Efficacy of tanshinone ⅡA and mesenchymal stem cell treatment of learning and memory impairment in a rat model of vascular dementia

OBJECTIVE: To investigate the efficacy of administration of tanshinone Ⅱ A(TSA) combined with mesenchymal stem cells(MSCs) for the treatment of learning and memory impairment caused by vascular dementia(Va D) and to determine the underlying ***: Modified four-vessel occlusion was used to establish a Va D model in rats, and their spatial learning and memory capacity was assessed by the Morris water maze. The rats were randomized into MSCs, TSA, MSCs combined with TSA, vehicle and sham groups. Histological changes were determined by hematoxylin and eosin staining, and the hippocampal neuron apoptosis ratio was assessed by flow cytometry. Western blotting was performed to detect Bcl-2 and Bax expression. The reactive oxidative species(ROS) levels and the activity of total superoxide dismutase(T-SOD), an antioxidant enzyme in the rat hippocampus, were ***: TSA combined with MSCs treatment administered by intravenous injection in the tail significantly attenuated cognitive deficits in the Va D model compared with the vehicle group(P < 0.01),and its protective effect on cognitive function was greater than that obtained by treatment with MSCs or TSA alone. Furthermore, TSA combined with MSCs treatment achieved synergistic effects in suppressing neuronal apoptosis in the rat hippocampus caused by global brain ischemia via up-regulating the expression of Bcl-2, an anti-apoptosis protein, and decreasing the expression of Bax, a pro-apoptotic protein. In addition, TSA combined with MSCs treatment attenuated ROS production and enhanced T-SOD activity in the rat hippocampus, and the antioxidant effect was greater than that of treatment with MSCs or TSA ***: TSA combined with MSCs treatment improved the spatial learning and memory capacity in a Va D model via suppressing neuronal apoptosis and antioxidant activity in the hippocampus, and this improvement was greater with combined treatment than with treatment with MSCs or TSA alone.

AIM： To discuss the relationship between onset of peptic ulcer （PU） and meteorological factors （MFs）. METHODS： A total of 24 252 patients were found with active PU in 104 121 samples of gastroscoic examination from 17 hospitals in Nanning from 1992 to 1997. The detectable rate of PU （DRPU） was calculated every month, every ten days and every five days. An analysis of DRPU and MFs was made in the same period of the year. A forecast model based on MFs of the previous month was established. The real and forecast values were tested and verified. RESULTS： During the 6 years, the DRPU from November to April was 24.4 -28.8%. The peak value （28.8%） was in January. The DRPU from May to October was 20.0 -22.6%, with its low peak （20.0%） in June. The DRPU decreased from winter and spring to summer and autumn （P 〈 0.005）. The correlated coefficient between DRPU and average temperature value was -0.8704, -0.6624, -0.5384 for one month, ten days, five days respectively （P 〈 0.01）. The correlated coefficient between DRPU and average highest temperature value was -0.8000, -0.6470,-0.5167 respectively （P 〈0.01）. The correlated coefficient between DRPU and average lowest temperature value was -0.8091, -0.6617, -0.5384 respectively （P 〈0.01）. The correlated coefficient between DRPU and average dew point temperature was -0.7812, -0.6246, -0.4936 respectively （P 〈0.01）. The correlated coefficient between DRPU and average air pressure value was 0.7320, 0.5777, 0.4579 respectively （P 〈0.01）. The average temperature, average highest and lowest temperature, average air pressure and average dew point temperature value of the previous month, ten days and five days could forecast the onset of PU, with its real and forecast values corresponding to 71.8%, 67.9% and 66.6% respectively. Telomerase activity and human telomerase reverse transcriptase expression in colorectal carcinoma

AIM： To study the activity of telomerase and the expression of human telomerase reverse transcriptase （hTERT） in colorectal carcinoma and its adjacent tissues, normal mucosa and adenomatoid polyp, and to evaluate their relation with carcinogenesis and progression of colorectal carcinoma. METHODS： Telomerase activity and hTERT expression were determined in 30 samples of colorectal carcinoma and its adjacent tissues, normal mucosa and 20 samples of adenomatoid polyp by modified telomeric repeat amplification protocol （TRAP）, enzyme-linked immunosorbent assay （ELISA） and immunohistochemical method. RESULTS： Telomerase activity and hTERT expression were 83.33% （25/30） and 76.67% （23/30） respectively in colorectal carcinoma, which were obviously higher than those in paracancerous tissues （13.33%, 16.67%）, normal mucosa （3.33%, 3.33%） and adenomatoid polyp （10%, 10%）. There was a significant difference between colorectal carcinoma and other tissues （P=0.027）. The telomerase activity and hTERT expression were higher in colorectal carcinoma with lymphatic metastasis than in that without lymphatic metastasis （P=0.034）. When the histological classification and clinical stage were greater, the telomerase activity and hTERT expression increased, but there was no significant difference between them. In colorectal carcinoma, the telomerase activity was correlated with hTERT expression （positive vs negative expression of telomerase activity and hTERT, P=0.021）. CONCLUSION： Telomerase activity is closely correlated with the occurrence, development and metastasis of colorectal carcinoma. Overexpression of hTERT may play a critical role in the regulation of telomerase activity.
