Shikimic acid accelerates phase change and flowering in Chinese jujube

The juvenile-to-adult phase change with first flowering as the indicator plays a crucial role in the lifecycle of fruit trees. However, the molecular mechanisms underlying phase change in fruit trees remain largely unknown. Shikimic acid (ShA) pathway is a main metabolic pathway closely related to the synthesis of hormones and many important secondary metabolites participating in plant phase change. So,whether ShA regulates phase change in plants is worth clarifying. Here, the distinct morphological characteristics and the underlying mechanisms of phase change in jujube (Ziziphus jujuba Mill.), an important fruit tree native to China with nutritious fruit and outstanding tolerance abiotic stresses, were clarified. A combined transcriptome and metabolome analysis found that ShA is positively involved in jujube(Yuhong’×Xing 16’) phase change. The genes in the upstream of ShA synthesis pathway (ZjDAHPS, ZjDHQS and ZjSDH), the contents of ShA and the downstream secondary metabolites like phenols were significantly upregulated in the phase change period. Further, the treatment of spraying exogenous ShA verified that ShA at a very low concentration (60 mg·L^(-1)) can substantially speed up the phase change and flowering of jujube and other tested plants including Arabidopsis, tomato and wheat. The exogenous ShA (60 mg·L^(-1)) treatment in jujube seedlings could increase the accumulation of endogenous ShA, enhance leaf photosynthesis and the synthesis of phenols especially flavonoids and phenolic acids, and promote the expression of genes (ZjCOs, ZjNFYs and ZjPHYs) involved in flowering pathway. Basing on above results, we put forward a propose for the underlying mechanism of ShA regulating phase change, and a hypothesis that ShA could be considered a phytohormone-like substance because it is endogenous, ubiquitous, movable and highly efficient at very low concentrations. GST family genes in jujube actively respond to phytoplasma infection

Jujube witches’broom(JWB)caused by phytoplasma has a severely negative effect on multiple metabolisms in *** GST gene family in plants participates in the regulation of a variety of biotic and abiotic *** study aims to identify and reveal the changes in the jujube GST gene family in response to phytoplasma ***,70 ZjGSTs were identified in the jujube genome and divided into 8 *** them,the Tau-class,including 44 genes,was the *** analysis indicated that Tau-class genes were highly conserved among species,such as Arabidopsis,cotton,chickpea,and *** chromosome location analysis,37.1%of genes were clustered,and 8 of 9 gene clusters were composed of Tau class *** RT-PCR,qRT-PCR and enzyme activity detection,the results showed that the expression of half(20/40)of the tested ZjGSTs was inhibited by phytoplasma infection in field and tissue culture conditions,and GST activity was also significantly *** the resistant and susceptible varieties under phytoplasma infection,ZjGSTU49-ZjGSTU54 in the cluster IV showed opposite expression patterns,which may be due to functional divergence during *** upregulated genes(ZjGSTU45,ZjGSTU49,ZjGSTU59,and ZjGSTU70)might be involved in the process of jujube against *** yeast two-hybrid results showed that all 6 Tauclass proteins tested could form homodimers or ***,the comprehensive analysis of the jujube GST gene family revealed that ZjGSTs responded actively to phytoplasma ***,some screened genes(ZjGSTU24,ZjGSTU49-52,ZjGSTU70,and ZjDHAR10)will contribute to further functional studies of jujube-phytoplasma interactions.

An efficient and universal protoplast-based transient gene expression system for genome editing in Brassica crops

Protoplast-based transient gene expression system has been widely used in plant genome editing because of its simple operation and less *** order to establish a universal protoplast-based transient transfection system for verifying activities of genome editing vectors containing targets in Brassica,we systematically optimized factors affecting protoplast isolation and transient gene *** established an efficient protoplast-based transient gene expression system(PTGE)in Chinese cabbage,achieving high protoplast yield of 4.9×10^(5)·g^(-1)FW,viability over 95%,and transfection efficiency of 76%.We showed for the first time that pretreatment of protoplasts with a hypotonic MMG could significantly enhance the transfection ***,protoplasts incubated at 37℃ for 6 min improved the transfection efficiency to 86%.We also demonstrated that PTGE worked well(more than 50%transfection efficiency)in multiple Brassica species including cabbage,Pak Choi,Chinese kale,and ***,PTGE was used for validating the activities of CRISPR/Cas9 vectors containing targets in Chinese cabbage,cabbage,and pak choi,demonstrating the broad applicability of the established PTGE for genome editing in Brassica crops.

The steel industry is a major source of CO_(2) emissions,and thus,the mitigation of carbon emissions is the most pressing challenge in this *** this paper,international environmental governance in the steel industry is reviewed,and the current state of development of low-carbon technologies is ***,low-carbon pathways for the steel industry at the current time are proposed,emphasizing prevention and treatment ***,the prospects of low-carbon technologies are explored from the perspective of transitioning the energy structure to a“carbon-electricity-hydrogen”***,steel enterprises should adopt hydrogen-rich metallurgical technologies that are compatible with current needs and process flows in the short term,based on the carbon substitution with hydrogen(prevention)and the CCU(CO_(2) capture and utilization)concepts(treatment).Additionally,the capture and utilization of CO_(2) for steelmaking,which can assist in achieving short-term emission reduction targets but is not a long-term solution,is *** conclusion,in the long term,the carbon metallurgical process should be gradually supplanted by a hydrogen-electric synergistic approach,thus transforming the energy structure of existing steelmaking processes and attaining near-zero carbon emission steelmaking technology.

Sanhua Tang(三化汤) protects against ischemic stroke by preventing blood-brain barrier injury: a network pharmacology and in vivo experiments

OBJECTIVE: To assess the effect and mechanism of Sanhua Tang(三化汤, SHT) in treating ischemic stroke(IS) through the "Kaitong Xuanfu" theory by using network pharmacology and animal experiments. METHODS: The active ingredients and targets of SHT and IS were screened by public databases such as Traditional Chinese Medicine systems pharmacology, GeneCards, and online mendelian inheritance in man. Visual network topographies were constructed using R, Cytoscape 3.6.0, AutoDockTools, a user-sponsored molecular visualization system on an open-source foundation, and other software to analyze the correlation between targets and active ingredients. The middle cerebral artery occlusion(MCAO) model was established by operation. Animals were divided into the Sham group, MCAO group(M group), aloe-emodin(AE) group(MCAO rats treated with aloe-emodin), SHT at low dosage(SL group)(MCAO rats treated with SL), SHT at medium dosage(SM group), and SHT at high dosage(SH group). 2,3,5-triphenyl tetrazolium chloride staining was used to detect the volume of cerebral infarction; Nissl staining was used to observe the morphology of neuronal cells; transmission electron microscopy was used to observe the integrity of the blood-brain barrier(BBB); enzymelinked immunosorbent assay was used to detect the content of interleukin-6(IL-6), IL-10, tumor necrosis factor α(TNF-α) in serum. Western blot was used to detect the expression of vascular endothelial growth factor A(VEGFA) protein in the cerebral ischemic penumbra. RESULTS: Using network pharmacology and molecular docking validation, four active ingredients(lignan, naringenin, aloe-rhodopsin, and β-sitosterol), seven target proteins(protein kinase b 1, IL-6, TNF, VEGFA, TP53, jun proto-oncogene, and cysteinyl aspartate specific proteinase 3), and inflammatory signaling pathways were identified. In this paper,we consider the parallel-machine customer order scheduling with delivery time and submodular rejection *** this problem,we are given m dedicated machines in parallel and n customer *** order has a delivery time and consists of m product types and each product type should be manufactured on a dedicated *** order is either rejected,in which case a rejection penalty has to be paid,or accepted and manufactured on the m dedicated *** objective is to find a solution to minimize the sum of the maximum delivery completion time of the accepted orders and the penalty of the rejected orders which is determined by a submodular *** design an LP rounding algorithm with approximation ratio of n+1 for this problem.

Friendship Inference Based on Interest Trajectory Similarity and Co-occurrence

Most of the current research on user friendship speculation in location-based social networks is based on the co-occurrence characteristics of users,however,statistics find that co-occurrence is not common among all users;meanwhile,most of the existing work focuses on mining more features to improve the accuracy but ignoring the time complexity in practical *** this basis,a friendship inference model named ITSIC is proposed based on the similarity of user interest tracks and joint user location *** utilizing MeanShift clustering algorithm,ITSIC clustered and filtered user check-ins and divided the dataset into interesting,abnormal,and noise *** interest trajectories were constructed from user interest check-in data,which allows ITSIC to work efficiently even for users without *** the same time,by application of clustering,the single-moment multi-interest trajectory was further proposed,which increased the richness of the meaning of the trajectory *** extensive experiments on two real online social network datasets show that ITSIC outperforms existing methods in terms of AUC score and time efficiency compared to existing methods.

Strong metal-support interaction(SMSI)has a great impact on the activity and selectivity of heterogeneous catalysts,which was usually adjusted by changing reduction temperature or processing catalyst in different ***,few researches concentrate on modulating SMSI through regulating the structure of the ***,we show how changing the surface environment of the anatase TiO_(2)(B–TiO_(2))can be used to modulate the *** moderate TiOx overlayer makes the Ni metal highly dispersed on the high specific surface area of support,resulting in a substantially enhanced CO_(2)methanation ***,a novel phenomenon was observed that boron dopants promote the for-mation of the B–O–Ti interface site,enhancing the catalytic performance of CO_(2)*** calculations confirm that the B–O–Ti structure facilitates the activation of CO_(2)and further hydrogenation to methane.

Solidification crack characteristics and sensitivity of peritectic steel

The characteristics and sensitivity of solidification cracks in peritectic steels were investigated using directional solidifi-cation *** cracks were observed in both hypoperitectic steels(12CrlMoV,15CrMo)and hyper-peritectic steel(20CrMo)during solidification at growth velocities of 15,50,and 80 pm/*** the dendritic boundaries,sulphide precipitates were found,promoting crack *** on the statistical analysis of interdendritic cracks in peritectic steels,the area ratio(RA)of interdendritic cracks in a directional solidification structure was proposed to evaluate the crack sensitivity of peritectic ***,the crack sensitivities of peritectic steels(12CrlMoV,15CrMo,and 20CrMo)were tested,evaluated,and compared with the surface crack rates of three types of steels produced from a steel *** results demonstrated that RA was in good agreement with that of the steel plant,and the crack sensitivity of 12CrlMoV steel was the strongest,followed by that of 15CrMo and 20CrMo ***,RA can be used to evaluate the crack sensitivity of peritectic steel.
