels compared to the other *** expression decreased in all flower tissues during *** fused protein of Boa ZDS was obtained by prokaryotic *** expression of Boa ZDS in Escherichia coli confirmed that Boa ZDS encodes a functionalζ-carotene desaturase that increases β-carotene accumulation in *** cells harboring a β-carotene-producing *** findings of the present study provide a molecular basis for the elucidation of ZDS gene function in Chinese kale.

Effects of transgenic Bt rice on non-target pests following pesticide applications plays an important role in evaluating the ecological safety of transgenic rice. Changes of the life history parameters of laboratory population of Nilaparvata lugens feeding on transgenic Bt rice T2A-1 and its parental line cv. MH63 as the control, and the contents of oxalic acid and soluble sugar following three pesticides （triazophos, jinggangmycin and chlorantraniliprole） treatments were investigated in this paper. Results showed that the population parameters of N. lugens and the physiological and biochemical parameters of rice did not differ significantly between T2A-1 and MH63 without pesticide application. But, the emergence rate （ER）, the nymphal number of next generation （NN）, and the index of population trend （IP） on T2A-1 treated with 10 mg L^-1 of triazophos were significantly higher than those on MH63, respectively, while the hatchability （HB） on T2A-1 treated with the three pesticides were obviously lower than those on MH 63. Furthermore, the content of oxalic acid in MH63 treated with 20 mg L-1 triazophos and, 40 and 80 mg L-1 chlorantraniliprole was significantly higher than that in T2A-1, while the content of soluble sugar in T2A-1 treated with 400 mg L-1 jinggangmycin was significantly higher than that in MH63. The results above indicated that triazophos in a low dose could cause a more favor of T2A-1 to population growth of N. lugens compared with MH 63.

The flower stalk is the product organ of flowering Chinese cabbage(Brassica campestris *** *** Tsen et Lee),which is cultivated extensively in South *** stalk formation and development,including bolting and flowering,determine the yield of flowering Chinese cabbage;however,the bolting and flowering mechanisms remain to be *** elucidate these processes,we studied the effects of low-temperature and gibberellin(GA)treatments,and their interaction,on stem elongation,bolting time,flowering time,hormone content,and cell morphology in stem of flowering Chinese *** results showed that both cold and GA treatments accelerated bolting time,stem elongation,and flowering ***,cold and GA cotreated plants displayed additive positive *** addition,cold treatments increased the GA,indole-3-acetic acid,and cytokinin contents and altered cell size in the shoot apices of flowering Chinese *** with uniconazole,a GA synthesis inhibitor,strongly delayed bolting time,stem elongation,and flowering time,whereas GA,but not cold treatment,rescued this inhibition,indicating that low temperature accelerates bolting and flowering not only through inducing GA in the shoot apices,but also other *** results provide a theoretical basis for further dissecting the regulatory mechanism of bolting and flowering in flowering Chinese cabbage.

The green miridbug,Cyrtorhinus lividipennis,is an important predator of the rice brown planthopper,Nilaparvata *** this study,the selective toxicity of seven commercial biopesticides for *** was examined under laboratory conditions; abamectin was the most selective to ***,followed by matrine and ***,rotenone,Bacillus thuringiensis and Beauveria bassiana showed less selectivity for ***,matrine,abamectin and azadirachtin were selected for sublethal assessments with respect to *** due to their high toxicities to *** treated with sublethal concentrations(LC10 and LC20) of the three biopesticides could distinguish volatiles released from healthy and ***-infested plants indicating that the biopesticides tested did not affect the foraging ability of surviving *** decreased the consumption capability of *** when the densities of *** were 20,30,40 and 50 insects per *** concentration treatment did not impact the pre-oviposition period or egg hatchability of ***,the fecundity of *** exposed to azadirachtin and abamectin increased by 27–41% compared to the untreated *** summary,abamectin or matrine together with *** could be considered an effective,sustainable pest management strategy for rice.

Lipoxygenases（LOXs） are a group of non-heme,iron-containing enzymes and extensively involved in plant growth and development,ripening and senescence,stress responses,biosynthesis of regulatory molecules and defense *** our previous study,18 LOXs in melon genome were screened and identified,and five 13-LOX genes（CmLOX08,CmLOX10,CmLOX12,CmLOX13 and CmLOX18） were predicted to locate in *** analysis result showed that the five genes have high homology with jasmonic acid（JA） biosynthesis-related LOXs from other *** addition,promoter analysis revealed that motifs of the five genes participate in gene expression regulated by hormones and ***,we analyzed the expressions of the five genes and LOX activity in leaves of four-leaf stage seedlings of oriental melon cultivar Yumeiren under abiotic stress：wounding,cold,high temperature and hydrogen peroxide（H_2O_2）,and signal molecule treatments：methyl jasmonate（MeJA）,abscisic acid（ABA） and salicylic acid（SA）.Real time qPCR revealed that wounding and H_2O_2 induced the expressions of all the five *** CmLOX08 was induced by cold while only CmLOX13 was suppressed by high *** induced the expressions of CmLOXIO and CmLOX12 while inhibited CmLOX13 and *** increased the 3 genes expressions except CmLOX08 and CmLOX13,whereas SA decreased the effect,apart from *** the abiotic stresses and signal molecules treatments increased the LOX activity in leaves of oriental *** summary,the results suggest that the five genes have diverse functions in abiotic stress and hormone responses,and might participate in defense *** data generated in this study will be helpful in subcellular localization and transgenic experiment to understand their precise roles in plant defense response.

Comparative Transcriptome Analysis of Actinidia arguta Fruits Reveals the Involvement of Various Transcription Factors in Ripening

The fruit of Actinidia arguta is typically *** ripening,the fruits produce a large amount of *** the molecular basis of climacteric fruit ripening has been extensively studied,some aspects such as its transcriptional regulation remain ***,we compared the transcriptomes of *** fruits collected at commercial harvest(sample d0)and at 6 days after harvest(sample d6)using RNA-seq.A total of 3 659 differentially expressed genes(DEGs)between samples d0 and d6 were detected,of which 2 983 and 1 104 could be functionally annotated with Gene Ontology(GO)and Clusters of Orthologous Groups(COG)*** involved in ethylene biosynthesis and signal transduction were identified,including ACO(c79627),ERF061(c105131),and ERF6(c99193).Transcription factors such as ERF,b HLH,NAC,MADS,and MYB were also differentially expressed between samples d0 and *** DEGs were subjected to further analysis using quantitative reverse transcription-polymerase chain reaction(q RT-PCR),and the results coincided with those of *** data revealed additional transcription factors that are involved in the regulation of fruit *** results provide useful information for future research on the transcriptional regulation of fruit ripening.
