Toxicity assessment and histopathological analysis of nano-ZnO against marine fish(Mugilogobius chulae) embryos

The toxicity of nano-materials has received increasing attention in recent years. Nevertheless, relatively few studies have focused on their oceanic distributions and toxicities. In this study, we assessed nano-ZnO toxicity in marine organisms using the yellowstriped goby （Mugilogobius chulae）. The relative differences in nano-ZnO dissolution and dispersal in seawater and fresh water were also investigated. The effects of nano-ZnO on embryonic development, deformity, hatching, mortality, and histopathology were analyzed. In addition, the effects of the Zn2＋ concentration on M. chulae hatching and mortality were compared. The results showed that nano-ZnO had higher solubility in seawater than in fresh water. Nano-ZnO significantly inhibited hatching. By the fifth day of exposure, the LC50 of nano-ZnO was 45.40 mg/L, and the mortality rate spiked. Hatching inhibition and lethality were dose-dependent over a range of 1-25 mg/L nano-ZnO. Zn2＋ inhibited hatching and increased lethality, but its effects were weaker than those of nano-ZnO at the same concentrations. Nano-ZnO also induced spinal bending, oedema, hypoplasia, and other deformities in M. chulae embryos and larvae. Histopathology revealed vacuolar degeneration, hepatocyte and enterocyte enlargement, and morphological abnormalities of the vertebrae. Therefore, nano-ZnO caused malformations in M. chulae by affecting embryonic growth and development. We conclude that nano-ZnO toxicity in seawater was significantly positively correlated with the associated Zn2＋ concentration and sedimentary behaviour. The toxicity of nano-ZnO was cumulative and showed a critical point, beyond which embryonic and developmental toxicity in marine fish was observed.

Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

Development of Neural Network for BLSOM Clustering of HA Genes of Avian Influenza Viruses Isolated in guangdong Province

A neural network classification method,and a batch-learning self-organizing map(BLSOM),was established using trinucleotide and tetranucleotide in the hemagglutinin gene sequences of 25 avian influenza viruses isolated in guangdong Province. Statistical analysis and normalization of the fragment number were done and MATLAB function was used to simulate the human brain thinking for self-organizing learning. When the number of training steps was 100 and above,the strains could be successfully clustered. H_1,H_3,H_5,H_7 and H_9 subtype strains fell within different classes,respectively,and the HA gene cluster map of H_3N_2 and H_7N_9strains was quite similar,suggesting that these strains shared the same origin; H_5N_1 strain was quite different in different years; H_1N_1 and H_9N_2 strains could be clustered into one group,indicating the natural recombinant variation in the two kinds of viruses,thereby providing a reference for high-risk strain screening and traceability.

Numerical verification of similar Cam-clay model based on generalized potential theory

From the mathematical principles, the generalized potential theory can be employed to create constitutive model of geomaterial directly. The similar Cam-clay model, which is created based on the generalized potential theory, has less assumptions,clearer mathematical basis, and better computational accuracy. Theoretically, it is more scientific than the traditional Cam-clay models. The particle flow code PFC3 D was used to make numerical tests to verify the rationality and practicality of the similar Cam-clay model. The verification process was as follows: 1) creating the soil sample for numerical test in PFC3 D, and then simulating the conventional triaxial compression test, isotropic compression test, and isotropic unloading test by PFC3D; 2)determining the parameters of the similar Cam-clay model from the results of above tests; 3) predicting the sample's behavior in triaxial tests under different stress paths by the similar Cam-clay model, and comparing the predicting results with predictions by the Cam-clay model and the modified Cam-clay model. The analysis results show that the similar Cam-clay model has relatively high prediction accuracy, as well as good practical value.

Conformations of surface atoms in various stages of nanogold-based genechip testing are scanned by the atomic force and scanning tunneling microscope. We intuitively observe the process and differences in probe combination, nucleic acid hybridization, and silver staining, which might be useful to validate the assay method of genechip. We hope to use this technology to make the other invisible chemical or biochemical reaction become visible and convincible in the future.

Effect of glycine site/NMDA receptor antagonist MRZ2/576 on the conditioned place preference and locomotor activity induced by morphine in mice

Objective: To study the effect of glycine site/NMDA (N-methyl-D-aspartate) receptor antagonist MRZ2/576 on the conditioned place preference (CPP) and locomotor activity induced by morphine in mice. Methods: Different doses (1.25, 2.5 and 5 mg/kg, i.p.) of MRZ2/576 were used to evaluate the effect of MRZ2/576 on the acquisition and expression of CPP induced by morphine (5 mg/kg) in mice. In addition, we examined the locomotor activity of mice in conditioning and testing phase of CPP paradigm. Results: MRZ2/576 alone could not establish place preference, but a 5 mg/kg dose of MRZ2/576 could block both acquisition and expression of morphine-induced CPP. In testing phase of CPP, there was no statistical difference for locomotor activity between the groups; injection of MRZ2/576 showed a dose-dependent decrease of locomotor activity on both control and morphine-treated mice, especially 5 mg/kg of MRZ2/576 significantly suppressed the locomotor activity of mice. Conclusion: Based on the present results, we assume that MRZ2/576 can antagonize the rewarding effect of morphine, suggesting that this glycine site/NMDA receptor antagonist could be used to treat addictions due to its light side effect profile.

Pseudomonas mendocina LYX:A novel aerobic bacterium with advantage of removing nitrate high effectively by assimilation and dissimilation simultaneously

The problem of nitrate accumulation in aerobic tank and total nitrogen excessive discharge in effluent was very common in traditional livestock and poultry farming wastewater treatment systems owing to the lengthy process flow and low process control level.A strain LYX of aerobic bacterium was isolated from the activated sludge of a wastewater treatment system in a pig farm,which could remove nitrate effectively in aerobic tank and was identified Pseudomonas mendocina by 16S rRNA *** the condition of nitrate as the sole nitrogen source,this strain removed over 90%of NO_(3)^(−)-N with an initial concentration of 110 mg/L under aerobic conditions within 48 *** them,37.9%of NO_(3)^(−)-N was assimilated into Bio-N,about 51.9%was reduced to gaseous nitrogen and less than 0.5%of nitrogen was replaced by NO_(3)^(−)-N and NH_(4)^(+)-N,9.7%NO_(3)^(−)-N remained in the effluent at the *** the same time,four key genes(napA,nirK,norB and nosZ)related to nitrate nitrogen removal were expressed during the denitrification process of *** LYX,in which the transcription level of the indicator genes of this aerobic denitrifying bacterium(napA)was the *** addition,it was found with the 15N tracer technique that inoculation of this strain on sludge increased the amount of nitrogen loss from 9.26 nmol N/(g·h)to 23.835 nmol N/(g·h).Therefore,*** LYX is a potential bioagent for advanced nitrogen removal by assimilating and reducing nitrate simultaneously in aerobic tanks.

Polydopamine-Assisted Fabrication of Fiber-Optic Localized Surface Plasmon Resonance Sensor Based on Gold Nanoparticles

A fast and facile method of fabricating fiber-optic localized surface plasmon resonance sensors baseff on spherical gold nanoparticles was introduced in this study. The gold nanoparticles with an average diameter of 55 nm were synthesized via the Turkevich method and were then immobilized onto the surface of an uncladded sensor probe using a polydopamine layer. To obtain a sensor probe with high sensitivity to changes in the refractive index, a set of key optimization parameters, including the sensing length, coating time of the potydopamine layer, and coating time of the gold nanoparticles, were investigated. The sensitivity of the optimized sensor probe was 522.80 nm per refractive index unit, and the probe showed distinctive wavelength shifts when the refractive index was changed from 1.328 6 to 1.398 7. When stored in deionized water at 4 ℃, the sensor probe proved to be stable over a period of two weeks. The sensor also exhibited advantages, such as low cost, fast fabrication, and simple optical setup, which indicated its potential application in remote sensing and real-time detection.

C. biprojicienus Wei ＆ Zhang and C. serratus Wang ＆ Yang are recorded from Shanxi for the first time. A key to the species of Chrysotus from Shanxi is presented.
