Neuropeptides are crucial in regulation of a rich variety of developmental,physiological,and behavioral functions throughout the life cycle of *** an integrated approach of multiomics,we identified neuropeptide precursors in the greater wax moth Galleria mellonella,which is a harmful pest of honeybee hives with a worldwide ***,a total of 63 and 67 neuropeptide precursors were predicted and annotated in the *** genome and transcriptome,in which 40 neuropeptide precursors were confirmed in the *** ***,we identified 12 neuropeptide precursor genes present in *** but absent in honeybees,which may be potential novel pesticide target *** hives were contaminated with heavy metals such as lead,enabling its bioaccumulation in *** bodies through the food chain,we performed transcriptome sequencing to analyze the effects of Pb stress on the mRNA expression level of *** neuropeptide *** treatment by Pb,the expression of neuropeptide F1 was found to be significantly downregulated,implying that this neuropeptide might be associated with responding to the heavy metal stress in *** study comprehensively identified neuropeptide precursors in ***,and discussed the effects of heavy metals on insect neuropeptides,with the example of *** results are valuable for future elucidation of how neuropeptides regulate physiological functions in *** and contribute to our understanding of the insect's environmental plasticity and identify potential new biomarkers to assess heavy metal toxicity in insects.

Identification and immune analysis of antimicrobial peptides from the cigarette beetle(Lasioderma serricorne)

Antimicrobial peptides(AMPs)in insects are endogenous peptides that are effector components of the innate defense system of the *** may serve as antimicrobial agents because of their small molecular weight and broad-spectrum antimicrobial *** this study,we performed transcriptome analysis of cigarette beetle(Lasioderma serricorne)larvae,parasitized by the ectoparasitic wasp,Anisopteromalus *** AMP genes were significantly upregulated following *** parasitism,postulating the hypothesis that the parasitization enhanced the host's resistance against pathogenic microorganisms through the regulation of host AMP ***,3 AMP genes(LsDef1,LsDef2,and LsCole)were significantly upregulated and we studied their immune function in *** challenge and functional analysis showed that LsCole was responsible for the immune response against Gram-negative and Gram-positive bacteria,while LsDef1 and LsDef2 were involved in insect defense against Gram-positive *** recombinant LsCole exhibited antimicrobial activities against the Gram-negative bacterium Escherichia coli and the Gram-positive bacterium Staphylococcus ***2 showed an antibacterial effect against *** and LsDef2 exhibited antibiofilm activity against *** 2 AMPs disrupted cell membranes and caused leakage of *** cell *** results indicated that the 3 AMPs in *** are involved in the innate immunity of this pest *** AMPs may have potential as antimicrobial agents for bacterial infection ***,data are discussed in relation to new control strategies with greater biosafety against pest insects with use of microbial biocontrol agents in combination with RNA interference against the insect's defensive AMP genes.

Evaluation of Resistance of Different Kiwifruit Varieties (Lines) to Canker Disease and Brown Spot Disease

Kiwifruit canker and brown spot are signiﬁcant diseases affecting kiwis,caused by Pseudomonas syringae patho-genic variations(Pseudomonas syringae ***(Psa))and Corynesporapolytica(Corynespora cassiicola).At present,the research on canker disease and brown spot disease mainly focuses on the isolation and identiﬁcation of pathogenic bacteria,drug control,resistance gene mining and functional veriﬁ*** has proved that breeding disease resistant varieties are an effective method to control canker disease and brown spot ***,most existing cultivars lack genes for canker and brown spot *** kiwifruit resources in nat-ure exhibit extensive genetic diversity due to prolonged natural selection,containing numerous resistance ***,due to insufﬁcient understanding of the resistance of most kiwifruit varieties(lines)to canker disease and brown spot disease,some high-quality resources have not been fully *** incidence of canker and brown spot of 18 kiwifruit cultivars(lines)was measured by inoculating isolated branches and leaves,and their resistance to canker and brown spot was analyzed according to the length,disease index,mean diameter,and systematic *** results were as follows:Among 18 different kiwifruit varieties(lines)for canker disease,there were two highly resistant materials,eight disease-resistant materials,four disease-susceptible materials,and two highly susceptible ***,regarding brown spot disease,there were one highly resistant material,ﬁve dis-ease-resistant materials,four susceptible materials,and three highly susceptible ***,four resources were resistant to both *** outcomes provided a theoretical basis for breeding kiwifruit against canker and brown spot.

MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population ***/threonine kinase Akt is an indispensable component of the insulin signaling *** of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect ***,the post-transcriptional pathway of LsAkt in *** remains *** study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of *** expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult *** reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of *** of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian *** also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of ***,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian *** of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt *** also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin *** results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of *** by targeting Akt in response to insulin *** data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

Identification and Analysis of the WRKY Transcription Factor Gene Family in Verbena bonariensis

The WRKY transcription factor gene family is one of the unique gene families in *** plays an important role in response to abiotic stresses such as cold and drought,hormone signal transduction,regulation of biosynthesis,leaf senescence seed germination,***,little information is available about WRKY transcription factors in Verbena *** this study,70 VbWRKY genes were identified from the whole *** phylogenetic analysis of the WRKY gene family in *** and Arabidopsis shows that the WRKY genes in *** can be divided into three groups:I,II,and III,which contain 13,47,and 10 members,*** II can be further divided into five subclasses:IIa(5),IIb(10),IIc(18),IId(6),and IIe(8).Conservative motif analysis showed that 64 proteins encoded by the VbWRKY gene had conserved motifs 1,2 and 3,and the same subclass motif elements were approximately the *** collinearity analysis showed that there were 44 homologous gene pairs among the VbWRKYs,and these homologous gene pairs may have the same *** sequence analysis showed that the VbWRKY gene has multiple cis-acting elements,including not only cis-acting elements related to low-temperature and light responses,but also cis-acting elements related to hormone regulation,Among them,most VbWRKY genes contain response elements about low-temperature,and 30 VbWRKY genes contain low-temperature response elements(LTR),and 61 VbWRKY genes contain abscisic acid response elements(ABRE),indicating that VbWRKY plays a crucial role in plant growth and abiotic *** to the expression of VbWRKY in the cold stress and different tissues transcriptome,70 VbWRKY genes played their respective roles in various tissues and stages to regulate plant growth,Also,some of them participated in the process of cold stress tolerance,52 VbWRKYs showed significant differences in expression under cold stress,and 37 VbWRKY genes were up-regulated under cold stress.9 VbWRKY genes w

Disruption of a microvitellogenin gene impairs eggshell formation in Mythimna separata

Microvitellogenin(mVg) belongs to the lepidopteran-specific lipoprotein＿11 super family and is thought to be involved in insect reproduction. However, the function of mVg in reproduction has not been established. In this study, we identified and characterized MsmVg in Mythimna separata, a destructive insect pest of grain crops. MsmVg is comprised of a 1,310 bp open reading frame(ORF) encoding 437 amino acids. Structural analysis showed that MsmVg contains three conserved domains in the lipoprotein＿11 super family, including the all-α-N-terminal domain(NTD), a Pseudomonas putida homologous domain(PPD), and an all-β-C-terminal domain(CTD). MsmVg was highly expressed in mature female adults and enriched in the ovaries of 3-day-old female adults. Disruption of MsmVg by RNA interference did not change the expression of MsmVg, vitellogenin(MsVg) or the vitellogenin receptor(MsVgR) in fat bodies, but it inhibited their transcription in ovaries. Phenotype analysis showed that knockdown of MsmVg did not affect yolk deposition in ovaries, but impaired eggshell formation and led to a reduction in the reproductive capacity. Furthermore, knockdown of MsmVg significantly inhibited the expression of genes associated with eggshell formation(MsChP1-3 and MsFcP3C). Taken together, these data suggest that MsmVg plays an important role in female reproduction by affecting eggshell formation in M. separata, which provides a potential target for managing this insect pest.

Detection of Ochratoxin A and Human Exposure Risk Assessment in Rasa roxburghii by SPE-UHPLC-MS/MS

SPE-UHPLC-MS/MS was firstly used to evaluate the contamination of ochratoxin A (OTA) in Rasa roxburghii and firstly assessed the human exposure risk of OTA in Rasa roxburghii. A more suitable method for OTA extraction and purification of R. roxburghii was obtained， 25 mL of R. roxburghii juice was treated with enzymatic hydrolysis at a concentration of 0.06 mg/mL. The resulting mixture was filtered and concentrated to dry， then redissolved with 0.2 mL of methanol and diluted with 0.4 mL of ultra-pure water. The activated HLB column， which was added the sample solution， was washed with 6 mL of ultra-pure water and then eluted with 6 mL of methanol to purify. The eluent was collected and dried by nitrogen at 40 ℃， then redissolved in 1 mL of methanol and filtered for detection. The hazard quotient (HQ) values of all samples ranged from 0.077% to 35.792%， which were within the allowable limits for human consumption. But from the perspective of OTA contamination， we recommend the max stored time were 14 days at room temperature， 35 days at 4 ℃ and 63 days at -20 ℃ for R. roxburghii fruit， and 7 days for sealed storage and the same-day for open storage at room temperature， 14 days for sealed storage and 7 days for open storage at 4 ℃， and 63 days for sealed storage and 56 days for open storage at -20 ℃ for R. roxburghii juice. And all thirty samples randomly sampled from the market were OTA negative. The results of this study can lay a foundation for the formulation of OTA limit standards in fruits and juice in the future， and provide a reference for consumers to consume R. roxburghii more healthily.

Transcriptome and proteome analysis reveals the characteristics of carbon metabolism in the mycelial development of Gomphus purpuraceus

Gomphus purpuraceus is an ancient wild edible mushroom. However， the niche and lignocellulose degradation system of G. purpuraceus have remained cryptic. In the current study， transcriptome and proteome sequencing of G. purpuraceus in four mediums was performed. A total of 3489 differentially expressed genes (DEGs) and 891 differentially expressed proteins (DEPs) were screened out. For the lignocellulose degradation system， a total of 61 expressed genes were annotated， including 19 genes encoding cellulase， 8 genes encoding hemicellulase， 5 genes encoding pectinase， 22 genes encoding lignin oxidase， and 7 lignin degrading auxiliary enzyme. From two-omics， morphology， and enzyme activities， we speculated that G. purpuraceus might have a limited capacity to degrade polysaccharides (starch and lignocellulose). In addition， trace amount of fumonisins (FB1 and FB2) were found in the G. purpuraceus. These results provided a new perspective for understanding the key pathways and hub genes involved in carbon metabolism of G. purpuraceus development.
