Therapeutic Effect and Mechanism of Caulophyllum robustum Maxim Extract on Adjuvant Arthritis Rats

[Objectives]To study the anti-inflammation and anti-swelling effect of Caulophyllum robustum Maxim( CRM) extract on secondary lesions of adjuvant arthritis( AA),associated inflammatory factors of AA rats and mice immune function. [Methods]Rats were divided into high,middle and low dose CRM extract groups( 69. 23,34. 61,17. 31 mg/kg),methotrexate group( 0. 68 mg/kg),triptolide group( 7. 88 mg/kg),model group and normal control group. The AA rat model was induced by intradermal injection of Freund's complete adjuvant( FCA). After the AA model was established successfully,the paw swelling of AA rats was observed and recorded. The levels of interleukin-1β( IL-1β),interleukin-4( IL-4),interleukin-10( IL-10),Tumor Necrosis Factor-α( TNF-α),interleukin 17( IL-17),interferon( IFN-γ),granulocyte colony-stimulating factor( G-CSF) and transforming growth factor( TGF-β) in the serum of AA rats were detected by enzyme-linked immunosorbent assay( ELISA). The spleen was removed and weighed for detecting the drug influence on the immune *** were observed by gavage administration at different dosages of 25 mg/kg,50 mg/kg,100 mg/kg for seven days,adopting carbon granules clearance test,delayed-type hypersensitivity induced by dinitrofluorobenzene( DNFB) coupled with flow cytometry evaluating lymphocytes( CD3^+,CD4^+,CD8^+) activation and serolysin test. [Results] Compared with the model group,the joint swelling,spleen index and inflammatory cytokines( IL-1β,IL-4,IL-10,TNF-α,IL-17,IFN-γ,G-CSF,TGF-β) were reduced significantly( P < 0. 05,P < 0. 01). And the activity of CRM was significantly superior to methotrexate and triptergium. Moreover,the results indicated that different dosages of 25 mg/kg,50 mg/kg,100 mg/kg of CRM could significantly reduce the carbon granules clearance index( K) and phagocytic index( a),inhibit the phagocytosis of mononuclear macrophages,reduce the mice swelling extent and inhibit DTH in mice induced by DNFB,decrease the quantity of serum hemolysin.

Tripterygium glycosides tablet(TGT),the classical commercial drug of Tripterygium wilfordii *** been effectively used in the treatment of rheumatoid arthritis,nephrotic syndrome,leprosy,Behcet's syndrome,leprosy reaction and autoimmune ***,due to its narrow and limited treatment window,TGT-induced organ toxicity(among which liver injury accounts for about 40%of clinical reports)has gained increasing *** present study aimed to clarify the cellular and molecular events underlying TGT-induced acute liver injury using single-cell RNA sequencing(scRNA-seq)*** TGT-induced acute liver injury mouse model was constructed through short-term TGT exposure and further verified by hematoxylin-eosin staining and liver function-related serum indicators,including alanine aminotransferase,aspartate aminotransferase,alkaline phosphatase and total *** the mouse model,we identified 15 specific subtypes of cells in the liver tissue,including endothelial cells,hepatocytes,cholangiocytes,and hepatic stellate *** analysis indicated that TGT caused a significant inflammatory response in liver endothelial cells at different spatial locations;led to marked inflammatory response,apoptosis and fatty acid metabolism dysfunction in hepatocytes;activated hepatic stellate cells;brought about the activation,inflammation,and phagocytosis of liver capsular macrophages cells;resulted in immune dysfunction of liver lymphocytes;disturbed the intercellular crosstalk in liver microenvironment by regulating various signaling ***,these findings elaborate the mechanism underlying TGT-induced acute liver injury,provide new insights into the safe and rational applications in the clinic,and complement the identification of new biomarkers and therapeutic targets for liver protection.

目的 :　研究共轭亚油酸 (CLA)两种主要异构体 (9,1 1 - CLA和 1 0 ,1 2 - CLA)对两种人乳腺癌细胞 (MCF- 7,MDA- MB- 2 31 )和大鼠扩散乳腺癌细胞 (F3 )活性的异同。方法 :　采用MTT方法研究两种主要 CLA异构体在不同浓度、不同作用时间及不同给药方式下 ,抑制上述几种癌细胞增殖的能力。结果 :　 1 0 ,1 2 - CLA对所有的试验癌细胞均呈现较强的抑制活性 ,而 9,1 1 -CLA仅对 MCF- 7呈现抑制活性 ,对 MDA- MB- 2 31和 F3 基本无活性 ;1 0 ,1 2 - CLA对 F3 的抑制活性与处理时间和给药方式密切相关 ,加药后不更换培养液处理 72 h可使其抑制能力达到最大值 ;细胞周期研究显示 1 0 ,1 2 - CLA对 MCF- 7呈微弱的 G0 / G1期抑制 ,而对 F3 细胞周期则无影响。结论 :　 CLA的两种主要异构体在该实验条件下对上述三种癌细胞增殖抑制能力和作用机制有着明显的差异 ,其中 1 0 ,1 2 - CLA活性明显强于 9,1 1 - CLA。 1 0 ,1 2 - CLA是通过其代谢物对 F3

OBJECTIVE:To investigate the safety profiles of Motherwort injection(MI).METHODS:A multi-center,prospective and drugderived hospital intensive monitoring method was conducted to assess the safety of MI in real world *** study was based on a very large population after the injection was approved and marketed in *** patients using the injection in participating hospitals were monitored to determine the incidence,pattern,severity and outcome of associated adverse ***:The post-marketing surveillance was performed in 10 094 female patients from April to December,*** incidence of adverse drug reactions(ADRs) was 0.79‰(8/10 094).Among the 8 patients,the reported adverse events mainly included systemic abnormalities,such as fever,chills and eyelid edema;skin and appendages disorders,such as pruritus and rash;gastrointestinal disorders,such as nausea,abdominal distension and pain;heart rate and rhythm disorders,such as palpitation and increased heart *** of these ADRs were mild in ***:In this study the ADRs incidence rate of MI is very low,which supports that it is generally safe for use in obstetric and gynecological ***,the total number of 8 ADRs recorded over a relatively short time span seems limited,and the low number of reports could not represent an absolute guarantee of safety.

Improvement of Poplar Bark Extract on Physiological Characters of Lactococcus lactis in Intestine of White Feather Broilers

[Objective]The paper was to study the improvement of poplar bark extract on intestinal Lactococcus lactis of white feather broilers.[Method]Totally 450 Ross 308 white-feather broilers were randomly divided into five groups:control group,low dose group,medium dose group,high dose group,and antibiotic group(oxytetracycline hydrochloride).The feeding duration was 45 *** probiotics were screened and isolated through homology,and the physiological and biochemical characteristics of chicken intestinal bacteria in different groups were compared to determine the properties of bacterial *** drug resistance,antibacterial ability,proliferation ability,acid resistance and bile salt resistance of isolated strain were tested,and a strain of *** was obtained.[Result]The isolated *** was sensitive to other drugs except natural tetracyclines,and there was no significant difference among the four groups except oxytetracycline group;as the concentration of extract increased,the inhibition of *** against Salmonella ***;the medium dose extract had the largest increase in the ability to tolerate the proliferation of ***.[Conclusion]Feeding poplar bark extract will produce positive effects on the physiological characters of intestinal *** in broiler chicken,which will provide potential probiotic strain for chicken production.

Background： The food and drug administration recently announced that the use of morcellation may cause fibroids or pelvic dissemination and metastasis of uterine sarcoma; therefore, the use of morcellation is limited in the USA. A large sample study is necessary to assess the proportion of uterine malignant tumors found in patients with laparoscopic myomectomy. Methods： A national multicenter study was performed in China. From 2002 to 2014, 33,723 cases were retrospectively selected. We calculated the prevalence and recorded the clinical characteristics of the patients with malignancy after morcellation application. A total of 62 cases were finally pathologically confirmed as malignant postoperatively. Additionally, the medical records of the 62 patients were analyzed in details. Results： The proportion of postoperative malignancy after morcellation application was 0.18% （62/33,723） for patients who underwent laparoscopic myomectomy. Nearly 62.9% （39/62） of patients had demonstrated blood flow signals in the uterine fibroids before surgery. And, 23 （37.1%） patients showed rapid growth at the final preoperative ultrasound. With respect to the pathological types, 38 （61.3%） patients had detectable endometrial stromal sarcoma, 13 （21.0%） had detectable uterine leiomyosarcoma, only 3 （3.2%） had detectable carcinosarcoma, and 5 （8.1%） patients with leiomyoma had an undetermined malignant potential. Conclusions： The proportion of malignancy is low after using moreellation in patients who undergo laparoscopie myomectomy. Patients with fast-growing uterine fibroids and abnormal ultrasonic tumor blood flow should be considered for malignant potential, and morcellation should be avoided.
