CGG repeat expansions in LOC642361/NUTM2B-AS1 have recently been identified as a cause of oculopharyngeal myopathy with ***,since only three patients from a single family were reported,it remains unknown whether their clinicopathological features are typical for CGG repeat expansions in LOC642361/***,using repeat-primed-polymerase chain reaction and long-read sequencing,we identify 12 individuals from 3 unrelated families with CGG repeat expansions in LOC642361/NUTM2B-AS1,typically presenting with oculopharyngodistal *** CGG repeat expansions range from 161 to 669 repeat *** of the patients present with ptosis,restricted eye movements,dysphagia,dysarthria,and diffuse limb muscle *** one patient shows T2-weighted hyperintensity in the cerebellar white matter surrounding the deep cerebellar nuclei on brain magnetic resonance *** biopsies from three patients show a myopathic pattern and rimmed *** of muscle biopsies suggest that CGG repeat expansions in LOC642361/NUTM2B-AS1 may deleteriously affect aggrephagic capacity,suggesting that RNA toxicity and mitochondrial dysfunction may contribute to *** study thus expands the phenotypic spectrum for the CGG repeat expansion of LOC642361/NUTM2B-AS1 and indicates that this genetic variant typically manifests as oculopharyngodistal myopathy with chronic myopathic changes with rimmed vacuoles and filamentous intranuclear inclusions in muscle fibers.

Effect of lime application on microbial community in acidic tea orchard soils in comparison with those in wasteland and forest soils

Lime application is a conventional technology to control acidification in tea orchard soils. We investigated the effect of lime application on soil microbial community diversity in the soils of three tea orchards, wasteland and forest. The BIOLOG data showed that both the average well color development of all carbon sources and the functional diversity index increased with the liming rate in the tea orchards and the forest, but decreased in the wasteland. The phospholipid fatty acid （PLFA） analysis showed that the structural diversity index of soil microbial community increased with the liming rate in all the tea orchards, the wasteland and the forest. Lime application also increased the soil-bacterial PLFA content in all the soils. Soil fungal and actinomycete PLFAs in the tea orchards showed an increasing trend from 0 to 3.2 g CaCO 3 /kg application and then a decreasing trend from 3.2 to 6.4 g CaCO 3 /kg application. The principal component analysis of BIOLOG and PLFA data suggested that lime application had a significant effect on soil microbial community structure, and land use had a greater effect on soil microbial community structure compared to lime application.

Nitrification potentials of Chinese tea orchard soils and their adjacent wasteland and forest soils

To investigate the nitrifying activities of different soil types, soil samples collected from 8-, 50- and 90-year old tea orchards, the adjacent wasteland, and 90-year old forest were measured for their nitrification potentials using the conventional soil incubation and the liquid incubation method. Among different soil types, the nitrification potential of soil in tea orchards was higher than that of wasteland and forest soils. The slurry shaken liquid incubation method was confirmed to be more accurate and have reliable results than the soil incubation. Interestingly, experimental result revealed that the generally applied pH value of 7.2 for the liquid media was not the optimal pH for these acid soils with a strong buffer capacity. This suggested that tea orchard soils may have nitrifiers requiring pHneutral condition for the best activity. Our data also showed that treatment with the commonly used nitrogen fertilizer urea significantly improved nitrification potential of the soils; such enhancement effect was stronger on all of three tea orchard soils than on wasteland and forest soils, and also stronger on the younger （8- and 50-year old） tea orchard soils than on the older one （90-year old）.

Soil Microbial Community Structure in Diverse Land Use Systems:A Comparative Study Using Biolog,DGGE,and PLFA Analyses

Biolog, 16S rRNA gene denaturing gradient gel electrophoresis （DGGE）, and phospholipid fatty acid （PLFA） analyses were used to assess soil microbial community characteristics in a chronosequence of tea garden systems （8-, 50-, and 90- year-old tea gardens）, an adjacent wasteland, and a 90-year-old forest. Biolog analysis showed that the average well color development （AWCD） of all carbon sources and the functional diversity based on the Shannon index decreased （P 〈 0.05） in the following order： wasteland 〉 forest 〉 tea garden. For the DCCE analysis, the genetic diversity based on the Shannon index was significantly lower in the tea garden soils than in the wasteland. However, compared to the 90-year-old forest, the tea garden soils showed significantly higher genetic diversity. PLFA analysis showed that the ratio of Gram positive bacteria to Cram negative bacteria was significantly higher in the tea garden soils than in the wasteland, and the highest value was found in the 90-year-old forest. Both the fungal PLFA and the ratio of fungi to bacteria were significantly higher in the three tea garden soils than in the wasteland and forest, indicating that fungal PLFA was significantly affected by land-use change. Based on cluster analysis of the soil microbial community structure, all three analytical methods showed that land-use change had a greater effect on soil microbial community structure than tea garden age.

Interaction of Human Genes WT1 and CML28 in Leukemic Cells

The molecular pathogenesis of leukemia is poorly understood. Earlier studies have shown both Wilms＇ tumor 1 suppressor gene （WT1） and CML28 abnormally expressed in malignant diseases of the hematopoietic system and WT1 played an important role in leukemogenesis. However, the rela- tionship between molecular CML28 and WT1 has not been reported. Here we described the use of small interfering RNA （siRNA） against WT1 and CML28 in leukemic cell line K562 to examine the interac- tion between CML28 and WT1. WT1 and CML28 gene expression in transfected K562 cells was de- tected by using RQ-PCR and Western blotting. K562 cells transfected with WTI-siRNA could greatly decrease both mRNA and protein expression levels of WT1 and CML28. In contrast, CML28-siRNA did not exert effect on WT1. Further, subcellular co-localization assay showed that the two proteins could co-localize in the cytoplasm of K562 cells, but WT1/CML28 complexes were not detected by us- ing immunoprecipitation. It was suggested that there exists the relationship between CML28 and WT1. CML28 may be a downstream target molecule of WT1 and regulated by WT1, which will provide im- portant clues for further study on the role of CML28 and WT1 in leukemic cells.

In this study,maize stalk tegument separated from the maize pith was crushed to obtain the *** cross-linking maize starch adhesives considering four main factors(water content,gelatinization temperature,NaOH as gelatinization agent and Na2B4O7·10H2O as cross-linking agent)with three levels were prepared based on an orthogonal test scheme L9(34)in order to increase the water-resisting property and the bonding strength of the common maize starch *** bonding properties of maize starch adhesives were characterized using shearing strength under compression *** models of fiber reinforced composites were established according to the microstructure analysis of the four species of insects’elytra including Protaetia orentalis,Copris ochus Motschulsky,Anoplophora chinensis and Cytister bengalensis Aube,which will provide the biomimetic models for the biomimetic laminated *** maize stalk fiber biomimetic laminated boards were prepared based on the structural models of the elytra *** flexural strength and flexural elastic modulus of the biomimetic boards were *** results showed that the flexural strengths of the single layer jute fiber,-reinforced maize stalk fiber boards and the dual layer jute fiber reinforced maize stalk fiber boards are higher than those of the common maize stalk fiber boards and the other three groups of jute fiber hybrid reinforced stalk fiber boards because of the biomimetic laminated design.
