Genetic and pathogenic characterization of new infectious bronchitis virus strains in the GVI-1 and GI-19 lineages isolated in central China

Avian infectious bronchitis(IB)is a highly contagious infectious disease caused by infectious bronchitis virus(IBV),which is prevalent in many countries worldwide and causes serious harm to the poultry *** present,many commercial IBV vaccines have been used for the prevention and control of IB;however,IB outbreaks occur *** this study,two new strains of IBV,SX/2106 and SX/2204,were isolated from two flocks which were immunized with IBV H120 vaccine in central *** and recombination analysis indicated that SX/2106,which was clustered into the GI-19 lineage,may be derived from recombination events of the GI-19 and GI-7 strains and the LDT3-A *** analysis showed that SX/2204 belongs to the GVI-1 lineage,which may have originated from the recombination of the GI-13 and GVI-1 strains and the H120 *** virus cross-neutralization test showed that the antigenicity of SX/2106 and SX/2204 was different from *** experiments found that both SX/2106 and SX/2204 could replicate effectively in the lungs and kidneys of chickens and cause disease and death,and H120 immunization could not provide effective protection against the two IBV *** is noteworthy that the pathogenicity of SX/2204 has significantly increased compared to the GVI-1 strains isolated previously,with a mortality rate up to 60%.Considering the continuous mutation and recombination of the IBV genome to produce new variant strains,it is important to continuously monitor epidemic strains and develop new vaccines for the prevention and control of IBV epidemics.

Escherichia coli has become one of the most important causes of calf *** aim of this study is to determine the patterns of antimicrobial resistance of *** isolates from six cattle farms and to identify prominent resistance genes and virulence genes among the strains isolated from the diarrhea of *** susceptibility tests were performed using the disk diffusion method,and PCR was used to detect resistance and virulence *** prevalence of multidrug resistant(MDR)*** was 77.8%in dairy cattle and 63.6%in beef *** were high resistance rates to penicillin(100%,100%)and ampicillin(96.3%,86.4%)in *** from dairy cattle and beef ***,resistance rate to antimicrobials and distribution of resistance genes in£coli isolated from dairy cattle were higher than those in beef *** analysis showed that the most prevalent resistance genes were blojm and oodAl in dairy cattle and beef cattle,***,seven diarrheagenic virulence genes(irp2,fyuA,Stx1,eoeA,F41,K99 and STa)were present in the isolates from dairy cattle,with a prevalence ranging from 3.7%to 22.22%.Six diarrheagenic virulence genes(irp2,fyuA,Stx1,eoeA,hylA and F41)were identified in the isolates from beef cattle,with a prevalence ranging from 2.27%to 63.64%.Our results provide important evidence for better exploring their interaction *** studies are also needed to understand the origin and transmission route of£coli in cattle to reduce its prevalence.

Selenium(Se)is an essential trace element that has a large number of biological functions for ***,the absorption kinetics of Se from sodium selenite in the small intestine of broilers remains ***,two experiments were conducted with 28-d-old commercial male broilers to study the kinetics of Se absorption in ligated small intestinal *** experiment 1,the Se absorption in duodenal,jejunal,and ileal segments at different post-perfusion time points(0,20,40,60,80,100 and 120 min)were *** experiment 2,a kinetic study of Se absorption was conducted with the duodenal,jejunal,and ileal loops perfused with solutions containing 0,0.0375,0.075,0.15,0.30,or 0.60μg mL^-1 of Se as sodium selenite,and Se contents in perfusates were determined at 100 min after *** results from experiment 1 showed that the Se absorption increased in an asymptotic response(P0.05)in the Se absorption rate among different ligated small intestinal segments perfused with solutions containing 0.0375–0.15μg mL^-1 of Se,however,the Se absorption rate was higher(P<0.05)in the jejunum than that in the duodenum perfused with solutions containing 0.30–0.60μg mL^-1 of *** kinetic curves of Se absorption demonstrated that the Se absorption was a saturated carrier-mediated process in the duodenum,and the maximum absorption rate was 1271 pg min^-1 cm^-1;whereas the Se absorptions were a non-saturated diffusion process in the jejunum and ileum,and the diffusive constants were 2107 and 1777 cm2 min^-1,*** results from the present study indicate that the jejunum is the main Se absorption site,and the Se absorption i

A novel live attenuated vaccine candidate protects chickens against subtype B avian metapneumovirus

Avian metapneumovirus(aMPV) is a highly contagious pathogen that causes acute upper respiratory tract diseases in chickens and turkeys, resulting in serious economic losses. Subtype B aMPV has recently become the dominant epidemic strain in China. We developed an attenuated aMPV subtype B strain by serial passaging in Vero cells and evaluated its safety and efficacy as a vaccine candidate. The safety test showed that after the 30th passage, the LN16-A strain was fully attenuated, as clinical signs of infection and histological lesions were absent after *** LN16-A strain did not revert to a virulent strain after five serial passages in chickens. The genomic sequence of LN16-A differed from that of the parent wild-type LN16(wtLN16) strain and had nine amino acid mutations. In chickens, a single immunization with LN16-A induced robust humoral and cellular immune responses, including the abundant production of neutralizing antibodies, CD4^(+) T lymphocytes, and the Th1(IFN-γ) and Th2(IL-4 and IL-6)cytokines. We also confirmed that LN16-A provided 100% protection against subtype B aMPV and significantly reduced viral shedding and turbinate inflammation. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype B aMPV.

After coronavirus disease 2019(COVID-19),previously known as pneumonia of unknown etiology(PUE)and 2019 novel coronavirus(2019-nCoV),was first discovered in Wuhan City,Hubei Province,the Chinese government took measures to stop the spread of the epidemic nationwide and interrupted local transmission in China(1–2).Although there have been several localized outbreaks that were caused by overseas importation of the virus—including heilongjiang,Jilin,Beijing,Liaoning,and other provincial-level administrative divisions(PLADs)—these outbreaks have been stopped within two to four weeks and were limited in scope to less than ten cases or a few hundred cases for each *** the COVID-19 pandemic still uncontrolled globally,the threat of importing the virus continues unabated and still threatens China’s population,virtually all of which is susceptible.

Establishment and Preliminary Application of One-step Reverse Transcriptase Droplet Digital PCR Assay for Bovine Viral Diarrhea Virus

[Objective]The paper was to establish a one-step reverse transcriptase droplet digital PCR(RT-ddPCR)assay for bovine viral diarrhea virus(BVDV).[Method]Based on one-step real-time quantitative PCR(RT-qPCR)assay,BVDV-specific primers and probes were designed in this *** reverse transcriptase,annealing temperature,primer and probe concentrations and reaction conditions of RT-ddPCR assay were ***,the specificity,sensitivity and repeatability of RT-ddPCR assay were evaluated.[Result]The optimal reverse transcription system for the established RT-ddPCR assay was as follows:commercial one-step reverse transcriptase droplet digital PCR kit with matching reagents,a final primer concentration of 900 nmol/L,a final probe concentration of 250 nmol/L and an optimal annealing temperature of 57℃.The results were negative when the method was used to detect other common epidemic viruses;the minimum detection limit was 3.2 copies/μL with good repeatability,and the coefficient of variation was less than 5%.RT-ddPCR and RT-qPCR assays were used to test 24 bovine swab samples and the test results showed that the established RT-ddPCR assay was superior to RT-qPCR assay.[Conclusion]The RT-ddPCR assay established in this study has strong specificity,high sensitivity and good repeatability,and is suitable for nucleic acid detection of clinical *** study provided a technical support for early detection and quantitative diagnosis of BVDV infection.
