Effects of sampling strategies and DNA extraction methods on eDNA metabarcoding: A case study of estuarine fish diversity monitoring

Environmental DNA(eDNA)integrated with metabarcoding is a promising and powerful tool for species composition and biodiversity assessment in aquatic ecosystems and is increasingly applied to evaluate fish diversity.To date,however,no standardized eDNA-based protocol has been established to monitor fish diversity.In this study,we investigated and compared two filtration methods and three DNA extraction methods using three filtration water volumes to determine a suitable approach for eDNA-based fish diversity monitoring in the Pearl River Estuary(PRE),a highly anthropogenically disturbed estuarine ecosystem.Compared to filtration-based precipitation,direct filtration was a more suitable method for eDNA metabarcoding in the PRE.The combined use of DNeasy Blood and Tissue Kit(BT)and traditional phenol/chloroform(PC)extraction produced higher DNA yields,amplicon sequence variants(ASVs),and Shannon diversity indices,and generated more homogeneous and consistent community composition among replicates.Compared to the other combined protocols,the PC and BT methods obtained better species detection,higher fish diversity,and greater consistency for the filtration water volumes of 1000 and 2000 mL,respectively.All eDNA metabarcoding protocols were more sensitive than bottom trawling in the PRE fish surveys and combining two techniques yielded greater taxonomic diversity.Furthermore,combining traditional methods with eDNA analysis enhanced accuracy.These results indicate that methodological decisions related to eDNA metabarcoding should be made with caution for fish community monitoring in estuarine ecosystems.

Fracture distribution in overburden strata induced by underground mining

Coal-mining activities give rise to a series of ecological environmental problems,such as ground settlement and groundwater pollution.In fact,they are mainly caused by mining-induced fractures.Hence,it is necessary to study the mining-induced fracture distribution to identify the behavior of rock mass movement.However,the fractures in overburden strata cannot be directly measured owing to the special condition.Therefore,the majority of previous studies are based on experiments or experience.For this reason,this study first used a discrete element method to simulate the shape of mining-induced fractures in overburden strata.Then,a geophysical tool of transient electromagnetic method(TEM)was used to investigate the mining-induced fracture distribution.Based on the low-resistivity anomaly area,the water-rich area in overburden strata was analyzed to be mainly caused by fracture seepage.Through the mutual authentication between numerical simulation and TEM results,the mining-induced fractures in overburden strata were explored.This study can enhance the understanding of mining-induced fracture distribution on the one hand and guarantee the coal mining safety on the other,thus guiding the coordinated development between coal mining and environmental protection.

The development of sericultural industry in the Pearl River Delta has contributed greatly to good ecological environment and economic prosperity of the people’s livelihood.Comprehensive utilization of sericulture resources can turn waste into valuable resources and expand the development approaches of sericultural industry.For example,using abundant silkworm excrement to produce chlorophyll is one of the good practices in comprehensive utilization of sericulture resources,which has leading technologies and good economic benefits.In addition to chlorophyll,the products produced from silkworm excrement can be used as the main raw materials of medicine,food,plant growth regulator and cosmetics.For instances,phytol,the important raw material for synthesis of vitamin E and vitamin K,was extracted from silkworm excrement,enabling China to become an important production base of vitamin E and vitamin K.Triacontanol extracted from silkworm excrement is a new crop growth regulator,which can increase the yield of rice and vegetable.Pectin,dietary fiber and carotenoid extracted from silkworm excrement as well can be used as natural raw material in many fields.China is the biggest manufacturer and supplier of silkworm excrement products in the world,which has a development history of about 50 years and generated high export revenue.It is believed that close combination of academic research and industry development is the only way to promote the industrial development of silkworm excrement resources.
