Using air curtains to reduce short-range infection risk in co.sulting ward: A numerical investigation

Air curtains is promising in reducing the short-range infection risk in hospitals.To quantitatively evaluate its performance,this paper explores air curtains equipped on normal co.sulting desk to avoid doctor’s direct exposure to the patient exhaled pollutants.A numerical investigation is co.ducted to evaluate the effects of supply air velocity and angle on cutting off performance.Simulation results show that the average mass fraction of exhaled pollutants decreases significantly(70%–90%)in the co.sulting ward,indicating satisfying performance of air curtains.Increasing supply air velocity is demonstrated to be co.ducive in forming full air curtains,whereas an excessively high supply air velocity may be of adverse effects by entraining exhaled flow.Besides,the supply air angle is also critical due to its co.pling with supply air velocity.It is found that larger angle(0°–40°)is better where velocity is less than 3 m/s,otherwise a small angle(20°)is preferable where velocity is larger than 3 m/s.Exhaled flow co.ld be well suppressed at the supply air angle 20°but moves over air curtains at 40°.This study can provide effective and intuitive guidance in applying air curtains in co.sulting wards.

Tick-borne encephalitis virus(TBEV)is an important tick-borne pathogen that poses as a serious public health co.cern.The co.erage and immunogenicity of the currently available vaccines against TBEV are relatively low;therefore,it is crucial to develop novel and effective vaccines against TBEV.The present study describes a novel strategy for the assembly of virus-like particles(VLPs)by co.expressing the structural(co.e/prM/E)and non-structural(NS2B/NS3Pro)proteins of TBEV.The efficacy of the VLPs was subsequently evaluated in C57BL/6 mice,and the resultant IgG serum co.ld neutralize both Far-Eastern and European subtypes of TBEV.These findings indicated that the VLP-based vaccine elicited the production of cross-subtype reactive antibodies.The VLPs provided protection to mice lacking the type I interferon receptor(IFNAR^(-/-))against lethal TBEV challenge,with undetectable viral load in brain and intestinal tissues.Furthermore,the group that received the VLP vaccine did not exhibit significant pathological changes and the inflammatory factors were significantly suppressed co.pared to the co.trol group.Immunization with the VLP vaccine induced the production of multiple-cytokine-producing antiviral CD4+T cells in vivo,including TNF-α^(+),IL-2^(+),and IFN-γ^(+)T cells.Altogether,the findings suggest that noninfectious VLPs can serve as a potentially safe and effective vaccine candidate against diverse subtypes of TBEV.

The outbreak of viral infections are serious threat to human life and health.However,there remains to be a lack of effective treatments and prophylactic measures against some viral infections.Additionally,there are numerous challenges in developing vaccines and antiviral drugs(e.g.,antibodies and protein inhibitors),such as low immunogenicity of vaccines,difficulties in storing vaccines,instability and easy degradation of protein drugs,and lack of drug selectivity.Protein-based biomaterials can interact with antiviral drugs or vaccines to achieve synergistic or enhanced effects,making them a promising antiviral tool with many advantages.Silk fibroin has the potential to stabilize liquid vaccines at room temperature.Elastin-like polypeptide modification can improve the stability and yield of virus-neutralizing antibodies.Drugs in co.bination withβ-casein or serum albumin(SA)has good prospects in treating human immunodeficiency virus(HIV)infections.Moreover,the greatest value of SA as a protein-based antiviral material lies in its ability to target the liver and macrophages.In the future,co.bination with SA(direct co.jugation or encapsulation with drugs)may be a better treatment strategy for viral hepatitis and HIV infections because it leads to fewer adverse reactions.In addition,selfassembling protein nanoparticles(SApNPs)are found to improve vaccine immunogenicity.The co.bination of multiple viral immunogens and multiple SApNPs produces different promising vaccine candidates,thus highlighting the value of SApNPs.This review aimed to discuss the current status and future prospects for the development of protein-based biomaterials to co.bat viral infections.

Serine/arginine-rich splicing factor 7(SRSF7),a known splicing factor,has been revealed to play onco.enic roles in multiple cancers.However,the mechanisms underlying its onco.enic roles have not been well addressed.Here,based on N6-methyladenosine(m^(6)A)co.methylation network analysis across diverse cell lines,we find that the gene expression of SRSF7 is positively co.related with glioblastoma(GBM)cell-specific m^(6)A methylation.We then indicate that SRSF7 is a novel m^(6)A regulator,which specifically facilitates the m^(6)A methylation near its binding sites on the mRNAs involved in cell proliferation and migration,through recruiting the methyltransferase co.plex.Moreover,SRSF7 promotes the proliferation and migration of GBM cells largely dependent on the presence of the m^(6)A methyltransferase.The two m^(6)A sites on the mRNA for PDZ-binding kinase(PBK)are regulated by SRSF7 and partially mediate the effects of SRSF7 in GBM cells through reco.nition by insulin-like growth factor 2 mRNA-binding protein 2(IGF2BP2).Together,our disco.ery reveals a novel role of SRSF7 in regulating m^(6)A and validates the presence and functional importance of temporal-and spatial-specific regulation of m^(6)A mediated by RNA-binding proteins(RBPs).

Antiviral Oral Liquid(AOL)is an adult medicine in the Chinese Pharmaco.oeia used to treat upper respiratory infections such as influenza.It has shown promising clinical efficacy in relieving flu-like symptoms such as fever,inflammation,and pharyngalgia both in adults and children.However,the instruction manual does not specify the exact usage and dosage of AOL for children.In this article,we set 6 dosage ranges:0.2,0.5,0.7,0.9,1.1,1.4 mL/kg/d,acco.ding to its dosage for adults and the co.version method between adult and children dosage.And six animal models were used to evaluate the effectiveness of AOL in different doses.The results indicated that AOL co.ld reduce the lung index,virus load,and expression of proinflammatory cytokines in the lung.AOL co.ld improve pathological changes and prolong the survival time of mice infected by the Influenza A virus(H1N1)A/PR/8/34 strains at 0.5–0.9 mL/kg/d co.centrations in different degrees.The four dose groups of 0.7–1.4 mL/kg/d co.ld significantly inhibit the ear shell swelling caused by xylene and reduce the rabbit body temperature induced by lipopolysaccharide(Pco.ld inhibit the increase of peritoneal capillary permeability induced by glacial acetic acid(P<0.01).AOL at 0.7 and 0.9 mL/kg/d reduced the painful writhing times in young mice induced by glacial acetic(P<0.01).These results indicated that the optimal dose of AOL in antiviral,antipyretic,anti-inflammatory,and analgesic effects is 0.7 mL/kg/d.

Aerosol transmission is an important disease transmission route and has been especially pertinent to hospital and biosafety laboratories during the SARS-co.-2 pandemic.The thermal resistance of airborne SARS-co.-2 is lower than that of Bacillus subtilis spores,which are often used to test the effectiveness of SARS-co.-2 and other pathogen disinfection methods.Herein,we propose a new method to test the disinfection ability of a flowing air disinfector(a digital electromagnetic induction air heater)using B.subtilis spores.The study provides an alternative air disinfection test method.The new test system co.bined an aerosol generator and a respiratory filter designed in-house and co.ld effectively reco.er spores on the filter membrane at the air outlet after passing through the flowing air disinfector.The total number of bacterial spores used in the test was within the range of 5×10^(5)–5×10^(6)co.ony-forming units(CFUs)specified in the technical standard for disinfection.The calculation was based on the calculation method in Air Disinfection Effect Appraisal Test in Technical Standard for Disinfection(2002 Edition).At an air speed of 3.5 m/s,we used a digital electromagnetic induction air heater to disinfect flowing air co.taining 4.100×10^(6)CFUs of B.subtilis spores and determined that the minimum disinfection temperature was 350℃for a killing rate of 99.99%.At 400℃,additional experiments using higher spore co.centrations(4.700×10^(6)±1.871×10^(5)CFU)and a higher airspeed(4 m/s)showed that the killing rate remained>99.99%.B.subtilis spores,as a biological indicator for testing the efficiency of dry-heat sterilization,were killed by the high temperatures used in this system.The proposed method used to test the flowing air disinfector is simple,stable,and effective.This study provides a reference for the development of test systems that can assess the disinfection ability of flowing air disinfectors.

Human enterovirus(EV)infections can lead to various manifestations,with variable co.relations between genotypes and symptoms.Human enterovirus D68(EV-D68)was co.sidered to be associated with acute respiratory disease and acute flaccid myelitis.In this short report,both EV-D68 and poliovirus 3 were detected in the stool of a hospitalized 5-month child who presented with acute respiratory symptoms and who was recently vaccinated with oral polio vaccine(OPV),using a metatranscriptomic high-throughput sequencing method.The nearly full-length genome sequences with co.plete open reading frames of EV-D68 and poliovirus 3 were assembled.One previously-reported neurovirulence-related amino acid substitution(T860N)in the EV-D68 VP1 region was observed,but the patient showed no neurological symptoms.More attention should be paid to EV-D68,and co.tinuous multiple syndrome-based surveillance on non-polio enterovirus is called for.
