AIM: To compare transforming growth factor-β1 (TGF- β1 ) levels in tears and the degree of corneal haze formation following epithelial laser in situ keratomileusis (Epi -LASIK) with and without the use of mitomycin C (MMC) and to investigate the effect of MMC on corneal wound healing. ·METHODS: Thirty-two patients (64 eyes) with high myopia underwent Epi -LASIK surgery, and MMC was randomly used in one eye in each patient. The epithelialization process was observed, and the TGF -β1 level in tears was measured at 1 day, 3, and 7 days postoperatively for comparison with baseline. Corneal haze was graded at 1 month, 3, and 6 months after surgery. · RESULTS: Mean preoperative spherical equivalent refraction was -8.24 ±2.18D (range-6.00 to -10.50D) in the MMC group and -7.82±1.55D (range -6.00 to -9.75D) in the non -MMC group. There was no significant difference between the two groups (P =0.38). Mean epithelialization time was (5.02 ±0.68) days in the MMC group and (4.86±0.57) days in the non-MMC group (P = 0.31). Tear fluid TGF-β1 levels were similar before surgery (P =0.34), but were significantly higher in the non-MMC group at 1 day, 3, and 7 days postoperatively (P =0.004, 0.008, and 0.012, respectively). Corneal haze scores 1 month after surgery were significantly higher in the non -MMC group (P =0.03), and similar at 3 and 6 months after surgery (P =0.28 and 0.62, respectively). ·CONCLUSION: MMC did not delay epithelialization. Inearly postoperative period, lower TGF -β1 levels in tears and a lower grade of corneal haze were observed in the MMC group. Our findings suggest that the ability of MMC to inhibit Epi -LASIK -induced haze might be mediated through TGF-β1 suppression.

AIM: To describe the clinical features of acute myopic onset of optic neuropathy and observe the effects of retrobulbar and systemic glucocorticoid therapy in a realworld ***: A retrospective observational case series included 18 patients with a clinical diagnosis of acute onset of myopic optic neuropathy in a real-world setting. While the patients were using retrobulbar and systemic glucocorticoid therapy, various imaging examination data were analysed, and the clinical features of myopic optic neuropathy were summarized for 6 mo to 2 y. RESULTS: The included group of patients with acute onset of myopic optic neuropathy consisted mostly of females(n=11). The visual field(VF) showed abnormalities in bilateral eyes, including the spread of physiological blind spots, central and paracentral dark spots, and centripetal peripheral VF reduction;but central vision with no subjective changes. The visual evoked potential(VEP) was abnormal in all eyes with vision loss. The best corrected visual acuity(BCVA) was improved from 1.04±0.63 to 0.47±0.57(log MAR) af ter glucocor ticoid treatment(P0.05). There was no recurrence during long-term followup ***: The acute onset of myopic optic neuropathy is characterized by BCVA and VF abnormalities in bilateral eyes. Retrobulbar and systemic glucocorticoid therapy is effective.

comitant esotropia(AACE).METHODS:A retrospective cohort study was conducted by analyzing 83 patients(case group)with AACE who underwent strabismus correction surgery from January 1,2021 to June 30,*** 73 outpatient volunteers were recruited during the same period as the normal control *** case group’s binocular vision time,near and distance esotropia angle,and near stereo vision function were recorded,and the age,gender,refractive status,and best-corrected visual acuity(BCVA)of both groups were ***,multiple logistic regression analysis was conducted using an eye usage condition questionnaire to determine the independent risk factors for ***:In the case group,61 patients(73.49%)had myopia,with a mean equivalent spherical power(SE)of-3.35±3.31 D(range:+2.75 to-10.62 D)of the right eye and-2.87±3.35 D(range:+2.75 to-11.12 D)of the left *** average duration of diplopia in the case group was 29.83±35.72mo,of which 80 patients(96.39%)were primarily with distance *** near and distance esotropia angle after wearing glasses were 52.36±20.95 prism degree(PD)and 56.71±19.54 PD,respectively,and there was no statistically significant difference between the two(t=1.38,P=0.169).The incidence of improper glasses wearing and unhealthy eye habits in the case group was significantly higher than those in the control group(P<0.05).Close-up work without glasses[β=2.30,odds ratio(OR)=10,95%confidence interval(CI)2.35-42.51,P=0.002]and near work in supine position(β=1.80,OR=6.02,95%CI 3.29-11.02,P<0.001)were independent risk factors for ***:Patients with AACE mainly present with distance diplopia,and there is a high degree of variation in *** work without wearing glasses and in supine position are independent risk factors for AACE.

AIM： To explore the state of autophagy and related mechanisms in the murine retinal microvascular endothelial cells（RMECs） under hypoxia ***： The murine RMECs were primarily cultured and randomly divided into three groups： hypoxia group（cultured in 1% O_2 environment）, hypoxia＋autophagy inhibition group [pretreated with 5 mmol/L 3-methyladenine（3-MA） for 4 h followed by incubation in 1% O_2] and control group（cultured under normoxic condition）. The state of autophagy in RMECs was examined by assaying the turnover of light chain 3 B（LC3BB） and expression of Beclin-1, Atg3 and Atg5 proteins with Western blotting, by detecting formation of autophagosomes with transmission electron microscopy（TEM） and by counting the number of GFP＋ puncta in RMECs. The protein levels of AMPK, P-AMPK, Akt, P-Akt, m-TOR and P-m TOR were also assayed by Western ***： Primary murine RMECs were successfully cultured. Under hypoxic conditions, the ratio of LC3BB-Ⅱ/Ⅰ and the expression of Beclin-1, Atg3 and Atg5 proteins were increased when compared with the control group. In addition, the numbers of autophagosome and the GFP＋ puncta were also increased under hypoxia. However, pretreatment with 3-MA obviously attenuated these changes in autophagy in RMECs under hypoxia. Protein expression of P-Akt and P-AMPK was increased but P-m TOR level was decreased in cells exposed to hypoxia. CONCLUSION： In murine RMECs autophagy is activated under hypoxia possibly through activation of the AMPK/mTOR signaling pathway.
