The application of microorganisms as probiotics is limited due to lack of safety ***,a novel multi-stress-tolerant yeast Meyerozyma guilliermondii GXDK6 with aroma-producing properties was identified from marine mangrove *** safety and probiotic properties were assessed in accordance with phenotype and whole-genome sequencing *** showed that the genes and phenotypic expression of related virulence,antibiotic resistance and retroelement were rarely *** morphogenesis genes(SIT4,HOG1,SPA2,ERK1,ICL1,CST20,HSP104,TPS1,and RHO1)and phospholipase secretion gene(VPS4)were *** hyphae and phospholipase were *** one retroelement(Tad1-65_BG)was *** biogenic amines(BAs)encoding genes were absent,except for spermidine synthase(JA9_002594),spermine synthase(JA9_004690),and tyrosine decarboxylase(inx).The production of single BAs and total BAs was far below the food-defined ***6 had no resistance to common antifungal *** enzymes,such as gelatinase,DNase,hemolytic,lecithinase,and thrombin were *** toxicity test with mice demonstrated that GXDK6 is ***6 has a good reproduction ability in the simulation gastrointestinal ***6 also has a strong antioxidant ability,β-glucosidase,and inulinase *** sum up,GXDK6 is considered as a safe probiotic for human consumption and food fermentation.

Insertion sequence transposition activates antimycobacteriophage immunity through an Isr2-silenced lipid metabolism gene island

Insertion sequences(ISs)exist widely in bacterial genomes,but their roles in the evolution of bacterial antiphage defense remain to be ***,we report that,under the pressure of phage infection,the IS1o96 transposition of Mycobacterium smegmatis into the Isr2 gene can occur at high frequencies,which endows the mutant mycobacterium with a broad-spectrum antiphage ***2 functions as a negative regulator and directly silences expression of a gene island composed of 11 lipid metabolism-related *** complete or partial loss of the gene island leads to a significant decrease of bacteriophage adsorption to the mycobacterium,thus defending against phage ***,a phage that has evolved mutations in two tail-filament genes can re-escape from the Isr2 inactivation-triggered host *** study uncovered a new signaling pathway for activating antimycobacteriophage immunity by Is transposition and provided insight into the natural evolution of bacterial antiphage defense.

Facet effects on bimetallic ZnSn hydroxide microcrystals for selective electrochemical CO2 reduction

Employing crystal facets to regulate the catalytic properties in electrocatalytic carbon dioxide reduction reaction（eCO2RR） has been well demonstrated on electrocatalysts containing single metals but rarely explored for bimetallic systems. Here, we synthesize Zn Sn（OH）6（ZSO）microcrystals（MCs） with distinct facets and investigate the facet effects in eCO2RR. Electrochemical studies and in situ Fourier Transform Infrared Spectroscopy（in situ-FTIR） reveal that ZSO MCs produce mainly C1 products of HCOOH and CO. The {111} facet of the ZSO MCS exhibits higher selectivity and faradaic efficiency（FE） than that of the {100} facet over a wide range of potentials（-0.9 V ～-1.3 V versus RHE）. Density Functional Theory（DFT） calculations elucidate that the {111} facet is favorable to the adsorption/activation of CO2molecules,the formation of intermediate in the rate-determining step, and the desorption of C1 products of CO and HCOOH molecules.

Identification and Expression Profile of a Neuropeptide LFRFamide-Like Gene During Different Stages of Gonadal Development in the Cephalopod Sepia pharaonis

Neuropeptides are widely distributed in vertebrates and invertebrates,regulating a variety of physiological activities in the organisms,such as metabolism,feeding and *** this study,to explore the function of neuropeptide LFRFamide in Sepia pharaonis,the full-length cDNA of LFRFamide-like gene(named SpLFRFL,MG869822.1)was identified with rapid amplification of cDNA ends(RACE)*** sequence of SpLFRFL was 860 bp in length and encoded 188 amino acids containing 4 different mature peptides:1 copy of PHTPFRFamide,NSLFRFamide,TIFRFamide,and 3 copies of *** alignment and phylogenetic analysis results showed that SpLFRFL shared high identity with LFRFamides of Sepia officinalis and Sepiella japonica and had the closest relationship with *** quantitative Real-time PCR(qRT-PCR),it was found that the SpLFRFL gene was highly expressed in the optic lobe and brain at three different stages during gonad development in both ***,the four mature peptides at a concentration of 0.01μmol L^(−1) could inhibit the protein synthesis in the Chinese hamster ovary cell strain-K1(CHOK1)induced by *** data suggest that SpLFRFL might be involved in the development and reproduction of *** results can contribute to future studies on neuropeptide evolution and function and benefit the cuttlefish farming.

To maintain,develop and rationally utilize marine organisms,understanding their genetic structure and habitat adaptation pattern is *** punctatus,which is a commercial fish species inhabiting the Indo-west Pacific Ocean,has shown an obvious annual global capture and aquaculture production decline due to climate changes and human *** the present study,restriction-site associated DNA sequencing(RAD-seq)was used to describe its genome-wide single nucleotide polymorphisms panel(SNPs).Among 146 individuals collected at nine locations scattered in China,Korea and Japan,a set of 632,090 SNPs were *** genetic analysis showed that *** individuals were divided into two significant genetic ***,potential genetic differentiation between northern and southern population of *** was *** results indicated that gene flow existed among sampling locations of ***,especially from southern Japan to ***,candidate genes associated with habitat adaptations of *** were identified,which are involved in diverse physiological processes of *** including growth and development(e.g.,KIDINS220,PAN3),substance metabolism(e.g.,PGM5)and immune response(e.g.,VAV3,CCT7,HSPA12B).Our findings may aid in understanding the possible mechanisms for the population genetic structure and local adaptation of ***,which is beneficial to establish the management and conservation units of ***,guiding the rational use of resources,with reference significance for a profound understanding of the adaptative mechanisms of other marine organisms to the environment.

Experimental study on the dynamic response of a 3-D wedge under asymmetric impact

Numerical Simulation on Production Trials by Using Depressurization for Typical marine Hydrate Reservoirs:Well Type and Formation Dip

Natural gas hydrate has huge reserves and is widely distributed in marine *** commercial development is of great significance for alleviating the contradiction between energy supply and *** an efficient research method,numerical simulation can provide valuable insights for the design and optimization of hydrate ***,most of the current production models simplify the reservoir as a two-dimensional(2D)horizontal layered model,often ignoring the impact of formation dip *** improve the accuracy of production prediction and provide theoretical support for the optimization of production well design,two three-dimensional(3D)geological models with different dip angles based on the geological data from two typical sites are *** vertical well,horizontal well and multilateral wells are deployed in these reservoirs with different permeabilities to perform production trial,and the sensitivity analysis of dip angles is also carried *** short-term production behaviors in high and low permeability reservoirs with different dip angles are *** simulation results show that 1)the gas and water production behaviors for different well types in the two typical reservoirs show obviously different variation laws when the short-term depressurization is conducted in the inclined formation;2)the inclined formation will reduce the gas production and increase the water extraction,and the phenomena becomes pronounced as the dip angle increases,particularly in the low-permeability reservoirs;3)and the impact of formation dip on hydrate recovery does not change significantly with the variation of well type.

Buckling Properties of Water-Drop-Shaped Pressure Hulls with Various Shape Indices Under Hydrostatic External Pressure

The water-drop-shaped pressure hull has a good streamline,which has good application prospect in the underwater ***,this study conducted analytical,experimental and numerical investigation of the buckling properties of water-drop-shaped pressure hulls under hydrostatic pressure.A water-drop experiment was conducted to design water-drop-shaped pressure hulls with various shape *** critical loads for the water-drop-shaped pressure hulls were resolved by using Mushtari’s *** numerical simulations including linear buckling analysis and nonlinear buckling analysis including eigenmode imperfections were *** results indicated that the critical loads resolved by Mushtari's formula were in good agreement with the linear buckling loads from the numerical *** formula can be extended to estimate the buckling capacity of water-drop-shaped pressure *** addition,three groups of pressure hulls were fabricated by using stereolithography,a rapid prototyping ***,three groups of the pressure hulls were subjected to ultrasonic measurements,optical scanning,hydrostatic testing and numerical *** experimental results were consistent with the numerical *** results indicate that the sharp end of the water-drop-shaped pressure hulls exhibited instability compared with the blunt *** paper provides a new solution to the limitations of experimental studies on the water-drop-shaped pressure hulls as well as a new configuration and evaluation method for underwater observatories.

In the context of carbon capture,utilization,and storage,the high-value utilization of carbon storage presents a significant *** address this challenge,this study employed the bipolar membrane electrodialysis integrated with carbon utilization technology to prepare Na_(2)CO_(3)products using simulated seawater concentrate,achieving simultaneous saline wastewater utilization,carbon storage and high-value production of Na_(2)CO_(3).The effects of various factors,including concentration of simulated seawater concentrate,current density,CO_(2)aeration rate,and circulating flow rate of alkali chamber,on the quality of Na_(2)CO_(3)product,carbon sequestration rate,and energy consumption were *** the optimal condition,the CO_(3)^(2-)concentration in the alkaline chamber reached a maximum of 0.817 mol/L with 98 mol%*** resulting carbon fixation rate was 70.50%,with energy consumption for carbon sequestration and product production of 5.7 k Whr/m^(3)CO_(2)and1237.8 k Whr/ton Na_(2)CO_(3),*** coupling design provides a triple-win outcome promoting waste reduction and efficient utilization of resources.
