Assembly of perilla seed protein isolate-pectin nanocomplex to deliver curcumin: Properties, characterization, molecular interactions and antioxidant activity

In the present study, nanocomplexes composed of perilla seed protein isolate (PSPI) and pectin (PEC) were assembled for the delivery of curcumin (Cur) based on the pH-driven self-assembly method. The encapsulation efficiency of Cur was 87.77%, and the PSPI-Cur-PEC exhibited controlled release of Cur during the in vitro digestion. Moreover, the PSPI-Cur-PEC possessed efficient antioxidant activity and provided greater defense for Cur against thermal treatment and ultraviolet irradiation. Fourier transform infrared spectroscopy, fluorescence spectroscopy and X-ray diffraction results indicated that entrapped Cur was existed in the form of amorphous state in PSPI-PEC, and the process was driven by hydrophobic effects, hydrogen bonding and electrostatic interactions. Scanning electron microscopy images revealed substantial morphological changes of the nanocomplexes throughout the process, while transmission electron microscopy images observed the core-shell structure of PSPI-Cur-PEC nanocomplexes. Furthermore, PSPI-PEC exhibited excellent biocompatibility by the in vitro cytotoxicity assay on Caco-2 cells, and the intermolecular interactions and possible formation mechanism of PSPI-Cur-PEC were further determined by molecular docking. This work elucidated that PSPI-PEC nanocomplexes assembled by a green and energy-efficient pH-driven method was feasible to protect sensitive hydrophobic bioactive components and provided a possible utilization of perilla seed protein in delivery system for nutraceuticals.

Objectives: Bacillus cereus isolates from vegetables can cause diarrheal syndrome food poisoning. Lettuce, the most highly consumed ready-to-eat vegetable, poses a significant risk of B. cereus exposure. Hence, B. cereus monitoring and evaluation during cultivation are essential for preventing foodborne illnesses caused by this food-pathogen combination. However, the impact of different cultivation practices on B. cereus contamination in lettuce currently remains unclear. Furthermore, a validity indicator for the experimental models cannot be effectively validated under field conditions. Materials and Methods: Herein, we assessed the risk of B. cereus exposure using lettuce grown under two different cultivation farming systems in China (open-field versus greenhouse) using a quantitative microbial risk assessment model. Tukey's mean comparison method and kernel density estimation frequency distribution statistics were used for model validation using environmental samples and lettuce. Results: The mean concentration of B. cereus in open-field-grown lettuce was significantly higher than that in greenhouse-grown lettuce (P0.05), verifying the accuracy of the risk model. Conclusion: Soil solarization, complete manure treatment, usage of mulch films, and irrigation water quality should be considered in agricultural practices to control B. cereus concentrations in lettuce. Overall, we found that B. cereus was more abundant in open-field-grown lettuce, and we provide a risk evaluation framework that can guide farmers and policy-makers to control B. cereus contamination in farm-to-fork lettuce chains in China.

Plant-based milk is considered a healthy and environmentally sustainable option. In order to solve the physical and chemical instability problem of most plant milk and increase the chance of consumer acceptance of the flavor, this experiment used soybean as raw material, fructo-oligosaccharides (FOS) as prebiotics, and added kombucha for fermentation. The microbial composition, functional components, antioxidant capacity, physico-chemical properties and flavor relationship of fermented soy milk at different fermentation times (up to 96 h) were studied by high-throughput sequencing, laser confocal scanning imaging, and gas chromatography-mass spectrometry. According to the results, the synbiotic interaction of FOS and kombucha consortium promoted the growth of yeast and lactic acid bacteria in soy milk. The ideal fermentation time was 84 h. At this time, fer-mented soy milk added with FOS displayed increased beta-glucosidase activity (by 68.10 mU/mL), isoflavones mainly represented by genistein content (by 612.41%), and DPPH radical scavenging activity (by 25.02%). Furthermore, the addition of FOS intensified the sour taste of the 84 h-fermented soy milk. The content of hexanal was reduced, while favorable flavor substances, such as citric acid and linalool, were also produced. Soymilk fermented with kombucha and FOS provides an opportunity to explore the physical and chemical characteristics and the mechanism of flavor formation in multi-strain co-fermentation systems and to develop commercial plant-based fermentation products.

Cloning and characterization of a C genome-specific repetitive sequence in Aegilops caudata L.

pAeca212 is 204 bp in length, and the G + C content is 51%. It disperses on all seven chromosome pairs of Aegilops caudata except centromeres and secondary *** with the 316893 DNA sequences registered in Genbank/EMBL/DDJB/PDB, pAeca212 is a new C-genome specific repetitive sequence. The results of genomic specificity analysis of pAeca212 show that there are no hybridization signals detected in all donor Poaceae plants except in rye. pAeca212 is a very useful molecular marker in the study of the origin of Triticeace and the detection of C chromatin in wheat background.

Expression of tachyplesin gene in yeast Pichia pastoris

Tachyplesin gene was designed and chemically synthesized with codon usage bias. The stop codon TAG and some restriction sites convenient for further cloning were added to this gene. The synthesized gene cloned into yeast expression vector pPIC9 (with α_secretion signal) was transformed into host strain GS115 by electroporation. The tachyplesin expressed from recombinants Y PIC27 and Y PIC42 showed an inhibition activity against the germination of the spores of \%Magnaporthe grisea\%. Southern blot performed with chromosome genome of the two recombinants indicated a single copy of the expression cassette was integrated at the chromosomal AOX 1 locus by which the genomic AOX 1 gene was functionally disrupted and Northern blot showed the presence of transcripts of the tachyplesin gene.

Expression of rabbit defensin NP-1 gene in transgenic tobacco plants and its activity against bacterial wilt

A cDNAderived 200 bp fragment encoding a rabbit defensin NP1 mature peptide was ligated into the GUSless pBI121 to yield pBIC35NP1. Tobacco leaf discs were genetically transformed with an agrobacterium tumefaciens strain harbouring pBIC35NP1. Northern blot analysis showed that the defensin NP1 expression cassette was normally transcribed in transgenic plants. Resistance tests demonstrated that expression of native defensin NP1 can confer partial resistance to the bacterial wilt pathogen, Ralstonia solanacearum.

Gene cloning and function analysis of ABP9 protein which specifically binds to ABRE2 motif of maize Cat1 gene

A cDNA library was constructed using mRNA extracted from 17 days post-pollination (dpp) maize embryos and was screened by employing a yeast one-hybrid system for proteins specifically interacting with ABRE2 motif of maize Call gene. Three truncated overlapping positive clones designated ABP9 were obtained and the full-length cDNA was isolated by 5’ RACE. Searching the database revealed that ABP9 protein belongs to a bZIP-type transcription factor family. ABP9 protein speciflcally binds to ABRE2 motif and activates the expression of downstream reporter gene in yeast cells. Our results strongly suggest that the ABP9 protein functions as a transcription activator.

Evaluation of transgenic tobacco expressing two insecticidal genes to delay resistance development of Helicoverpa armigera

Chromosomal location of yellow rust resistance gene(s) in Triticum aestivum-Lophopyrum elongatum substitution lines

A set of T. aestivum-L. elongatum substitution lines were studied on yellow rust resistance at seedling stage, inheritance of the resistance and esterase-5 (Est-5) analysis. The results demonstrated that L. elongatum carried a new yellow rust resistance gene(s), which was dominant and located on chromosome 3E of L. elongatum. The biochemical locus encoding Est-5 was also located on chromosome 3E, and was tentatively named Est-E5, which was co-segregant with the Yr gene(s) in wheat background. In addition, the transmission frequencies of chromosome 3E in 3A/3E and 3D/3E hybrids selfing were significantly higher than that of chromosome 3E in 3B/3E hybrid, which was probably due to the difference on genetic relationships among A, B, D and E genomes.

Genetic study on the behavior of honeybees influenced by polyandry system

Within a honeybee population, due to polyandry, there are super-sister and half-sister relations, thus many sub-families exist. For the Chinese honeybee (Apis cerana cerana F.) a phylogenetic dendrogram has been constructed in which 4 sub-families are clustered based on DNA fingerprint patterns. And it has been observed that each kind of workers is distributed to several different sub-families.
