OBJECTIVES:Proteins are the major component and play a key role in nutritious and therapeutic functions of edible bird’s nest(EBN);however,limited studies have been conducted on the protein due to difficulties in extraction,isolation as well as *** study aimed to provide comprehensive information for the quality evaluation of EBN peptides,which would be a valuable reference for further study on EBN ***:Here,we developed a quality control method using high performance liquid chromatography(HPLC)peptide fingerprints deriving from EBN being digested with simulated gastric *** characteristic peptide peaks were collected and identified by LC-MS/***:The characteristic peptide peaks,corresponding to the protein fragments of acidic mammalian chitinase-like,lysyl oxidase,and Mucin-5AC-like,were identified and ***,the principal component analysis indicated that the fingerprints were able to discriminate colour of EBN(white/red)and production sites(cave/house)of White EBN on the same weight *** proposed by the model developed in this study,Muc-5AC-like and AMCaselike proteins were the markers with the highest discriminative ***:The overall findings suggest that HPLC peptide fingerprints were able to clearly demonstrate peptide profile differences between genuine and adulterated EBN samples;and classify EBN samples by its color and production *** addition,the protein identification results suggested that Muc-5AC-like protein was the major protein in EBN.

[Objectives]To establish the quality standard of Shema mixture.[Methods]Thin-Layer Chromatography(TLC)was used to qualitatively identify Ephedra sinica Stapf,Belamcanda chinensis(L.)Redoutéand Citrus maxima(Burm.)*** in the mixture,and High Performance Liquid Chromatography(HPLC)was established for the determination of the content of ephedrine hydrochloride,which was an effective component of principle drug *** Stapf in the mixture.[Results]TLC was used to identify *** Stapf,***(L.)Redoutéand ***(Burm.)*** in Shema mixture,and the characteristic spots were stable and *** hydrochloride had a good linear relationship in the range of 2.01-14.06μg,the average recovery of samples was 98.77%,RSD=1.58%.[Conclusions]TLC and HPLC,as the quality control methods of Shema mixture,have high accuracy and good repeatability,which laid a foundation for the quality control of Shema mixture.

[Objectives] This study was conducted to observe the protective effect of Pratia extract on acute liver injury in mice.[Methods] Mice were randomly divided into 6 groups according to body weight,10 in each group: normal control group,ethanol-induced/CCl4 liver injury model,low-dose,middle-dose and high-dose Pratia extract groups,and bifendatatum *** the blank group,other groups were given 50% ethanol intragastrically at a dose of 12 ml/kg to cause acute alcoholic liver injury,or intraperitoneally injected with 10% CCl4 soybean oil solution to cause acute CCl4 liver *** serum ALT and AST activity were measured as well as liver SOD and MDA concentrations.[Results] Pratia extract effectively reduced serum ALT and AST,decreased MDA content and increased liver tissue SOD activity.[Conclusions] Pratia extract has certain protective effect on acute liver injury.

This article explores the mechanisms of acupuncture meridians by determining characteristics of the tissue fluid flow in the connective tissue along meridians. Based on deep dissection of acupoints on the upper and lower limbs of the human body and micro and macro observation and measurement, a mathematical model of the flow of tissue fluid in interosseous membranes is constructed. It is shown that the signal transmission along acupuncture meridians may be determined by unique anatomical and physiological factors that govern the flow of tissue fluid in the connective tissue and involve mast cell degranulation. Our results provide a preliminary explanation of the phenomenon of the ＂de qi＂ sensation and the mechanism of acupuncture signal transmission along acupuncture meridians.

Pheretima,also called“earthworms”,is a well-known animal-derived traditional Chinese medicine that is extensively used in over 50 Chinese patent medicines(CPMs)in Chinese Pharmacopoeia(2020 edition).However,its zoological origin is unclear,both in the herbal market and *** this study,a strategy for integrating in-house annotated protein databases constructed from close evolutionary relationship-sourced RNA sequencing data from public archival resources and various sequencing algorithms(restricted search,open search,and de novo)was developed to characterize the phenotype of natural peptides of three major commercial species of Pheretima,including Pheretima aspergillum(PA),Pheretima vulgaris(PV),and Metaphire magna(MM).We identified 10,477 natural peptides in the PA,7,451 in PV,and 5,896 in MM *** specific signature peptides were screened and then validated using synthetic peptides;these demonstrated robust specificity for the authentication of PA,PV,and ***,all marker peptides were successfully applied to identify the zoological origins of Brain Heart capsules and Xiaohuoluo pills,revealing the inconsistent Pheretima species used in these *** conclusion,our integrated strategy could be used for the in-depth characterization of natural peptides of other animal-derived traditional Chinese medicines,especially non-model species with poorly annotated protein databases.

The excessive reactive oxygen species(ROS)is a hallmark associated with the initiation and progression of inflammatory bowel disease(IBD),which execrably form a vicious cycle of ROS and inflammation to continually promote disease ***,the gold nanoparticles-embedded ceria nanoparticles(Au/CeO_(2))with enhanced antioxidant activities are designed to block this cycle reaction for treating IBD by scavenging overproduced *** Au/CeO_(2) with core-shell and porous structure exhibits significantly higher enzymatic catalytic activities compared with commercial ceria nanoparticles,likely due to the effective exposure of catalytic sites,higher content of Ce(III)and oxygen vacancy,and accelerated reduction from Ce(IV)to Ce(III).Being coated with negatively-charged hyaluronic acid,the Au/CeO_(2)@HA facilitates accumulation in inflamed colon tissues via oral administration,reduces pro-inflammatory cytokines,and effectively alleviates colon injury in colitis ***,the Au/CeO_(2)@HA with good biocompatibility is a promising nano-therapeutic for treating IBD.

Deep learning(DL)is currently revolutionizing peptide drug development due to both computational advances and the substantial recent expansion of digitized biological ***,progress in oligopeptide drug development has been limited,likely due to the lack of suitable datasets and difficulty in identifying informative features to use as inputs for DL ***,we utilized an unsupervised deep learning model to learn a semantic pattern based on the intrinsically disordered regions of~171 known osteogenic ***,oligopeptides were generated from this semantic pattern based on Monte Carlo simulation,followed by in vivo functional characterization.A five amino acid oligopeptide(AIB5P)had strong bone-formation-promoting effects,as determined in multiple mouse models(e.g.,osteoporosis,fracture,and osseointegration of implants).Mechanistically,we showed that AIB5P promotes osteogenesis by binding to the integrinα5 subunit and thereby activating FAK *** summary,we successfully established an oligopeptide discovery strategy based on a DL model and demonstrated its utility from cytological screening to animal experimental verification.
