Longitudinal assessment of peripheral organ metabolism and the gut microbiota in an APP/PS1 transgenic mouse model of Alzheimer’s disease

Alzheimer’s disease not only affects the brain,but also induces metabolic dysfunction in peripheral organs and alters the gut *** aim of this study was to investigate systemic changes that occur in Alzheimer’s disease,in particular the association between changes in peripheral organ metabolism,changes in gut microbial composition,and Alzheimer’s disease *** do this,we analyzed peripheral organ metabolism and the gut microbiota in amyloid precursor protein-presenilin 1(APP/PS1)transgenic and control mice at 3,6,9,and 12 months of ***-month-old APP/PS1 mice exhibited cognitive impairment,Alzheimer’s disease-related brain changes,distinctive metabolic disturbances in peripheral organs and fecal samples(as detected by untargeted metabolomics sequencing),and substantial changes in gut microbial composition compared with younger APP/PS1 ***,a strong correlation emerged between the gut microbiota and kidney metabolism in APP/PS1 *** findings suggest that alterations in peripheral organ metabolism and the gut microbiota are closely related to Alzheimer’s disease development,indicating potential new directions for therapeutic strategies.

OBJECTIVE:To observe the intervention of Chushizi(Fructus Broussonetiae)(CSZ)on drug-induced liver injury(DILI)in rats,as well as indicators of liver function,serum levels of inflammatory cytokines,and expression of proteins and m RNA associated with toll-like receptor 3(TLR3)and the signal transducer and activator of transcription 3(STAT3)pathway in the liver[TLR3,janus protein tyrosine kinase 2(JAK2),c-jun,c-fos,c-Jun N-terminal kinase 2(JNK2),and STAT3].METHODS:Forty specified pathogen free grade Sprague-Dawley rats were randomly divided into the control group,the model group,the silybin group and the CSZ *** were given acetaminophen(APAP)to trigger *** of the liver was observed by hematoxylin-eosin *** levels of alanine aminotransferase(ALT),aspartate transaminase(AST),direct bilirubin(DBIL),and total bilirubin(TBIL)in serum were detected by a semi-automatic biochemical *** of tumor necrosis factor alpha(TNF-α),interleukin(IL)-6,IL-13,and IL-22 in serum were detected by the enzyme-linked immunosorbent assay,the expression of TLR3,phosphorylation of JAK2(p-JAK2),while c-jun and c-fos proteins in the liver were determined by immunohistochemistry;expression of JNK2,and STAT3 in the liver were assayed by Western blot and real-time quantitative polymerase chain reaction.P-JNK2 and p-STAT3 in the liver were assayed by Western ***:After treatment,the activity of ALT,AST,and concentrations of TBIL,DBIL,TNF-α,IL-6,as well as IL-13 in serum,were lower than those in the model group,and expression of p-JAK2,TLR3,c-jun,c-fos,p-STAT3,and p-JNK2 could be ***:Our findings suggest that CSZ is a valid medicine.to alleviate APAP-induced DILI,while its partial mechanism may regulate the TLR3/JNK/c-jun/c-fos/JAK/STAT3 pathway.

Effects of modified Wenjing decoction on microcirculation in reproductive organs in rats with symptom patterns of cold coagulation and blood stasis

OBJECTIVE: To investigate the relationship between symptom patterns of cold coagulation and blood stasis(CCBS) and microcirculation disturbance. In addition, we determined the efficacy of modified Wenjing decoction(WJD) for the treatment of ***: CCBS was induced in rats with an icewater bath treatment. The ovarian function, microvascular and circulatory status of reproductive organs, and function of local microvascular endothelial cells(VECs) and vascular smooth muscle cells(VSMCs) were ***: Ovarian dysfunction was observed in the rats with CCBS. It was characterized by the presence of an estrous cycle disorder and a decrease in reproductive hormone levels. Microvascular circulation disorders were associated with an imbalance in vasoconstriction, relaxation substances, nitric oxide, abnormal blood flow in whole blood, and decreased blood flow in the auricle and uterus. VECs were damaged, and VSMCs contracted and proliferated in ovarian and uterine ***: Our findings suggest that the dysfunctional reproductive organs observed in gynecological CCBS may be closely related to the microcirculation disturbance of local tissues, microvascular contraction, and vascular remodeling. Modified WJD can be used to treat CCBS by improving microcirculation in reproductive organs.

Laparoscopic liver resection utilizing the ventral avascular area of the inferior vena cava:A retrospective cohort study

Optimizing the key parameter to accelerate the recovery of AMOC under a rapid increase of greenhouse gas forcing

Atlantic Meridional Overturning Circulation(AMOC)plays a central role in long-term climate variations through its heat and freshwater transports,which can collapse under a rapid increase of greenhouse gas forcing in climate *** studies have suggested that the deviation of model parameters is one of the major factors in inducing inaccurate AMOC *** this work,with a low-resolution earth system model,the authors try to explore whether a reasonable adjustment of the key model parameter can help to re-establish the AMOC after its *** a new optimization strategy,the extra freshwater flux(FWF)parameter is determined to be the dominant one affecting the AMOC’s *** traditional ensemble optimal interpolation(EnOI)data assimilation and new machine learning methods are adopted to optimize the FWF parameter in an abrupt 4×CO_(2) forcing experiment to improve the adaptability of model parameters and accelerate the recovery of *** results show that,under an abrupt 4×CO_(2) forcing in millennial simulations,the AMOC will first collapse and then re-establish by the default FWF parameter ***,during the parameter adjustment process,the saltier and colder sea water over the North Atlantic region are the dominant factors in usefully improving the adaptability of the FWF parameter and accelerating the recovery of AMOC,according to their physical relationship with FWF on the interdecadal timescale.

Difference in the anti-fatigue effect of moxibustion at different acupoints in one-off exhausted rats

Objective:To explore the difference in the anti-fatigue effect of moxibustion at different acupoints through observing the impacts of moxibustion at"Shènshù(肾俞BL23)","Zúsānlǐ(足三里ST36)","Guānyuán(关元CV4)"and"Shénquè(神阙CV8)"on energy metabolic indexes in the one-off exhausted ***:A total of 56 SPF male rats were randomized into 7 groups,named a blank group,a model group,a non-acupoint group,a"BL23"group,a"ST36"group,a"CV4"group and a"CV8"group,8 rats in each *** one-off exhaustion model was established by weight-bearing swimming *** after the exhaustive swimming of rats in each intervention group,the mild moxibustion was applied at the corresponding acupoints for 15 min immediately.4 h after the exhaustive swimming,the blood was collected from the femoral artery and the quadriceps femoris of the left hindlimb was *** levels of blood lactate(BLD),lactate dehydrogenase(LDH),creatine kinase(CK)and blood urea nitrogen(BUN),the expressions of adenosine triphosphatease(ATP),fructose-6-phosphofructokinase(PFK)and malomdialdehyde(MDA)in the quadriceps femoris and the activity of superoxide dismutase(SOD)and catalase(CAT)were ***:Compared with the model group and the non-acupoint group,the levels of serum BLD,LDH,CK and BUN were all lower those in the"BL23"group,the"ST36"group,the"CV4"group and the"CV8"group,indicating a statistical significance(all P<0.05).Compared with the model group and the nonacupoint group,PFK was higher in the"BL23"group,indicating a statistical significance(both P<0.05).Compared with the model group and the non-acupoint group,PFK was higher in the"ST36"group and the"CV4"group,indicating a statistical significance(all P<0.05).Conclusion:It was found in the research that moxibustion at"BL23""ST36""CV4"and"CV8"regulated the energy metabolism in the one-off exhausted rats so as to bring the anti-fatigue effect into play and the effect was different among the *** at"CV8"may work on the preve
