Pangolins are among the most critically endangered animals due to widespread poaching and worldwide trafficking. Captive breeding is considered to be one way to protect them and increase the sizes of their populations. However, comparative studies of captive and wild pangolins in the context of gut microbiota are rare. Here, the gut microbiome of captive and confiscated-rescued wild pangolins is compared, and the effects of different periods of captivity and captivity with and without antibiotic treatment are *** show that different diets and periods of captivity, as well as the application of antibiotic therapy, can alter gut community composition and abundance in pangolins. Compared to wild pangolins, captive pangolins have an increased capacity for chitin and cellulose/hemicellulose degradation, fatty acid metabolism, and short-chain fatty acid synthesis, but a reduced ability to metabolize exogenous substances. In addition to increasing the ability of the gut microbiota to metabolize nutrients in captivity, captive breeding imposes some risks for survival by resulting in a greater abundance of antibiotic resistance genes and virulence factors in captive pangolins than in wild pangolins. Our study is important for the development of guidelines for pangolin conservation, including health assessment, disease prevention, and rehabilitation of wild pangolin populations.

MicroRNAs(miRNAs) are a group of small, endogenous, single-stranded non-coding RNAs that post-transcriptionally regulate gene expression levels. Previous studies have revealed that miRNAs play key roles in multiple biological processes, such as growth and development in both animals and plants. Computational identification is an efficient method for miRNA prediction in organisms with a reference genome before high-throughput miRNA sequencing experiments. In this study, we employed an integrated strategy combining the homology-based and ab initio approaches to predict miRNAs from the genome and transcriptome of large yellow croaker, one of the most commercially important marine fish in China and East Asia. A total of 418 miRNA molecules, including 287 miRNAs by the homology-based method and 131 miRNAs by the ab initio approach, were identified for large yellow croaker. Additionally, 16 053 target genes were predicted and annotated for Gene Ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) databases. Meanwhile, we analysed single nucleotide polymorphisms(SNPs) around large yellow croaker miRNA and found that the miRNA seed regions were significantly less prone to mutations, indicating that the miRNA sequences were under strict natural selection based on their essential regulation functions in living cells. Twenty-two SNPs were identified in large yellow croaker miRNA seed regions, which dramatically influenced the miRNA-gene regulation networks. This is the first reported miRNA detection from both the genome and transcriptome using the integrated strategy for large yellow croaker species, and the miRNA and SNP analyses in this work provide important resources and a reference for subsequent miRNA functional investigations in large yellow croaker.

Attenuation of Virulent Porcine Reproductive and Respiratory Syndrome Virus Strain CH-1a and Genetic Variation of ORF5 Gene

To develop a modified live vaccine （MLV） against porcine reproductive and respiratory syndrome virus （PRRSV）, virulent CH-Ia strain was attenuated by serial passages up to 130 passage （P130） in Marc-145 cells. The virulence and immune efficacy of the attenuated CH-1 a were evaluated in pigs. The results showed that animals inoculated with P130 did not develop any clinical sign of the disease, but produced rapid and effective humoral immune responses against PRRSV challenge, indicating that attenuated CH-1 a P 130 is the candidate as the effective vaccine against PRRSV. To define the potential mutations in the attenuated CH-la genome, we sequenced and analyzed the ORF5 gene of CH-la strain of different passages （P39, P55, P65, P70, P85, P100, P115, P120, P125, and P130） and found that three mutations （C5Y, H38Q and L146Q） which may be related with the attenuation of CH-la. In addition, we also found a unique restriction enzyme site （TspEI） in the ORF5 gene of attenuated CH-la, which can be used as a genetic marker to distinguish original and attenuated CH- 1 a.

Locust Behaved Particle Swarm Optimization Technique

The collective behavior of certain animals and insects has the characteristic of self-organization. The simple interactions among individuals can produce complex adaptive patterns at the level of the group. Recently,new scientific investigation pointed out that desert locusts show extreme phenotypic plasticity in transforming between the lonely phase and the swarming gregarious phase depending on the population density,which is controlled by a serotonin called 5-hydroxytryptamine( 5HT). In this paper,based on the mechanism of the locusts' collective behavior,a new particle swarm optimization technique called LBPSO is studied. The number of swarms is selfadaptively adjusted by the acquired outstanding particles coming from behind the previous global best solution. The swarm sizes are related to the corresponding serotonin 5HT,which is determined by the optimization parameters such as global best and iteration number. And each swarm adopts one of three rules below according to its density, generalized social evolution strategy, generalized cognition evolution strategy and the independent moving strategy. A comparative study of LBPSO,social particle swarm optimization( SPSO), improved SPSO and the standard particle swarm optimization( StdPSO) on their abilities of tracking optima is carried out. And the results under four static benchmark functions and a dynamic function generator moving peaks benchmark( MPB)show that LBPSO outperforms the other three functions in both static and dynamic landscapes due to the introduced locusts' collective behavior.

Ten thousands of bone fragments were recovered from the Ma’anshan Paleolithic site, of which 4358 pieces can be identified to skeletal elements or species. In this research, the bone assemblages are quantified based on elements of MNI, MNE, and MAU. Then bone surface modifications and skeletal element profiles of the bone assemblages are studied to understand shlepping behaviors and reveal the technique of disarticulating and defleshing the animals’ bodies, etc. Based on the analyses of bone assemblages and the comparative study with the ethnoarchaeological as well as experimental data, it is assumed that later hominids at the site made fuller use of the game animals than the earlier hominids. Incorporating the lines of evidence from chronological dating, paleoenvironment, and polished bone tools between the two cultural layers, here we propose three hypotheses suggesting that the changes of environment, the migrations of the hominids from the North, or the demographic pulses may cause such a behavioral difference. However, it is yet to be determined which hypothesis can better explain the changes.

The present study provides a comparative understanding of florfenicol deposition kinetics following oral administration at a single dose of 30 mg/kg body weight to Japanese eel （ Anguilla japonica） at the temperature of 20℃, 24℃ and 28℃, respectively. Approximate 0.3 mL of blood sample from each eel in these three groups was collected in a row at the time 0.5, 1, 2, 4, 8, 12, 24 and 36 h after medicated-feed gavage. The concentrations of florfenicol in eel plasma were detected by reversed-phase high performance liquid chromatography （ RP - HPLC ）. The plasma concentration-time data of each eel were analyzed by non-compartmental methods based on statistical moment theory using DAS software. Pharmacokinetics parameters of different groups were tested by one-way analysis of variance using SPSS software. It was found that most of parameters were different significantly among groups （ P 〈 0.05 ）. In the three groups at 20℃, 24℃ and 28℃, pharmacokinetics parameters of peak plasma concentration （ Cmax ） were （7. 839 ± 1. 125）, （ 13. 010 ± 2. 334） and （ 18. 267 ± 3. 717 ） μg/mL, and the time to reach the Cmax（Tmax） were （6.500± 2.070）, （4.500 ± 1.414） and （3.429 ± 0.926） h, respectively. These suggested that eels at higher temperature absorbed more drug and more quickly. The volumes of distribution （Vz/F） were （3. 964 ± 0. 594）, （2. 466 ± 0. 672） and （1. 841 ± 0.485） IZkg, respectively. The difference deduced that more florfenicol was bound to tissue in eels at lower temperature. The mean residence time （ MRT0-∞ ） and the half-life of drug （ t1/2, ） in the three respective groups were （31.503 ±7.117）, （22.881 ± 4.940） and （22.134 ± 6.204） h, and （21.243 ±5.166）, （14.994±4.293） and （14. 656± 5. 061 ） h. These parameters showed that the elimination rate of florfenicol in eels at 24℃ or 28℃ was more quickly than that at 20℃. The areas under the concentration -time curve （AUC0-∞ ） were （235.580 ±62.013）, （271.983±75.023） and （353. 192 ± 92.491） μg h/mL, respectively. It indicat
