Effects of the concentration and nature of total dissolved solids in drinking water on feed intake, nutrient digestion, energy balance,methane emission, ruminal fermentation, and blood constituents in different breeds of young goats and hair sheep

Understanding how different livestock species and breeds respond to consumption of brackish water could improve usage of this resource.Therefore,Angora,Boer,and Spanish goat doelings and Dorper,Katahdin,and St.Croix ewe lambs (6 animals per animal type[AT];initial age=296±2.1 days)consuming water with varying concentrations of minerals of a natural brackish water source (BR) and sodium chloride (NaCl;SL) were used to determine effects on water and feed intake,nutrient digestion,heat energy,methane emission,ruminal fluid conditions,and blood constituent concentrations.There were 6 simultaneous 6 (water treatments[WT])×6 (AT) Latin squares with 3-wk periods.The WT were fresh (FR),BR alone (100-BR),a similar total dissolved solids (TDS) concentration as 100-BR via NaCl addition to FR (100-SL),BR with concentrations of all minerals increased by approximately 50%(150-BR),a similar TDS level as 150-BR by NaCl addition to FR (150-SL),and a similar 150 TDS level achieved by addition of a 1:1 mixture of BR minerals and NaCl to 100-BR (150-BR/SL).Concentrations (mg/kg) in BR were 4928 TDS,85.9 bicarbonate,224.9 calcium,1175 chloride,60.5 magnesium,4.59 potassium,1387sodium,1962 sulfate,and 8.3 boron,and TDS in other WT were 209,5684,7508,8309,and 7319 mg/kg for FR,100-SL,150-BR,150-SL,and 150-BR/SL,respectively.There were very few significant effects of WT or AT×WT interactions,although AT had numerous effects.Water intake was affected by AT (P=0.02)and WT (P=0.04),with greater water intake for 150-SL than for FR,100-BR,100-SL,and 150-BR.Dry matter intake among AT was lowest (P<0.05) for Angora.Digestion of organic matter and neutral detergent fiber and heat energy differed among AT (P<0.05),but nitrogen digestion and ruminal methane emission were similar among AT.Blood aldosterone concentration was higher (P<0.05) for FR than for other WT.In conclusion,all AT seemed resilient to these WT regardless of mineral source and concentrations,with TDS less than 8300 mg/kg,which did not influence nutrient utilization,ruminal ferme

Globally,the prevalence of anxiety and depression has reached epidemic proportions.food-derived protein hydrolysates and peptides delivered through dietary supplementation can avoid the negative risks associated with traditional pharmaceuticals while delivering superior anxiolytic and antidepressant effects.This review summarizes current research on food-derived anxiolytic and antidepressant protein hydrolysates and peptides,and subsequently analyses their physicochemical characteristics and elaborates on their mechanisms.The aim of this work is to contribute to the in-depth study and provide a theoretical foundation for the development of related products to better serve patients with anxiety and depression.

Liberica coffee as an alternative cropping system for sustainable farming on Indonesian Peatlands

Indonesian peatlands are a fragile ecosystem,and to protect it,growing Liberica coffee is a promising way for both the environment and the economy.This study aimed to evaluate the performance of the liberica coffee on peatland with different water tables and develop an improved cultivation system in this ecosystem.The study area was in Tanjung Jabung Barat District,Jambi Province,Indonesia.The plant age is greater than 15 years and the average plant density was about 1000 trees/hm2.The study was conducted in two stages.The 1st stage was a survey to identify and characterize smallholder liberica coffee farming at three peatland zones,namely 0-100 m,200-300 m,and>400 m from the principal drainage canal,and the 2nd stage was a field experiment to test the effectiveness of amendments in improving liberica coffee growth and improving degraded peatland.The treatments were arranged in a randomized complete block design with four replications,including manure(M),at the rate of 10 t/hm^(2);lime(L),3 t/hm^(2);peat surface elevation(P),and a farmer’s practice(Control)without manure and/or lime as a control.This research revealed that liberica coffee can not tolerate high water table as it may stimulate disease incidence and cause low-quality yield.However,recycling of organic C of about 17.14 t/(hm^(2)·a)through cherry residue application and litter compensated part of the loss of organic C through CO_(2)emissions,coffee bean removal,and maintaining peat fertility for sustainable farming.The low yield of(0.70±0.12)t/(hm^(2)∙a)coffee bean could be increased to(0.87±0.24)and(0.94±0.14)t/(hm^(2)∙a)by adding 3 t/(hm^(2)∙3a)of lime or 10 t/(hm^(2)∙a)of manure,respectively.This research revealed that water table management and amendments are two main factors in liberica coffee farming on peatlands.It is of great significance to study the cultivation technology of coffee in peatland.

Differences in the effects and action modes of gut commensals against dextran sulfate sodium-induced intestinal inflammation

Inflammatory bowel disease(IBD)is a complex relapsing inflammatory disease in the gut and is driven by complicated host-gut microbiome interactions.Gut commensals have shown different functions in IBD prevention and treatment.To gain a mechanistic understanding of how different commensals affect intestinal inflammation,we compared the protective effects of 6 probiotics(belonging to the genera Akkermansia,Bifidobacterium,Clostridium,and Enterococcus)on dextran sulfate sodium(DSS)-induced colitis in mice with or without gut microbiota.Anti-inflammatory properties(ratio of interleukin(IL)-10 and IL-12)of these strains were also evaluated in an in vitro mesenteric lymph nodes(MLN)co-culture system.Results showed that 4 probiotics(belonging to the species Bifidobacterium breve,Bifidobacterium bifidum,and Enterococcus faecalis)can alleviate colitis in normal mice.The probiotic strains differed in regulating the intestinal microbiota,cytokines(IL-10,IL-1βand interferon(IFN)-γ),and tight junction function(Zonulin-1 and Occludin).By constrast,Akkermansia muciniphila AH39 and Clostridium butyricum FHuNHHMY49T1 were not protective.Interestingly,B.breve JSNJJNM2 with high anti-inflammatory potential in the MLN model could relieve colitis symptoms in antibiotic cocktail(Abx)-treated mice.Meanwhile,E.faecalis FJSWX25M1induced low levels of cytokines in vitro and showed no beneficial effects.Therefore,we provided insight into the clinical application of probiotics in IBD treatment.

4-Hydroxy-2-Oxoglutaric Acid,A KeyMetabolite Involved in Trypsin-Regulation of Arginine Metabolism in Hylocereus undatus during Storage

Trypsin,a novel superoxide scavenger,significantly enhances the storage quality of Hylocereus undatus(H.undatus).To elucidate the preservation mechanism of trypsin on H.undatus,a widely targeted metabolomic analysis,and transcriptomics analysis were conducted.Firstly,a total of 453 metabolites were identified,with organic acids and their derivatives constituting the largest proportion(25%).Amino acids and their metabolites,prominent among organic acids,were further analyzed.Among them,73 metabolites were associated with amino acids,and 37 exhibited significant differences.The most enriched Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway was arginine biosynthesis(map00220),with polyamine metabolites showing the most pronounced differences,particularly spermine(FC=1.7594).Compared with the control group,4-hydroxy-2-oxoglutaric acid was significantly upregulated(FC=2.117)in the process of spermine biosynthesis.Furthermore,the results of Gene Ontology(GO)and KEGG enrichment analysis of the H.undatus transcriptome profile revealed that trypsin treatment led to 187 differentially expressed genes associated with arginine.Both GO and KEGG analyses exhibited significant enrichment in the spermine biosynthetic process(GO:0006597)(map:00220)within the arginine biosynthesis pathway.Moreover,most enzymes and metabolites within the spermine biosynthesis pathway in H.undatus were upregulated.The results of the PPI network highlighted that ADC,SPDS,and SAMDC,among others,were pivotal proteins involved in trypsin-regulated arginine metabolism and spermine synthesis.This study revealed that trypsin could significantly delay postharvest senescence of H.undatus at room temperature.This effect might be attributed to trypsin triggering the synthesis of 4-hydroxy-2-oxoglutaric acid in the fruit peel,thereby promoting the biosynthesis of spermine and other polyamines.

Determining minimum sample size for the conditioned Latin hypercube sampling algorithm

In digital soil mapping(DSM),a fundamental assumption is that the spatial variability of the target variable can be explained by the predictors or environmental covariates.Strategies to adequately sample the predictors have been well documented,with the conditioned Latin hypercube sampling(cLHS)algorithm receiving the most attention in the DSM community.Despite advances in sampling design,a critical gap remains in determining the number of samples required for DSM projects.We propose a simple workflow and function coded in R language to determine the minimum sample size for the cLHS algorithm based on histograms of the predictor variables using the Freedman-Diaconis rule for determining optimal bin width.Data preprocessing was included to correct for multimodal and non-normally distributed data,as these can affect sample size determination from the histogram.Based on a user-selected quantile range(QR)for the sample plan,the densities of the histogram bins at the upper and lower bounds of the QR were used as a scaling factor to determine minimum sample size.This technique was applied to a field-scale set of environmental covariates for a well-sampled agricultural study site near guelph,Ontario,Canada,and tested across a range of QRs.The results showed increasing minimum sample size with an increase in the QR selected.Minimum sample size increased from 44 to 83 when the QR increased from 50% to 95% and then increased exponentially to 194 for the 99%QR.This technique provides an estimate of minimum sample size that can be used as an input to the cLHS algorithm.

Effect of L-theanine combined with geniposide on depression-like behavior induced by the chronic unpredictable mild stress(CUMS) in mice

Depression is a prevalent mental disorder with limited effective treatments， posing a significant global issue. This study explored L-theanine and geniposide， key components in "food-medicine homology" materials， to determine if their combination (TG) could alleviate depression-like behaviors and hippocampal neuronal damage in a chronic unpredictable mild stress (CUMS) mouse model. Male C57BL/6J mice were divided into control， CUMS model， and CUMS+TG groups with varying doses. The CUMS group displayed depression-like behaviors， including reduced activity and sucrose preference. TG treatment partially reversed these changes， significantly increasing antioxidant enzyme activities， decreasing pro-inflammatory cytokine levels， and improving neuromodulator levels. RNA-seq analysis identified the transthyretin (TTR) gene， upregulated in the model group but downregulated after TG treatment. TG treatment modulated intestinal microbiota composition compared to the CUMS group， including increased Firmicutes， reduced Bacteroidetes and Prevotella， and variable changes in Bifidobacterium abundance. In conclusion， our study indicates that CUMS exposure upregulates stress hormones and TTR expression， associated with neuroinflammation， oxidative stress， monoamine depletion， depression-like behaviors， and intestinal microbiota dysbiosis. TG treatment alleviates these effects and modulates intestinal microbiota， suggesting L-theanine and geniposide’s potential as a novel depression therapy.

Alpha-lactalbumin(α-LA)is a major whey protein found in breast milk and plays a crucial role in the growth and development of infants.In this study,Bacillus subtilis RIK1285 harboring AprE signal peptide(SP)was selected as the original strain for the production ofα-LA.It was found thatα-LA was identified in the pellet after ultrasonic disruption and centrifugation instead of in the fermentation supernatant.The original strain most likely only producedα-LA intracellular,but not extracellular.To improve the expression and secretion ofα-LA in RIK1285,a library of 173 homologous SPs from the B.subtilis 168 genome was fused with target LALBA gene in the pBE-S vector and expressed extracellularly in RIK1285.SP YjcN was determined to be the best signal peptide.Bands in supernatant were observed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and purified by nickel column to calculate the highest yield signal peptide.In addition,different promoters(P_(aprE),P_(43),and P_(glv))were compared and applied.The results indicated that the strain RIK1285-pBE-P_(glv)-YjcN-LALBA had the highestα-LA yield,reaching 122.04μg/mL.This study demonstrates successful expression and secretion of humanα-LA in B.subtilis and establishes a foundation for simulating breast milk for infant formulas and developing bioengineered milk.

Blautia producta displays potential probiotic properties against dextran sulfate sodium-induced colitis in mice

Blautia has attracted attention because of its potential efficacy in ameliorating host energy metabolism and inflammation.This study aims to investigate the influences of Blautia producta D4 on colitis induced by dextran sulfate sodium(DSS)and to reveal the underlying mechanisms.Results showed that B.producta D4 intervention significantly relieved body weight loss,and suppressed the elevation of pro-inflammatory cytokines(including interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β))and excessive oxidative stress(myeloperoxidease(MPO)activity,superoxide dismutase(SOD)activity,glutathione peroxidase(GSH-Px)activity,and malondialdehyde(MDA)level)in colitis mice.Moreover,the concentrations of tight junction proteins(occludin,claudin-1,and ZO-1)related to the intestinal barrier were obviously elevated,and colitis-related TLR4/NF-κB pathway activation was remarkably inhibited after B.producta D4 intervention.The intestinal microbial disorder was evidently ameliorated by increasing the relative abundance of Clostridium sensu stricto 1,Bifidobacterium,GCA-900066225,Enterorhabdus,and reducing the relative abundance of Lachnospiraceae NK4A136 group.In conclusion,oral administration of B.producta D4 could ameliorate DSS-induced colitis by suppressing inflammatory responses,maintaining the intestinal barrier,inhibiting TLR4/NF-κB pathway,and regulating intestinal microbiota balance.These results are conducive to accelerate the development of B.producta D4 as a functional probiotic for colitis.

Akkermansia muciniphila-directed polyphenol chlorogenic acid intervention for obesity in mice

Akkermansia muciniphila play an important in ameliorating obesity but is not allowed for direct consumption in most countries.To date,microbiota-directed foods selectively promote the targeted human gut microbes,providing a strategy for A.muciniphila enhancement.Multiple studies have indicated the potential regulation of the polyphenol on A.muciniphila.Therefore,a polyphenol screening based on A.muciniphila upregulation was performed in mice.Chlorogenic acid(CGA)exhibited a greater response to A.muciniphila upregulation.Furthermore,we found that CGA did not directly promote A.muciniphila growth or mucin secretion.Microbiome and metabolomics revealed that the increased abundance of A.muciniphila resulted from the inhibition of CGA on Desulfovibrio and Alistipes and the influence of docosahexaenoic acid,β-hydroxybutyrate,and N-acetyl-lactosamine.Finally,to confirm the regulation of CGA on A.muciniphila under disease conditions,high-fat diet-fed mice were established.The results showed CGA promoted A.muciniphila growth,and we expectedly found that CGA suppressed the augment in body weight of mice,significantly attenuated adipose tissue abnormality,provided liver protection and improved gut barrier integrity.These results suggest that CGA inhibits the development of obesity.Overall,our results indicate that microbiota-directed food is a promising approach for the treatment of obesity.
