Effect of peroxiredoxin 6 on total and progressive motility of human spermatozoa after cryopreservation

Sperm cryopreservation is useful in assisted reproductive technology and male fertility ***,freezing and thawing significantly reduces the total and progressive motility of human *** the present study,we explored the effects of peroxiredoxin 6(PRDX6)on total and progressive motility of human spermatozoa after *** samples of 20 males with normal parameters were collected and frozen in media supplemented with different concentrations of PRDX6(0 mM,10−5 mM,10−7 mM,and 10−9 mM,respectively).Postthaw total and progressive motility of sperms were *** results showed that in comparison with 0 mM,the concentrations of 10−5 mM,10−7 mM,and 10−9 mM of PRDX6 all significantly improved total motility and progressive motility of sperms(p<0.05).The 10−7 mM of PRDX6 showed the best *** conclusion,the supplementation of PRDX6 helps to maintain the total and progressive motility of human spermatozoa.

Tolerance of neurite outgrowth to Rho kinase inhibitors decreased by cyclooxygenase-2 inhibitor

In this study, PC12 Adh cells and Neuro-2a cells were treated with Rho-associated kinase inhibitors （Y27632 and Fasudil）, a cyclooxygenase-1 selective inhibitor （SC560）, and a cyclooxygenase-2 inhibitor （NS398）. We found that these cells became tolerant to Rho-associated kinase inhibitors, as neurite outgrowth induced by these inhibitors diminished following more than 3 days of exposure in either cell line. The proteins cyclooxygenase-2 and cytosolic prostaglandin E synthetase were upregulated at day 3. NS398 decreased the tolerance to neurite outgrowth induction in both cell lines, whereas SC560 had almost no effect. These findings indicate that cells become tolerant to neurite outgrowth induced by Rho-associated kinase inhibitors, this is at least partly associated with upregulation of proteins involved in the cyclooxygenase-2 pathway, and cyclooxygenases-2 inhibition prevents this tolerance.

Present situation and analysis on the training of chinese basketball talents

At present, china has entered the era of market economy. Reserve youths training system has changed greatly. The training system formed by the planned economy period reserve talents, has been incompatible with the society, resulting in the merchandise of reserve basketball talents. Talented young players have no qualitative progress and there is no highlight on them, so do the player groups. The training of reserve sports talents is being adjusted and converted, and training a great diversity of reserve sports talents will coexist for long. The whole nation training system in the era of the planned economy, through policy intervention, the highly centralized amateur sports school, provincial sports team promoting to the national team management system, guarantees the stability of the sports teams in china, and provides reserve youth force. However, the cultural quality of the athletes and the level of scientific training are not high.

Comparison of Aerosol Effects on Simulated Spring and Summer Hailstorm Clouds

Numerical simulations are carried out to investigate the effect of cloud condensation nuclei（CCN） concentrations on microphysical processes and precipitation characteristics of hailstorms. Two hailstorm cases are simulated, a spring case and a summer case, in a semiarid region of northern china, with the Regional Atmospheric Modeling System. The results are used to investigate the differences and similarities of the CCN effects between spring and summer hailstorms. The similarities are：（1） The total hydrometeor mixing ratio decreases, while the total ice-phase mixing ratio enhances, with increasing CCN concentration;（2） Enhancement of the CCN concentration results in the production of a greater amount of small-sized hydrometeor particles, but a lessening of large-sized hydrometeor particles;（3） As the CCN concentration increases, the supercooled cloud water and rainwater make a lesser contribution to hail, while the ice-phase hydrometeors take on active roles in the growth of hail;（4） When the CCN concentration increases, the amount of total precipitation lessens,while the role played by liquid-phase rainfall in the amount of total precipitation reduces, relatively, compared to that of icephase precipitation. The differences between the two storms include：（1） An increase in the CCN concentration tends to reduce pristine ice mixing ratios in the spring case but enhance them in the summer case;（2） Ice-phase hydrometeor particles contribute more to hail growth in the spring case, while liquid water contributes more in the summer case;（3） An increase in the CCN concentration has different effects on surface hail precipitation in different seasons.

Effect of Taichong(LR 3) acupuncture in spontaneously hypertensive rats

OBJECTIVE: To evaluate the effects of Taichong(LR3) acupuncture points(acupoints) on the expression of glucose transporter protein 1(GLUT1) in the hypothalamus of spontaneously hypertensive rats(SHRs) as measured by combined positron emission tomography and computed tomography(PETCT).METHODS: Spontaneously hypertensive rats(SHR)were divided into model, Taichong(LR 3) acupuncture, and sham groups. Additionally, Tokyo Wistar rats were used as the control group. Changes in blood pressure were recorded in different groups of rats before and after the corresponding treatment. Hematoxylin and eosin(HE) staining was used to study basic morphological changes, and immunohistochemistry was used to determine GLUT1 expression in the hypothalamus. Further,PET-CT was utilized to elucidate the antihypertensive mechanism after acupuncture at the Taichong(LR 3) ***: PET-CT indicated activation of the hypothalamus. Measurement of blood pressure showed that acupuncture at the Taichong(LR 3) acupoints lowered blood pressure. HE staining did not show any significant pathological changes, although differences in cell number were observed. Immunohistochemical analysis indicated a GLUT1 downregulation in the SHRs of the Taichong(LR 3) acupuncture group after the ***: Acupuncture at Taichong(LR 3) acupoints lowered blood pressure in SHRs, with possible mechanisms being changes in cell number and GLUT1 expression in the hypothalamus.

Therapeutic mechanism of Yīn-Chén-Hāo decoction in hepatic diseases

Yīn-Chén-Hāo decoction(YCHD) is a traditional chinese medicine.formula composed of capillaris(Artemisia capillaris), gardenia(Gardenia jasminoides), and rhubarb(Rheum rhabarbarum) that is used for the treatment of damp-heat jaundice. In modern clinics, YCHD is mostly used for hepatic diseases. This review summarizes the biological activities of YCHD and its medical applications. The main active compounds of YCHD are chlorogenic acid, rhein, geniposide, emodin, and scoparone. The pharmacological actions of YCHD include inhibition of hepatic steatosis, apoptosis, necrosis, anti-inflammation, and immune regulation. YCHD could be developed as a new therapeutic strategy for the treatment of hepatic diseases.

Effect of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV 6) for the treatment of diarrhea-dominant irritable bowel syndrome： a patient-blinded pilot trial with orthogonal design

OBJECTIVE: To investigate the effects of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV6) in patients with diarrhea-dominant irritable bowel syndrome(D-IBS) by assessing predominant factors, and determining the best factor combinations and their interactions. To identify the optimal quantity of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV 6) to achieve optimal ***: An L9(3~4) orthogonal design was applied to 233 confirmed subjects based on the three most influential factors and the three most frequently used levels. Nine programs were *** were assigned to four subgroups according to age and gender; each subject underwent one of the nine programs randomly. We selected Tianshu(ST 25, bilateral) and Qihai(CV 6). The scores of symptoms and the IBS Quality of Life(IBS-QOL) were evaluated after the final ***: After treatment, clinical symptoms of all patients were significantly improved(P = 0.001).Quality of life also improved significantly(P < 0.05).For all the participants, frequency was the dominating factor regarding symptoms(P = 0.01). Duration was the dominating factor for quality of life(P =0.0001). The best combination for improving symptoms in 30-40-year-old male and female patients was two courses of 10 min each once a day, and for41-50-year-old male and female patients it was three courses of 10 min(males) or 30 min(females)twice a day. The best combination for quality of life was three courses of 20 min once a day. No interaction was found between frequency and course, or frequency and ***: Warming moxibustion Tianshu(ST25, bilateral) and Qihai(CV 6) is a promising therapy for D-IBS. To acquire optimal efficacy, the three main factors(duration, frequency and treatment course) of the warming moxibustion should reach a specific quantity and be combined appropriately.

Right-and left-sided colorectal cancers respond differently to traditional chinese medicine

AIM To explore the differences in the responses of left-sided colorectal cancer(LSCRC) and right-sided colon cancer(RSCC) to traditional chinese medicine.TCM).METHODS Patients with postoperative stage I-III colorectal cancer(CRC) were enrolled and divided into the LSCRC with or without TCM and RSCC with or without TCM groups depending on the primary tumor side and TCM administration. Patients in the TCM group were given TCM for at least 6 mo. Our research adopted diseasefree survival(DFS) as the primary endpoint. We applied a Cox proportional hazards regression model for the multivariate factor analysis using Stata 12.0 and SPSS 22.0 software for data *** Of the 817 patients included in our study, 617 had LSCRC(TCM group, n = 404; Non-TCM group, n = 213), and 200 had RSCC(TCM group, n = 132; NonTCM group, n = 68). The 6-year DFS for patients with LSCRC was 56.95% in the TCM group and 41.50% in the Non-TCM group(P = 0.000). For patients with RSCC, the 6-year DFS was 52.92% in the TCM group and 37.19% in the Non-TCM group(P = 0.003). Differences between LSCRC and RSCC were not statistically significant regardless of TCM *** Patients with either LSCRC or RSCC and who took TCM experienced longer DFS; furthermore, patients with RSCC benefited more from TCM in DFS.

Establishment and reliability evaluation of the design space for HPLC analysis of six alkaloids in Coptis chinensis(Huanglian) using Bayesian approach

Coptis chinensis(Huanglian) is a commonly used traditional chinese medicine.TCM) herb and alkaloids are the most important chemical constituents in it. In the present study, an isocratic reverse phase high performance liquid chromatography(RP-HPLC) method allowing the separation of six alkaloids in Huanglian was for the first time developed under the quality by design(Qb D) principles. First, five chromatographic parameters were identified to construct a Plackett-Burman experimental design. The critical resolution, analysis time, and peak width were responses modeled by multivariate linear regression. The results showed that the percentage of acetonitrile, concentration of sodium dodecyl sulfate, and concentration of potassium phosphate monobasic were statistically significant parameters(P < 0.05). Then, the Box-Behnken experimental design was applied to further evaluate the interactions between the three parameters on selected responses. Full quadratic models were built and used to establish the analytical design space. Moreover, the reliability of design space was estimated by the Bayesian posterior predictive distribution. The optimal separation was predicted at 40% acetonitrile, 1.7 g·m L-1of sodium dodecyl sulfate and 0.03 mol·m L-1 of potassium phosphate monobasic. Finally, the accuracy profile methodology was used to validate the established HPLC method. The results demonstrated that the Qb D concept could be efficiently used to develop a robust RP-HPLC analytical method for Huanglian.

First-Principles Calculations of Phase Transition and Stability of Si2CN4 under High Pressure

A pressure-induced phase transition and stability in Si2 CN4 polymorphs under high pressure are studied by firstprinciples calculations. The result shows that the phase transition pressure of α- and β-Si2 CN4 to the cubic spinal phase is 29.9 GPa and 27.5 GPa predicted by thermodynamic method respectively. Under ambient condition, all of the three Si2CN4 polymorphs are metastable with positive formation enthalpy. Unlike the stability of Si3N4 polymorphs, α-Si2 CN4 is more stable than the β phase.
