Ionic liquids as the effective technology for enhancing transdermal drug delivery: Design principles, roles, mechanisms, and future challenges

Ionic liquids (ILs) have been proven to be an effective technology for enhancing drug transdermal absorption. However, due to the unique structural components of ILs, the design of efficient ILs and elucidation of action mechanisms remain to be explored. In this review, basic design principles of ideal ILs for transdermal drug delivery system (TDDS) are discussed considering melting point, skin permeability, and toxicity, which depend on the molar ratios, types, functional groups of ions and inter-ionic interactions. Secondly, the contributions of ILs to the development of TDDS through different roles are described: as novel skin penetration enhancers for enhancing transdermal absorption of drugs;as novel solvents for improving the solubility of drugs in carriers;as novel active pharmaceutical ingredients (API-ILs) for regulating skin permeability, solubility, release, and pharmacokinetic behaviors of drugs;and as novel polymers for the development of smart medical materials. Moreover, diverse action mechanisms, mainly including the interactions among ILs, drugs, polymers, and skin components, are summarized. Finally, future challenges related to ILs are discussed, including underlying quantitative structure-activity relationships, complex interaction forces between anions, drugs, polymers and skin microenvironment, long-term stability, and in vivo safety issues. In summary, this article will promote the development of TDDS based on ILs.

To the Editor:Medicinal plants have the ability to synthesize a large number of pharmaceutical metabolites,the production and regulation of which are influenced by both intrinsic signals(e.g.,the spatial and temporal stage)and extrinsic environmental conditions(e.g.,temperature,light availability,and water availability)*** to the Encyclopedia of DNA Elements(ENCODE)project2,accessible chromatin regions(ACRs)served essential roles in maintaining the genome architecture and gene regulation in both mammals and ***,identifying ACRs within medicinal plant genomes remains a challenge,as genomic sequences have been determined for numerous species yet the characterization of such functional elements *** this study,we selected Andrographis paniculata(APA)3 as a model system to map ACRs genome-wide by the assay for transposase-accessible chromatin with sequencing(ATAC-seq)*** aimed to uncover the genomic features of ACRs as well as their association with the expression of genes involved in secondary metabolite biosynthesis(e.g.,andrographolide)within medicinal plants(Supporting Information Fig.S1).

Land use/land cover represents the interactive and comprehensive influences between human activities and natural conditions,leading to potential conflicts among natural and human-related issues as well as among *** study introduced economic standards for farmers.A hybrid approach(CA-ABM)of cellular automaton(CA)and an agent-based model(ABM)was developed to effectively deal with social and land-use synergic issues to examine human–environment interactions and projections of land-use conversions for a humid basin in south *** attributes and socioeconomic data were used to analyze land use/land cover and its drivers of *** major modules of the CA-ABM are initialization,migration,assets,land suitability,and land-use change *** estimates of the factors influencing the urban land-use conversion probability were captured using parameters based on a spatial logistic regression(SLR)***,multicriteria evaluation(MCE)and Markov models were introduced to obtain empirical estimates of the factors affecting the probability of ecological land *** agent-based CA-SLR-MCE-Markov(ABCSMM)land-use conversion model was proposed to explore the impacts of policies on land-use *** model can reproduce observed land-use patterns and provide links for forest transition and urban expansion to land-use decisions and ecosystem *** results demonstrated land-use simulations under multi-policy scenarios,revealing the usefulness of the model for normative research on land-use management.

Oxidative therapies receive a limited antitumor efficiency due to the insufficient reactive oxygen species(ROS)levels at focal sites and the evolvement of antioxidant defense ***,we develop an albumin-based nanomedicine to co-deliver chlorin e6(Ce6)and COH-SR4(CS),which can simultaneously enhance the yield and lethality of intracellular ROS for amplified photodynamic therapy(PDT).In which,CS acts as both an activator of AMP-activated protein kinase(AMPK)and an inhibitor of glutathione S-transferases(GSTs).Benefiting from it,the prepared HSA-Ce6@COH-SR4(HCCS)enables positive feed-back uptake by promoting AMPK phosphorylation,leading to rapid and extensive tumor accumulation of *** a result,HCCS obviously increases the ROS production to elevate intracellular oxidative ***,HCCS can inhibit GSTs to disturb the antioxidant defense system of tumor cells,intensifying the oxidative damage of ***,the PDT of HCCS is significantly strengthened by improving the ROS yield and lethality,which greatly declines the proliferation of breast cancer in *** study may open a window in the development of drug co-delivery system for enhanced oxidative therapy of tumors.

An Interfacial Gel Electrode Patch with Tunable Hydrogen Bond Network for Electromyographic Sensing and Discrimination

The sensitivity and fidelity of surface electromyography(sEMG)signal monitoring is critical for muscle status and fatigue assessment,prosthetic control,and gesture ***,the incompatible skin-electrode interface and complex electrophysiological environment restrict the sensitive acquisition and accurate analysis of sEMG *** on the impedance of the skin-electrode interface issue,we developed an interfacial gel electrode patch with a tunable hydrogen bond network to simultaneously achieve a conformal interface,suitable adhesion,and high conductivity for sEMG signal *** exploiting hydroxyethylidene diphosphonic acid(HEDP)and 2-hydroxyphosphono-acetic acid(HPAA)as hydrogen bonding regulators were introduced into the polyvinyl alcohol(PVA)-based hydrogel network to regulate the hydrogen bond cross-linking *** a result,the balance of elastic modulus,adhesion,and electrical conductivity of PVA-HEDP-HPAA(PHH)hydrogel are *** reliable electrodeskin interface is manipulated to achieve conformal contact by matching the elastic modulus,reducing the gap of electrode-skin interface by adhesion,and promoting ion and electron conduction by electrical *** PHH electrode patches exhibit a lower interfacial impedance(12.56 kΩ)and a signal-to-noise ratio of 38.09±1.28 dB for accurate analysis of muscle strength and evaluation of the fatigue *** the assistance of the artificial neural network algorithm,seven gestures can be recognized with 100%prediction *** interfacial gel electrode patch contributes a bio-matching electrophysiological platform for prosthetic control,human–machine interaction,and clinical or athletic auxiliary monitoring.

Keemun black tea stems and leaves: Comparison of chemical characterization and anti-inflammatory activities

This work aims to compare the chemical composition and anti-inflammatory effects on RAW264.7 macrophages of Keemun black tea stems and leaves. Fifty volatile compounds were identified in tea stems and leaves， and aldehydes， alcohols， and esters were the main volatile compound categories. Eleven key volatile compounds， including geraniol， phenylacetaldehyde， methyl salicylate， linalool， etc. contributed to distinguishing the tea stems from the tea leaves. In the quantitative and LC-MS-based metabolomics analysis， higher contents of amino acids， monosaccharides， and quinic acid were found in stems than in leaves. Inversely， higher contents of tea pigments， flavan-3-ols， gallic acid， purine alkaloids， and flavonol glycosides were present in tea leaves than in stems. LC-MS-based metabolomics also revealed that organic acids were the most critical non-volatile compounds responsible for the differences between tea stems and leaves. Furthermore， tea stems had better inhibiting effects of pro-inflammatory cytokines (IL-1β and IL-6) in lipopolysaccharide-challenged RAW264.7 macrophages than tea leaves， while no significant differences exist between leaves and stems for inhibiting the secretion of TNF-α and NO. In conclusion， our results support using Keemun black tea stems as a novel source of anti-inflammatory compounds.

Parvalbumin-positive retinal ganglion cells(PV+RGCs)are an essential subset of RGCs found in various ***,their role in transmitting visual information remains ***,we characterized PV+RGCs in the retina and explored the functions of the PV+RGC-mediated visual *** applying multiple viral tracing strategies,we investigated the downstream of PV+RGCs across the whole ***,we found that the PV+RGCs provided direct monosynaptic input to PV+excitatory neurons in the superficial layers of the superior colliculus(SC).Ablation or suppression of SC-projecting PV+RGCs abolished or severely impaired the flight response to looming visual stimuli in mice without affecting visual ***,using transcriptome expression profiling of individual cells and immunofluorescence colocalization for RGCs,we found that PV+RGCs are predominant glutamatergic ***,our findings indicate the critical role of PV+RGCs in an innate defensive response and suggest a non-canonical subcortical visual pathway from excitatory PV+RGCs to PV+SC neurons that regulates looming visual *** results provide a potential target for intervening and treating diseases related to this circuit,such as schizophrenia and autism.

Background:Shenzao dripping pill(SZDP)is empirically prescribed for treating cardiac ***,there is a lack of comprehensive knowledge regarding the underlying mechanisms contributing to its therapeutic *** objective of this study is to investigate the underlying mechanism of SZDP against chronic myocardial ischemia(CMI)in a rat ***:In this study,we utilized electrocardiographic and echocardiographic detection along with pathological tissue analysis to evaluate the efficacy of *** integration of network pharmacology and metabolomics was conducted to investigate the *** docking and molecular dynamics simulations were used to validate the binding energy between the compounds of SZDP and the associated ***:The results showed that SZDP was able to improve T wave voltage,reverse CMI abnormalities in ejection fraction and fractional shortening,and restore histopathological heart *** results indicated that disturbances of metabolic profile in CMI rats were partly corrected after SZDP administration,mainly affecting purine metabolism.13-Docosenamide may be the potential metabolic biomarker of the therapeutic application of SZDP for *** network pharmacology and metabolomics,thiopurine S-methyltransferase(TPMT),xanthine dehydrogenase/oxidase(XDH),bifunctional purine biosynthesis protein ATIC(ATIC),and cytochrome p4501A1(CYP1A1)were identified as possible targets of SZDP to exert therapeutic effects by enhancing the metabolic levels of L-Tryptophan,Deoxyribose 1-phosphate and Phosphoribosyl ***:SZDP has a therapeutic effect on CMI by regulating metabolite levels,acting on the targets of TMPT,XDH,ATIC,and CYP1A1,and reducing cardiomyocyte injury and myocardial fibrosis.

High-precision nondestructive evaluation of a thermal barrier coating based on perovskite quantum dot anion exchange

Thermal barrier coatings(TBCs)in gas turbine engines are used in expressly harsh environments;thus,assessing TBC integrity status is critical for safety and ***,traditional periodic maintenance involves visual inspections of the TBCs,requiring the gas turbine to be decommissioned and partially *** importantly,tiny defects or internal damages that easily cause coating failure cannot be *** this work,a new nondestructive evaluation(NDE)technique of TBCs based on quantum dot(QD)anion exchange is first explored *** exchanging anions between the Cl ions and the CsPbBr_(3) QDs,the degrees of salt corrosion of the TBCs are *** resultant NDE technique shows that the colour of the TBCs obviously changes from green to blue,accompanied by a large blueshift(~100 nm)of the photoluminescence(PL)peak *** addition,the relationship between the PL peak position and coating thermophysical properties indicates that the precision of this NDE technique may easily identify theμm-level of the thermal growth oxide(TGO)changes inside the TBCs.
