Given the escalating impact of climate change on agriculture and food security,gaining insights into the evolutionary dynamics of climatic adaptation and uncovering climate-adapted variation can empower the breeding of climate-resilient crops to face future climate ***(Medicago sativa ***),the queen of forages,shows remarkable adaptability across diverse global environments,making it an excellent model for investigating species responses to climate *** this study,we performed population genomic analyses using genome resequencing data from 702 accessions of 24 Medicago species to unravel alfalfa’s climatic adaptation and genetic susceptibility to future climate *** found that interspecific genetic exchange has contributed to the gene pool of alfalfa,particularly enriching defense and stress-response *** introgression between *** ***(***)and alfalfa not only aids alfalfa’s climatic adaptation but also introduces genetic burden.A total of 1671 genes were associated with climatic adaptation,and 5.7%of them were introgressions from *** integrating climate-associated variants and climate data,we identified populations that are vulnerable to future climate change,particularly in higher latitudes of the Northern *** findings serve as a clarion call for targeted conservation initiatives and breeding *** also identified preadaptive populations that demonstrate heightened resilience to climate fluctuations,illuminating a pathway for future breeding ***,this study enhances our understanding about the local adaptation mechanisms of alfalfa and facilitates the breeding of climate-resilient alfalfa cultivars,contributing to effective agricultural strategies for facing future climate change.

There are only eight approved small molecule antiviral drugs for treating *** them,four are nucleotide analogues(remdesivir,JT001,molnupiravir,and azvudine),while the other four are protease inhibitors(nirmatrelvir,ensitrelvir,leritrelvir,and simnotrelvir-ritonavir).Antiviral resistance,unfavourable drug‒drug interaction,and toxicity have been reported in previous *** there is a dearth of new treatment options for *** this work,a three-tier cell-based screening was employed to identify novel compounds with anti-SARS-CoV-2 *** compound,designated 172,demonstrated broad-spectrum antiviral activity against multiple human pathogenic coronaviruses and different SARS-CoV-2 variants of *** studies validated by reverse genetics showed that compound 172 inhibits the 3-chymotrypsin-like protease(3CLpro)by binding to an allosteric site and reduces 3CLpro dimerization.A drug synergistic checkerboard assay demonstrated that compound 172 can achieve drug synergy with nirmatrelvir in *** vivo studies confirmed the antiviral activity of compound 172 in both Golden Syrian Hamsters and K18 humanized ACE2 ***,this study identified an alternative druggable site on the SARS-CoV-23CLpro,proposed a potential combination therapy with nirmatrelvir to reduce the risk of antiviral resistance and shed light on the development of allosteric protease inhibitors for treating a range of coronavirus diseases.

Monkeypox(mpox)outbreak in 2022 has caused more than 91,000 cases,has spread to 115 countries,regions,and territories,and has thus attracted much *** stability of poxvirus particles in the environment is recognized as an important factor in determining their ***,few studies have investigated the persistence of poxviruses on material surfaces under various environmental conditions,and their sensitivity to ***,we systematically measured the stability of vaccinia virus(VACV)under different environmental conditions and sensitivity to inactivation methods via plaque assay,quantitative real‐time polymerase chain reaction(qPCR),and Gaussia luciferase(G‐luciferase)reporter *** results show that VACV is stable on the surface of stainless steel,glass,clothing,plastic,towel,A4 paper,and tissue and persists much longer at 4℃ and?20℃,but is effectively inactivated by ultraviolet(UV)irradiation,heat treatment,and chemical *** study raises the awareness of long persistence of poxviruses in the environment and provides a simple solution to inactivate poxviruses using common disinfectants,which is expected to help the control and prevention of mpox virus and future poxvirus outbreaks.

HIV-1 reverse transcriptase(RT)has received great attention as an attractive therapeutic target for acquired immune deficiency syndrome(aids),but the inevitable drug resistance and side effects have always been major challenges faced by non-nucleoside reverse transcriptase inhibitors(NNRTIs).This work aimed to identify novel chemotypes of anti-HIV-1 agents with improved drugresistance profiles,reduced toxicity,and excellent druggability.A series of diarylpyrimidine(DAPY)derivatives were prepared via structural modifications of the leads K-5a2 and *** them,15a with dimethylphosphine oxide moiety showed the most prominent antiviral potency against all of the tested viral panel,being 1.6-fold(WT,EC_(50) Z 1.75 nmol/L),3.0-fold(L100I,EC_(50) Z 2.84 nmol/L),2.4-fold(K103N,EC_(50) Z 1.27 nmol/L),3.3-fold(Y181C,EC50 Z 5.38 nmol/L),2.9-fold(Y188L,EC_(50) Z 7.96 nmol/L),2.5-fold(E138K,EC_(50) Z 4.28 nmol/L),4.8-fold(F227L/V106A,EC_(50) Z 3.76 nmol/L)and 5.3-fold(RES056,EC_(50) Z 15.8 nmol/L)more effective than that of the marketed drug *** docking results illustrated the detailed interactions formed by compound 15a and WT,F227L/V106A,and RES056 ***,15a-HCl carried outstanding pharmacokinetic(t1/2 Z 1.32 h,F Z 40.8%)and safety profiles(LD_(50)>2000 mg/kg),which demonstrated that 15a HCl is a potential anti-HIV-1 drug candidate.

Effects of Synthetic Parameters in Pechini Method on the Formation and Properties of (ZrO_2)_(0.92)(Sc_2O_3)_(0.08) Solid Solution

Homogeneous (ZrO_2)_(0.92)(Sc_2O_3)_(0.08) solid solution in fluorite cubic structure was prepared from the gels with altered molar ratios of citric acid (CA) to metal ions (M) and ethylene glycol (EG) via a polymerization route (Pechini method). Due to the enhanced chemical homogeneity (high level of mixing of metal ions and ligands) in the polymeric gels, Sc-doped zirconia can be crystallized at temperatures as low as 400 ℃. During the evolution from amorphous gel to the crystallized (ZrO_2)_(0.92)(Sc_2O_3)_(0.08) fine powder, the bonding nature between carboxylate groups and Zr/Sc cations changed: unidentate→bridging→ionic upon calcination. The molar ratios of CA/M (1～4) and CA/EG (0.2～4) were demonstrated to affect the thermal behavior of the gels and thus the particle properties of the derived nanoparticulate oxide powders (including particle size and surface area). The as-sintered sample compacted from the nanosized powders prepared by calcining the gel with the highest content of organics (CA/M=4 and CA/EG=0.2) exhibited the best sinterability and the highest oxide ion conductivity.

Since the Cenozoic,the Tibetan Plateau has experienced large-scale uplift and outgrowth due to the India-Asia ***,the mechanism and timing of these tectonic processes still remain ***,using apatite fission track dating and inverse thermal modeling,we explore the mechanism of different phases of rapid cooling for different batholiths and intrusions in the southeastern Tibetan *** contrast to previous views,we find that the coeval granitic batholith exposed in the same tectonic zone experienced differential fast uplift in different sites,indicating that the present Tibetan Plateau was the result of differential uplift rather than the entire lithosphere uplift related to lithospheric collapse during Cenozoic *** addition,we also suggest that the 5-2 Ma mantle-related magmatism should be regarded as the critical trigger for the widely coeval cooling event in the southeastern Tibetan Plateau,because it led to the increase in atmospheric CO_(2)level and a hotter upper crust than before,which are efficient for suddenly fast rock weathering and ***,we propose that the current landform of the southeastern Tibetan Plateau was the combined influences of tectonic and climate.

Rare earth upconversion nanophosphors: synthesis, functionalization and application as biolabels and energy transfer donors

This review focused on rare earth upconversion nanophosphors (UCNPs), a particular class of emitters whose photoluminescence mechanism is of fundamental difference from that of conventional dyes and semiconductor quantum dots. We in the first section gave a brief summary on a variety of synthetic methodologies developed during the past decades. Instead of presenting an exhaustive reference list, we selected only a few representative examples, illustrating the merits and limits of each involved synthetic route. Then we surveyed the recent progress in the functionalization techniques for these nanomaterials, depicting the modification in microstructures and improvement in properties with respect to the parent nanoparticles. And finally, we emphasized their application in the research fields of biolabeling and energy transfer, narrating their superior performance benefiting from the unique excitation and emission properties.
