Identification and immune analysis of antimicrobial peptides from the cigarette beetle(Lasioderma serricorne)

Antimicrobial peptides(AMPs)in insects are endogenous peptides that are effector components of the innate defense system of the *** may serve as antimicrobial agents because of their small molecular weight and broad-spectrum antimicrobial *** this study,we performed transcriptome analysis of cigarette beetle(Lasioderma serricorne)larvae,parasitized by the ectoparasitic wasp,Anisopteromalus *** AMP genes were significantly upregulated following *** parasitism,postulating the hypothesis that the parasitization enhanced the host's resistance against pathogenic microorganisms through the regulation of host AMP ***,3 AMP genes(LsDef1,LsDef2,and LsCole)were significantly upregulated and we studied their immune function in *** challenge and functional analysis showed that LsCole was responsible for the immune response against Gram-negative and Gram-positive bacteria,while LsDef1 and LsDef2 were involved in insect defense against Gram-positive *** recombinant LsCole exhibited antimicrobial activities against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus ***2 showed an antibacterial effect against *** and LsDef2 exhibited antibiofilm activity against *** 2 AMPs disrupted cell membranes and caused leakage of *** cell *** results indicated that the 3 AMPs in *** are involved in the innate immunity of this pest *** AMPs may have potential as antimicrobial agents for bacterial infection ***,data are discussed in relation to new control strategies with greater biosafety against pest insects with use of microbial biocontrol agents in combination with RNA interference against the insect's defensive AMP genes.

MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population ***/threonine kinase Akt is an indispensable component of the insulin signaling *** of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect ***,the post-transcriptional pathway of LsAkt in *** remains *** study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of *** expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult *** reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of *** of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian *** also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of ***,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian *** of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt *** also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin *** results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of *** by targeting Akt in response to insulin *** data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

In vivo and in vitro antiproliferative and antimetastatic effects of hemolymph of Aspongopus chinensis Dallas on breast cancer cells

OBJECTIVE:To investigate the effects of Aspongopus(A.)chinensis hemolymph on the proliferation and metastasis of breast cancer ***:The in vitro effects of *** hemolymph were investigated in murine(4T1)and human(HCC1937)breast cancer cell ***,cell apoptosis,and cell migration were evaluated by using the cell counting kit-8 assay,Hoechst staining,and wound healing experiments,respectively.A syngeneic mouse model was established to evaluate the in vivo effects of the hemolymph extract on tumor growth and *** body weight,tumor size,blood levels of function-related enzymes,and pathological features of the liver and kidney tissues were ***:The hemolymph of *** significantly inhibited in vitro tumor cell migration and viability while inducing ***,it inhibited in vivo tumor growth and metastasis with a minimal effect on mouse body weight,and did not induce liver or kidney ***:Our results suggested that the *** hemolymph has antitumorigenic properties,suggesting it has potential as a novel therapeutic option for the treatment of metastatic breast cancer.

Smurf participates in Helicoverpa armigera diapause by regulating the transforming growth factor-βsignaling pathway

Diapause,an important strategy used by insects to avoid adverse environments,is regulated by various cell signaling *** results of our previous studies demonstrated that the transforming growth factor-β(TGF-β)signaling pathway regulated pupal diapause in Helicoverpa armigera,which was accompanied by downregulation of proteins in TGF-β***,to date the mechanism underlying this phenomenon remains ***,we cloned the E3 ubiquitin ligases gene *** vitro experiments showed that Smurf directly bound to TGF-βreceptor type I(TGFβRI)and *** Smurf promoted ubiquitination of TGFβRI and Smad2,thereby downregulating their protein ***,silencing of the Smurf gene suppressed ubiquitination of TGFβRI and Smad2 thereby increasing their protein *** from in vivo co-immunoprecipitation assays revealed that the binding of Smurf to TGFβRI or Smad2 was stronger in diapause pupae than in nondiapause *** of Smurf inhibitor A01 into diapause pupae markedly upregulated expression of TGFβRI and Smad2 proteins,leading to resumption of development in diapause *** together,these findings suggested that ubiquitin ligase E3 Smurf participated in *** diapause by regulating TGF-βsignaling,and thus could be playing a crucial role in insect diapause.

Neuropeptides are crucial in regulation of a rich variety of developmental,physiological,and behavioral functions throughout the life cycle of *** an integrated approach of multiomics,we identified neuropeptide precursors in the greater wax moth Galleria mellonella,which is a harmful pest of honeybee hives with a worldwide ***,a total of 63 and 67 neuropeptide precursors were predicted and annotated in the *** genome and transcriptome,in which 40 neuropeptide precursors were confirmed in the *** ***,we identified 12 neuropeptide precursor genes present in *** but absent in honeybees,which may be potential novel pesticide target *** hives were contaminated with heavy metals such as lead,enabling its bioaccumulation in *** bodies through the food chain,we performed transcriptome sequencing to analyze the effects of Pb stress on the mRNA expression level of *** neuropeptide *** treatment by Pb,the expression of neuropeptide F1 was found to be significantly downregulated,implying that this neuropeptide might be associated with responding to the heavy metal stress in *** study comprehensively identified neuropeptide precursors in ***,and discussed the effects of heavy metals on insect neuropeptides,with the example of *** results are valuable for future elucidation of how neuropeptides regulate physiological functions in *** and contribute to our understanding of the insect's environmental plasticity and identify potential new biomarkers to assess heavy metal toxicity in insects.

A water mite species is described as new to science, Atractides （Tympanomegapus） aprojectus sp. nov. The male ofAtractides （Tympanomegapus） huaxiensis Yi ＆ Jin, 2010 is described for the first time from guizhou Province, China. The materials were collected during a survey in 2011 from the rivers in guizhou. The specimens are deposited in the Institute of Entomology, guizhou University （GUGC）.

First record of Cubachipteria(Acari:Oribatida:Achipteriidae) in China,with descriptions of two new species

The genus Cubachipteria of Achipteriidae is firstly reported from China. Two new species of C. pianmaensis sp. nov. and C. gaoligongensis sp. nov., are described here, which are collected on moss and leaf litter from Yunnan Province, China. These two species can be distinguished from all known species of Cubachipteria by the absence of interlamellar setae. A key to the known species of Cubachipteria is given. All type materials are deposited in guizhou University, Guiyang City, China （GUGC）.

The Effects of Fertilizers on Rabbiteye Blueberry(Vaccinium ashei Reade.)Root Distribution and Fruit Yield

The root system plays an important role in the growth and development of *** aim of this study was to assess the impacts of different fertilizers on the root growth and root–yield relationship of blueberry to provide insight into the regulation of root growth and fruit yield by fertilizing from the perspective of the root *** blueberry variety‘Britewell’as the test material,and six fertilizers,including BF,OR,CF,SF,HF,and RT were used in single-factor fertilization experiments to analyze the effects of different fertilizer treatments on the root morphology,root distribution,and fruit yield of *** overall increased the root length density and root surface area in most soil layers,and the RT treatment significantly increased the total root length density and total root surface area 98.6%and 98.5%,respectively,compared with a control lacking *** addition,the effect of fertilization on the blueberry root system was mainly observed in the 0–20 cm *** yield was positively correlated with total root length density and total root surface area,and negatively correlated with average root *** summary,the SF and RT treatments increased the morphological indexes of the root system,particularly in the shallow soil layers,leading to an increase in blueberry fruit yield.

Disruption of a microvitellogenin gene impairs eggshell formation in Mythimna separata

Microvitellogenin(mVg)belongs to the lepidopteran-specific lipoprotein_11 super family and is thought to be involved in insect ***,the function of mVg in reproduction has not been *** this study,we identified and characterized MsmVg in Mythimna separata,a destructive insect pest of grain *** is comprised of a 1,310 bp open reading frame(ORF)encoding 437 amino *** analysis showed that MsmVg contains three conserved domains in the lipoprotein_11 super family,including the all-α-N-terminal domain(NTD),a Pseudomonas putida homologous domain(PPD),and an all-β-C-terminal domain(CTD).MsmVg was highly expressed in mature female adults and enriched in the ovaries of 3-day-old female *** of MsmVg by RNA interference did not change the expression of MsmVg,vitellogenin(MsVg)or the vitellogenin receptor(MsVgR)in fat bodies,but it inhibited their transcription in *** analysis showed that knockdown of MsmVg did not affect yolk deposition in ovaries,but impaired eggshell formation and led to a reduction in the reproductive ***,knockdown of MsmVg significantly inhibited the expression of genes associated with eggshell formation(MsChP1-3 and MsFcP3C).Taken together,these data suggest that MsmVg plays an important role in female reproduction by affecting eggshell formation in ***,which provides a potential target for managing this insect pest.
